    Sinomenine, a natural dextrorotatory morphinan analog, is anti-inflammatory and neuroprotective through inhibition of microglial NADPH oxidase

    <p>Abstract</p> <p>Background</p> <p>The mechanisms involved in the induction and regulation of inflammation resulting in dopaminergic (DA) neurotoxicity in Parkinson's disease (PD) are complex and incompletely understood. Microglia-mediated inflammation has recently been implicated as a critical mechanism responsible for progressive neurodegeneration.</p> <p>Methods</p> <p>Mesencephalic neuron-glia cultures and reconstituted cultures were used to investigate the molecular mechanisms of sinomenine (SN)-mediated anti-inflammatory and neuroprotective effects in both the lipopolysaccharide (LPS)- and the 1-methyl-4-phenylpyridinium (MPP<sup>+</sup>)-mediated models of PD.</p> <p>Results</p> <p>SN showed equivalent efficacy in protecting against DA neuron death in rat midbrain neuron-glial cultures at both micro- and sub-picomolar concentrations, but no protection was seen at nanomolar concentrations. The neuroprotective effect of SN was attributed to inhibition of microglial activation, since SN significantly decreased tumor necrosis factor-α (TNF-α, prostaglandin E<sub>2 </sub>(PGE<sub>2</sub>) and reactive oxygen species (ROS) production by microglia. In addition, from the therapeutic point of view, we focused on sub-picomolar concentration of SN for further mechanistic studies. We found that 10<sup>-14 </sup>M of SN failed to protect DA neurons against MPP<sup>+</sup>-induced toxicity in the absence of microglia. More importantly, SN failed to show a protective effect in neuron-glia cultures from mice lacking functional NADPH oxidase (PHOX), a key enzyme for extracellular superoxide production in immune cells. Furthermore, we demonstrated that SN reduced LPS-induced extracellular ROS production through the inhibition of the PHOX cytosolic subunit p47<sup><it>phox</it></sup>translocation to the cell membrane.</p> <p>Conclusion</p> <p>Our findings strongly suggest that the protective effects of SN are most likely mediated through the inhibition of microglial PHOX activity. These findings suggest a novel therapy to treat inflammation-mediated neurodegenerative diseases.</p

    Structure of the Mouse TRPC4 Ion Channel

    Members of the transient receptor potential (TRP) ion channels conduct cations into cells. They mediate functions ranging from neuronally mediated hot and cold sensation to intracellular organellar and primary ciliary signaling. Here we report a cryo-electron microscopy (cryo-EM) structure of TRPC4 in its unliganded (apo) state to an overall resolution of 3.3 Å. The structure reveals a unique architecture with a long pore loop stabilized by a disulfide bond. Beyond the shared tetrameric six-transmembrane fold, the TRPC4 structure deviates from other TRP channels with a unique cytosolic domain. This unique cytosolic N-terminal domain forms extensive aromatic contacts with the TRP and the C-terminal domains. The comparison of our structure with other known TRP structures provides molecular insights into TRPC4 ion selectivity and extends our knowledge of the diversity and evolution of the TRP channels

    Janus hollow fibre membranes with intrusion anchored structure for robust desalination and leachate treatment in direct contact membrane distillation

    High-performance Janus membranes have been intriguing for membrane distillation due to its distinctive features for improving fouling and wetting resistance. However, long-term structural stability remains challenging owing to delamination caused by the opposite affinity between the two different kinds of materials. Herein, a physically-anchored hydrophilic layer containing graphene oxide (GO)/poly(vinyl alcohol) (PVA)/sulfosuccinic acid (SSA) i.e., GOSP layer was formed on the surface of a microporous polypropylene (PP) hollow fibre membrane. Meanwhile, the nanostructure of the GOSP layer could be tuned from 2-dimensional lamellar to 3-dimensional mixed matrix, exhibiting various water transport properties. As a result, the fabricated Janus membrane with the partially intruded pore channels exhibited superior antiwetting and antifouling performance for desalination and wastewater treatment in the direct contact membrane distillation process. For surfactant containing saline water, the GOSP/PP Janus membrane with optimised PVA/GO ratio possessed noticeably enhanced anti-wetting property compared to pristine PP membrane. In particular for leachate treatment, the GOSP/PP membrane produced high quality permeate with <1 mg/L ions and 10 mg/L total organics and maintained stable water flux while the leachate was concentrated by 5 times (80 % water recovery), providing great potential for using membrane distillation towards zero liquid discharge

    Differential Expression Analysis of Olfactory Genes Based on a Combination of Sequencing Platforms and Behavioral Investigations in Aphidius gifuensis

    Aphidius gifuensis Ashmead is a dominant endoparasitoid of aphids, such as Myzus persicae and Sitobion avenae, and plays an important role in controlling aphids in various habitats, including tobacco plants and wheat in China. A. gifuensis has been successfully applied for the biological control of aphids, especially M. persicae, in green houses and fields in China. The corresponding parasites, as well as its mate-searching behaviors, are subjects of considerable interest. Previous A. gifuensis transcriptome studies have relied on short-read next-generation sequencing (NGS), and the vast majority of the resulting isotigs do not represent full-length cDNA. Here, we employed a combination of NGS and single-molecule real-time (SMRT) sequencing of virgin females (VFs), mated females (MFs), virgin males (VMs), and mated males (MMs) to comprehensively study the A. gifuensis transcriptome. Behavioral responses to the aphid alarm pheromone (E-β-farnesene, EBF) as well as to A. gifuensis of the opposite sex were also studied. VMs were found to be attracted by female wasps and MFs were repelled by male wasps, whereas MMs and VFs did not respond to the opposite sex. In addition, VFs, MFs, and MMs were attracted by EBF, while VMs did not respond. According to these results, we performed a personalized differential gene expression analysis of olfactory gene sets (66 odorant receptors, 25 inotropic receptors, 16 odorant-binding proteins, and 12 chemosensory proteins) in virgin and mated A. gifuensis of both sexes, and identified 13 candidate genes whose expression levels were highly consistent with behavioral test results, suggesting potential functions for these genes in pheromone perception

    A mini review on antiwetting studies in membrane distillation for textile wastewater treatment

    The textile industry is an important contributor to the growth of the global economy. However, a huge quantity of wastewater is generated as a by-product during textile manufacturing, which hinders the ongoing development of textile industry in terms of environmental sustainability. Membrane distillation (MD), which is driven by thermal-induced vapor pressure difference, is being considered as an emerging economically viable technology to treat the textile wastewater for water reuse. So far, massive efforts have been put into new membrane material developments and modifications of the membrane surface. However, membrane wetting, direct feed solution transport through membrane pores leading to the failure of separation, remains as one of the main challenges for the success and potential commercialization of this separation process as textile wastewater contains membrane wetting inducing surfactants. Herein, this review presents current progress on the MD process for textile wastewater treatment with particular focuses on the fundamentals of membrane wetting, types of membranes applied as well as the fabrication or modification of membranes for anti-wetting properties. This article aims at providing insights in membrane design to enhance the MD separation performance towards commercial application of textile wastewater treatment

    World review of capture fisheries and aquaculture insurance 2022

    This world review of capture fisheries and aquaculture insurance presents the findings of five regional and four national reports conducted in 2020. An estimated 450 000 fishing vessels worldwide are covered by marine hull insurance. Nearly all of the estimated 67 800 large-scale industrial fishing vessels are covered by marine hull insurance, as well as 50–60 percent of the estimated 430 000 semi-industrial fishing vessels. However, over 95 percent of the 2.3 million motorized small-scale fishing vessels operate uninsured. Most small-scale fishers do not have access to adequate insurance services. Between 2009 and 2019, underwriting experiences in fishing vessel insurance were generally reported as “Good”. What is more, access to accident, life and health insurance services for crew on fishing vessels and small-scale fishers in developing countries has improved in recent years. In 2020, the number of aquaculture insurance policies in force was estimated at over 40 000 worldwide, with China and Indonesia the largest markets for this type of insurance. While large-scale aquaculture producers are well served by the insurance industry, the provision of insurance is inadequate for mediumand small-scale farmers, particularly in Asia. Underwriting experiences for aquaculture stock mortality insurance were reported as “Good” to “Very good” (40 percent), or “Neutral” (36 percent). The insurance industry has consolidated the market and increased profitability in aquaculture insurance. This world review contains information on the capture fisheries and aquaculture insurance market, the prevailing underwriting practices, perils covered, policies in force, risk management and claim handling procedures. Finally, it offers a series of recommendations for increasing insurance service provision to the fisheries and aquaculture industries
