    Identification of a Novel Testis-specific Gene in Mice and Its Potential Roles in Spermatogenesis

    Aim Identification of a novel gene in mouse testis and its relation to spermatogenesis. Methods Genes expressed during different developmental stages of the mouse testis were screened by DNA microarray. The results of chip analysis were authenticated by reverse transcription-polymerase chain reaction (RT-PCR) technique, as well as the tissue distribution of the selected genes. The characteristics of the selected genes were analyzed by bioinformatics tools. Results A novel gene, TSC77, was identified and located at the mouse chromosome 2G1. The full cDNA length of TSC77 was 2280 bp, with a 2046 bp open reading frame encoding a 681 amino acids protein with a predicted molecular weight of 77.17 kDa. The results of subcellular localization of GFP-TSC77 fusion protein indicated TSC77 protein was located in the nucleus of Cos-7 cells. The analysis of multiple amino acid sequence alignment showed that TSC77 protein was highly homologous with the human CAI40813 (C2orf26 gene, 76%), and rat XP_230651 (77%).     Investigation and identification of let-7a related functional proteins in gastric carcinoma by proteomics

    Abstract. MicroRNAs are small noncoding RNA molecules that control expression of target genes. Our previous studies show that let-7a decreased in gastric carcinoma and that up-regulation of let-7a by gene augmentation inhibited gastric carcinoma cell growth both in vitro and in vivo, whereas it remains largely unclear as to how let-7a affects tumor growth. In this study, proteins associated with the function of let-7a were detected by high throughout screening. The cell line of SGC-7901 stablely overexpressing let-7a was successfully established by gene cloning. Two-dimensional gel electrophoresis (2-DEy was used to separate the total proteins of SGC-7901/let-7a, SGC-7901/EV and SGC-7901, and PDQuest software was applied to analyze 2-DE images. Ten different protein spots were identified by MALDI-TOF-MS, and they may be the proteins associated with let-7a function. The overexpressed proteins included Antioxidant protein 2, Insulin-like growth factor binding protein 2, Protein disulfide isomerase A2, C-1-tetrahydrofolate synthase, Cyclin-dependent kinase inhibitor1 (CDKN1) and Rho-GTPase activating protein 4. The underexpressed proteins consisted of S-phase kinase-associated protein 2 (Spk2), Platelet membrane glycoprotein, Fibronectin and Cks1 protein. Furthermore, the different expression levels of the partial proteins (CDKN1, Spk2 and Fibronectin) were confirmed by western blot analysis. The data suggest that these differential proteins are involved in a novel let-7a signal pathway and these findings provide the basis to investigate the functional mechanisms of let-7a in gastric carcinoma

    Equipo simulador de agua de lluvia para experimentos a campo en ecosistemas áridos y semiáridos

    The predicted changes in precipitation patterns because of global change have profound effects on terrestrial ecosystems. In the present study, the principle and design details of a rainfall enrichment system (RAINES) for open field experiments in semi-arid and arid ecosystems are presented. The rainfall intensity, validity and uniformity of this experimental facility were also tested. During the period from 2008 to 2010, our data showed that the RAINES was able to simulate rainfall events with different rainfall sizes, frequencies and timing. The greatest advantage of the RAINES was its high uniformity in rainfall distribution over a relatively large experimental surface area (>65 m2), which was important for experimental studies of semi-arid and arid ecosystems where vegetation distribution is sparse. The rainfall validity of RAINES was steadily at 66% or higher as long as the hydraulic pressure exceeded 1.4 KPa and the wind speed was below 2.5 m s-1. Since the RAINES is light-weight, inexpensive and versatile enough to be used to simulate various rainfall events with needed properties in remote fields, it is able to provide reliable simulated rainfall in the field for studying possible responses of soil and vegetation processes to rainfall change in arid and semi-arid ecosystems. The application of the RAINES will improve our understanding on the relationship between water availability and ecosystem processes in arid and semi-arid ecosystems, which will provide useful knowledge for the protection, restoration and sustainable management of semi-arid and arid desert ecosystems worldwide.Los cambios predichos en los modelos de precipitación como resultado del cambio global tienen efectos profundos en los ecosistemas terrestres. Un equipo que simule la lluvia es una herramienta de investigación efectiva para explorar los efectos de los cambios en los modelos de lluvia sin varias restricciones naturales. En el presente estudio, se presentan los principios y detalles de diseño de un equipo simulador de lluvia (RAINES) para estudios a campo en ecosistemas áridos y semiaridos. También se determinaron la intensidad de lluvia, validez y uniformidad del RAINES. Durante el período 2008 al 2010, nuestros datos mostraron que el RAINES fue capaz de simular eventos de lluvia con diferentes cantidades, frecuencias y momentos de lluvia. La mayor ventaja del RAINES fue su gran uniformidad en la distribución de la lluvia sobre una superficie experimental relativamente grande (>65 m2 ). Esto es importante para estudios experimentales en ecosistemas áridos y semiáridos donde la distribución de la vegetación es dispersa. La validez de la lluvia provista por el RAINES fue al menos de 66% siempre y cuando la presión hidráulica excedió los 1.4 KPa y la velocidad del viento fue menor que 2.5 m s-1. El RAINES es de bajo peso, no costoso y lo suficientemente versátil como para ser usado para simular varios eventos de lluvia bajo condiciones de campo distantes. Es capaz de proveer lluvia simulada en forma confiable en el campo para estudiar la respuesta de procesos en el suelo y la vegetación a cambios en la cantidad de lluvia en ecosistemas áridos y semiáridos.     Electric resistance tomography and stress wave tomography for decay detection in trees—a comparison study

    Background To ensure the safety of trees, two NDT (nondestructive testing) techniques, electric resistance tomography and stress wave tomography, were employed to quantitatively detect and characterize the internal decay of standing trees. Comparisons between those two techniques were done to make full use of the individual capability for decay detection. Methods Eighty trees (40 Manchurian ash and 40 Populus simonii) were detected, then wood increment cores were obtained from each cross disc trial. The Dt, which was defined as the value determined by the mass loss ratio of each wood core, was regarded as the true severity of decay. Using ordinary least-squares regression to analyze the relationship between Dt and De (De was defined as the severity of decay determined by electric resistance tomography) and between Dt and Ds (Ds was defined as the severity of decay determined by stress wave tomography). Results The results showed that both methods could estimate the severity of decay in trees. In terms of different stages of decay, when Dt < 30%, De had a strong positive correlation with Dt (R2 = 0.677, P < 0.01), while, when Dt ≥ 30%, Ds had a significant positive correlation relationship with Dt (R2 = 0.645, P < 0.01). Conclusion Electric resistance tomography was better than stress wave tomography for testing in the early stages of decay, while stress wave tomography can be used effectively in the late stage of decay.
