    Effect of two different source of forage on the organic matter digestibility in Mediterranean Italian Buffalo cows

    The present study aimed to evaluate the influence of two different source of forage (haysilage and hay) during the lactation on organic matter digestibility (OMD) in buffalo cows. Lactating buffaloes (n = 40) at 29.6 days in milk (DIM) were equally divided as function of previous milk yield into Group 1 (meadow hay, n = 20) and Group 2 [haysilage (Lolium multiflorum), n = 20]. The diets were isoenergetic [0.92 milk unit forage (MUF) on dry matter basis] and isoproteic (16.2 % crude protein on dry matter basis) and administred as total mixed ration (TMR). From all the buffaloes, for each group and in two sampling time (first sampling, DIM = 74.0 and second sampling, DIM = 129.0) the faeces were collected in order to evaluate the in vivo digestibility. Overall the trial, the subjects fed haysilage showed higher OMD than those fed hay (66.1 vs 45.7; P<0.01), moreover the in vivo digestibility was affected by the DIM, in particular in the group fed hay (40.6 vs 53.4; P<0.0001, respectively). This work underlines the importance of the administration of the haysilage (Lolium multiflorum) as source of high quality forage because it gives, overall the lactation, more nutritive principles compared with meadow hay

    Effect of Cereals and Legumes Processing on In Situ Rumen Protein Degradability: A Review

    The determination of the ruminal degradability rate of feeds, mainly starch and crude protein, is one of the most common methods to evaluate the nutritional value of ruminant feed. The protein requirements for ruminants are met from microbial protein and undegraded dietary protein digested in the small intestine. In order to reach maximum productivity, high-quality proteins are needed, and the requirement for undegraded dietary protein increases with the performance of the animal. This protein can be supplied by reducing the ruminal degradation to increase the amount of protein digested post-rumen, but the form in which a feed is administered influences degradability, and grain processing, especially, is a common practice to improve feed efficiency. Despite these aspects, studies on the effects of feed processing methods on protein degradability are limited, even though more and more ruminants are fed with processed feeds. For these reasons, this review investigated the protein degradability of different processed cereals and legumes in ruminants based on the analysis of available literature in order to take stock of the state of the art on this topic. Results showed that: First, the majority of the papers are focused on the energy aspects mainly due to carbohydrate-rich feeds; second, the majority of the studies in the literature are quite old, probably because the changes occurred in the animal testing legislation that made in vivo studies more and more difficult in the last 20 years; third, as a consequence, the few data available in recent years concern in vitro experiments; fourth, we found a high variability of the experimental conditions thus affecting protein degradability and making it quite difficult to compare the different results

    Follicular dynamics in synchronized Italian Mediterranean buffalo cows

    The aim of this study was to evaluate the length and the characteristics of the oestrous cycle in Italian Mediterranean buffalo cows, undergone synchronization of ovulation. The trial was performed on 32 buffaloes synchronized by the Ovsynch Program, which consists of an injection of GnRH on day 0, PGF2&alpha; on day 7 and GnRH on day 9. Starting on day 10 (Day 0 of the new cycle). Buffaloes undergone ultrasound examination of the ovaries on alternate days until the following heat. Follicular growth and corpus luteum formation and dimensions were recorded as well as the number of follicular waves. Statistical analysis was performed by ANOVA. Four animals (12.5%) did not show signs of oestrous and were excluded from the trial. The mean length of the oestrous cycle was 23.7&plusmn;3.4 days. In particular, 1 animal (3.6%) showed an oestrous cycle characterized by 1 follicular wave with a length of 16 days, 17 subjects (60.7%) showed 2 follicular waves with a cycle length of 22.4&plusmn;2.3 days and 10 buffaloes (35.7%) showed 3 follicular waves with a cycle of 26.8&plusmn;2.0 days. These results confirm previous reports performed in buffalo species, although the cycle resulted longer in the 3-waves group

    Buffalo milk samples differing significantly for cheese yield values were analysed by 2D electrophoresis in order to outline a protein profile, with specific regards to k-casein fractions. Four buffaloes, two of which showing high cheese yield and two with low cheese yield selected from a group of 135 subjects were chosen for the proteomic analyses. Six main spots in 2D gels were recognized as αs1-, αs2-, β- and k-casein, α-lactoalbumin, β-lactoglobulin. The main visible differences in the 2D gels between buffaloes with high vs. low cheese yield were found in the appearance of the four k-casein spots (spots numbers:20, 19, 16, 18) which differ in the number of phosphorilation and glycosilation. The area and the intensity of the four spots were calculated by using Melanie II (Bio-Rad) software. Samples with high cheese yield showed higher value of the by-products: area x intensity of spot 16, correspondent to k-casein with one phosphorilation site, and lower values of spots 19 and 20, of k-casein with more than one phosphorilation site and glycosilated

    Effect of pre-partum habituation to milking routine on behaviour and lactation performance of buffalo heifers

    The aim of this study was to investigate the effect pre-partum habituation in the milkingparlour on behaviour and lactation performance of buffalo heifers. Sixteen buffalo heifersin late gestation were used for this study. The animals, with an age ranging from 30 to44 months at the start of the study, were equally allocated into two treatments and bal-anced for estimated calving date. Eight animals received pre-partum habituation for 10 daysbefore the estimated calving date (group H), while the eight others received no treatmentand served as the control (group C). During the pre-partum habituation sessions, group Hanimals were moved to the milking parlour and left for 10 min in the milking stall onceeach day, where the udder was thoroughly washed with warm water, wiped with a dis-posable towel and massaged. The behaviours, registered from entrance into the milkingstall to exit, were step and kick. After calving milk flow profiles, milk yield and milk qual-ity variables were determined along with behavioural recordings. The H animals duringthe habituation procedure showed a reduction in the number of steps (P < 0.001) and kicks(P < 0.01). After calving the H animals performed fewer steps than the control animals at 0(P < 0.001), 3 (P < 0.01), 6 (P < 0.01), 13 (P < 0.01) and 20 (P < 0.01) days after calving. Animalsfrom group H also performed fewer kicks than control animals at 0 (P < 0.001), 3 (P < 0.01),6 (P < 0.01) and 13 (P < 0.01) days after calving. In the C group a reduction in the numberof steps (P < 0.001) and kicks (P < 0.001) was observed as lactation proceeded, whereas forgroup H only a tendency for a reduction over lactation was detected. Pre-partum habitu-ation did not significantly affect milk quality or milk flow variables. Milk yield in the first3 min of milking (P < 0.001), and average milk flow (P < 0.001), increased throughout theexperimental period, whereas the duration of the pre-milking phase decreased as lactationproceeded (P < 0.001). This study shows that buffalo heifers exposed to a pre-partum habit-uation programme performed fewer steps and kicks than control animals during milking.Therefore, it is concluded that using this treatment can reduce the level of restlessness inbuffalo heifers during milking

    A Multi-Analytical Diagnostic on an Outdoor Wall Painting: The Study on the Déesis of St. Maria Annunziata’s Church, Motta San Giovanni (Reggio Calabria, Italy)

    This article concerns the diagnostic campaign aimed at analyzing the mural painting representing the iconographic theme of the Deesis of the Church of St. Maria Annunziata, Motta San Giovanni, in the province of Reggio Calabria. In 1951, a flood caused the collapse of the building and the consequent breaking of the apse into two parts. The present study focused on the left side of the apse, hosting the figures of Christ and Mary, in order to plan the best conservation intervention strategy. For this purpose, non-invasive investigations and laboratory analytical methods were conducted in order to characterize the constituent materials and to identify the forms of alteration and degradation present on the surface of the painting. In particular, Raman spectroscopy, optical microscopy, scanning electron microscopy coupled to the chemical analysis by an EDS probe, Fourier-transform infrared spectroscopy and ion chromatography were employed. The results highlighted the presence of a single layer of plaster made with a lime-based binder. The chromatic palette of the painting is characterized by ochres and carbon black mixed with lime to obtain the different shades. Finally, the definition of the nature of the deposits and of the overlaid materials was fundamental in order to identify the best products and methods to restore the readability of the work

    Diet and Plastic Ingestion in the Blackmouth Catshark Galeus melastomus, Rafinesque 1810, in Italian Waters

    Simple Summary The blackmouth catshark has a habitat range that spans from the Norwegian seas to Senegal and throughout the Mediterranean Sea, and it is one of the most common sharks in Italian waters. The aim of this work is to investigate, through the analyses of the stomach contents of five populations from the Tyrrhenian and Ionian Seas, the diet of blackmouth catsharks. The analyses showed that the most frequent items were Osteichthyes of the family Myctophidae, except for one population, in which the most common items were Cephalopods and Crustacean of the Decapods order. Plastic debris was also found in all populations analysed and classified by colour and shape. This study ought to increase the knowledge of the feeding ecology of the blackmouth catshark, thus improving the meagre literature about Tyrrhenian and Ionian waters. Galeus melastomus is the most common Pentanchidae in the Mediterranean Sea. A scavenger and opportunistic feeder, and despite the wide distribution, little is known about its feeding habits in Italian waters. The main purpose of this study was to investigate the diet of the blackmouth catshark by analysing the stomach contents. The specimens analysed were obtained from five populations of the Tyrrhenian and of the Ionian Seas, collected from a depth between 40 and 700 m. A total of 259 stomachs were analysed. The stomach contents were grouped into macro-categories and identified to the lowest taxonomic level possible. Crustaceans such as Parapenaeus longirostris, the Cephalopods Heteroteuthis dispar and Onychoteuthis banksii, and Osteichthyes, mostly Myctophidae, were identified. Plastic debris was also found among the stomach contents and classified according to its colour and shape. Osteichthyes represent the most abundant item (44%), above all the Myctophidae family, except for the catshark population from Tuscany, in which the most frequent species were Cephalopods, such as Abralia veranyi and Heteroteuthis dispar. Differences in the plastic debris contents were also observed between the Tuscany population and other populations. These could be explained as a probable consequence of the different depths at which the blackmouth catshark populations were sampled

    The silane depletion fraction as an indicator for the amorphous/crystalline silicon interface passivation quality

    In silicon heterojunction solar cells, thin amorphous silicon layers passivate the crystalline silicon wafer surfaces. By using in situ diagnostics during plasma-enhanced chemical vapor deposition (PECVD), the authors report how the passivation quality of such layers directly relate to the plasma conditions. Good interface passivation is obtained from highly depleted silane plasmas. Based upon this finding, layers deposited in a large-area very high frequency (40.68 MHz) PECVD reactor were optimized for heterojunction solar cells, yielding aperture efficiencies up to 20.3% on 4 cm
