    Understanding Status Update in Microblog: A Perspective on Media Needs

    Microblog has grown popularly as a seminal social medium for timely information seeking and sharing. However, the reason why individuals update real-time information in microblog has not been well understood, and empirical research to address this specific information behavior is hardly available. As a felt urge can be conceptualized as a precursor of real-time updating in the microblog, we attempt to capture the underlying mechanism in causing this less reflective behavior urge. We apply the media needs theory to investigate how the individualsā€™ media needs spark their urge to update personal status in the microblog. In particular, we conceptualize the cognitive needs as related to information uniqueness, personal integrative needs as related to connectivity, social integrative needs as a unidirectional relationship, affective needs as positive emotions and tension release needs as negative emotions. An online survey was employed to validate the proposed model within 523 microblog users in China. The results suggest that the usersā€™ behavior urge is significantly influenced by information uniqueness, connectivity, unidirectional relationship and positive emotions. Furthermore, among the five media needs, the affective and social integrative related factors strongly determine the personal real-time status update in microblog. The theoretical and practical implications are discussed in this paper

    Research progress in plant molecular systematics of lauraceae

    Lauraceae is a large family of woody plants with high ecological and economic value. The tribal and generic division and phylogenetic relationship of Lauraceae have long been contro-versial. Based on morphological and molecular evidence, phylogenetic relationships within the Cinnamomeae, Laureae and Perseeae tribes, also called ā€˜the Core Lauraceaeā€™, have arisen particular attention. In this review, we comprehensively collated the literatures on the phylogeny of Lau-raceae published in recent years and summarized progress made in molecular systematic researches employing gene fragments, chloroplast genomes and DNA barcodings analyses. We clarified the phylogenetic relationships and main controversies of ā€˜the Core Lauraceaeā€™, the systemic position of fuzzy genera (Neocinnamomum, Caryodaphnopsis and Cassytha) and the development of chloroplast genome and DNA barcodes. We further suggested and proposed the whole genome analysis and different inflorescence types would be possible to provide more information for further research on phylogenetic relationships and taxonomy of Lauraceae

    Characterization of the Small RNA Transcriptomes of Androgen Dependent and Independent Prostate Cancer Cell Line by Deep Sequencing

    Given the important roles of miRNA in post-transcriptional regulation and its implications for cancer, characterization of miRNA facilitates us to uncover molecular mechanisms underlying the progression of androgen-independent prostate cancer (PCa). The emergence of next-generation sequencing technologies has dramatically changed the speed of all aspects of sequencing in a rapid and cost-effective fashion, which can permit an unbiased, quantitive and in-depth investigation of small RNA transcriptome. In this study, we used high-throughput Illumina sequencing to comprehensively represent the full complement of individual small RNA and to characterize miRNA expression profiles in both the androgen dependent and independent Pca cell line. At least 83 miRNAs are significantly differentially expressed, of which 41 are up-regulated and 42 are down-regulated, indicating these miRNAs may be involved in the transition of LNCaP to an androgen-independent phenotype. In addition, we have identified 43 novel miRNAs from the androgen dependent and independent PCa library and 3 of them are specific to the androgen-independent PCa. Function annotation of target genes indicated that most of these differentially expressed miRNAs tend to target genes involved in signal transduction and cell communication, epically the MAPK signaling pathway. The small RNA transcriptomes obtained in this study provide considerable insights into a better understanding of the expression and function of small RNAs in the development of androgen-independent prostate cancer

    Distribution of organochlorine pesticides and polychlorinated biphenyls in Ny-ƅlesund of the Arctic

    Sediment, moss and animal-dropping samples were collected from Ny-ƅlesund of the Arctic to investigate the distribution of organochlorine pesticides (OCPs) and polychlorinated biphenyls (PCBs) in the Arctic area. OCPs and PCBs were analyzed using the capillary gas chromatography (GC-ECD). Recoveries of three surrogates were at a range of 69.5ļ½ž92.2% (mean: 78.8% ) , 75.1ļ½ž112.1% (98.5%) and 67.7ļ½ž95.1% (79.5%) , and the relative standard deviations were 6.7% , 8.0% and 11.3% , respectively. The major monomers of OCPs were hexachlorocyclohexanes (HCHs) and DDTs. Their concentrations were at a range of 0.86ļ½ž4.50 ng/g (mean: 2.24 ng/g) and 0.22ļ½ž1.09 ng/g (mean: 0.55 ng/g). The concentrations of PCBs were 0.20ļ½ž3.25 ng/g (mean: 0.84 ng/g) and the major congeners were tetra-, penta-, and hexa-PCBs, which were up to 23.8%态26.7% and 32.3% of total PCB s, respectively. Based on the results of the ratio analysis and the principal component analysis, the atmospheric transport is believed be one of the major input ways of OCPs and PCB s in the Ny-ƅlesund of the Arctic

    Improving the Efficacy of Conventional Therapy by Adding Andrographolide Sulfonate in the Treatment of Severe Hand, Foot, and Mouth Disease: A Randomized Controlled Trial

    Background. Herb-derived compound andrographolide sulfonate (called Xiyanping injection) recommended control measure for severe hand, foot, and mouth disease (HFMD) by the Ministry of Health (China) during the 2010 epidemic. However, there is a lack of good quality evidence directly comparing the efficacy of Andrographolide Sulfonate combination therapy with conventional therapy. Methods. 230 patients were randomly assigned to 7ā€“10 days of Andrographolide Sulfonate 5ā€“10ā€‰mg/Kg/day and conventional therapy, or conventional therapy alone. Results. The major complications occurred less often after Andrographolide Sulfonate (2.6% versus 12.1%; risk difference [RD], 0.94; 95% CI, 0.28ā€“1.61; P=0.006). Median fever clearance times were 96 hours (CI, 80 to 126) for conventional therapy recipients and 48 hours (CI, 36 to 54) for Andrographolide Sulfonate combination-treated patients (Ļ‡2=16.57, P<0.001). The two groups did not differ in terms of HFMD-cause mortality (P=1.00) and duration of hospitalization (P=0.70). There was one death in conventional therapy group. No important adverse event was found in Andrographolide Sulfonate combination therapy group. Conclusions. The addition of Andrographolide Sulfonate to conventional therapy reduced the occurrence of major complications, fever clearance time, and the healing time of typical skin or oral mucosa lesions in children with severe HFMD

    mirTools: microRNA profiling and discovery based on high-throughput sequencing

    miRNAs are small, non-coding RNA that negatively regulate gene expression at post-transcriptional level, which play crucial roles in various physiological and pathological processes, such as development and tumorigenesis. Although deep sequencing technologies have been applied to investigate various small RNA transcriptomes, their computational methods are far away from maturation as compared to microarray-based approaches. In this study, a comprehensive web server mirTools was developed to allow researchers to comprehensively characterize small RNA transcriptome. With the aid of mirTools, users can: (i) filter low-quality reads and 3/5ā€² adapters from raw sequenced data; (ii) align large-scale short reads to the reference genome and explore their length distribution; (iii) classify small RNA candidates into known categories, such as known miRNAs, non-coding RNA, genomic repeats and coding sequences; (iv) provide detailed annotation information for known miRNAs, such as miRNA/miRNA*, absolute/relative reads count and the most abundant tag; (v) predict novel miRNAs that have not been characterized before; and (vi) identify differentially expressed miRNAs between samples based on two different counting strategies: total read tag counts and the most abundant tag counts.     Massive nutrients offshore transport off the Changjiang Estuary in flooding summer of 2020

    Flood events significantly increase water discharges and terrigenous material inputs to coastal waters. Riverine nutrients in the Changjiang Estuary are transported by the dispersion of Changjiang Diluted Water (CDW) plumes and detached low-salinity water patches. However, the effects of flooding on nutrient offshore transports have not been well explored. Here, we present the nutrient conditions in the Changjiang Estuary and adjacent East China Sea in the historical flooding year 2020. Comparisons of nutrient distributions between flooding years, drought year and non-flooding years were also made. Our results showed that nitrate flux from the Changjiang River in August 2020 was 1.5 times that of the multi-year averaged flux in non-flooding years. Enormous riverine nutrient input resulted in much higher nutrient concentrations in the outer estuary than those in non-flooding years. In addition, a detached low-salinity water patch was observed, which made the salinity of the northern estuary even lower than that in the historical flooding year 1998. Surface dissolved inorganic nitrate (DIN) level in the low-salinity water patch was even ~16 times of that at nearby station in the drought year 2006. While phosphate (PO43āˆ’) concentrations were less than 0.1 Ī¼mol Lāˆ’1 east of 123Ā°E, which was probably caused by intensive biological uptake, as indicated by a high Chlorophyll a (Chl a) concentration (29.08 Ī¼g Lāˆ’1). The depleted PO43āˆ’ and high N/P of the low-salinity water patch suggested PO43āˆ’ limitation even under flood conditions. A three end-member mixing model was adopted to identify the contributions of the CDW end-member (CDWend-member) and biological process to nutrient distributions. Our model results showed that the nutrient contribution of the CDWend-member to the estuary (122ā€“124Ā°E, 31ā€“32.5Ā°N) in flooding year 2020 was over double that in drought year 2006. Model-derived biological DIN uptake was as high as 24.65 Ī¼mol Lāˆ’1 at the low-salinity water patch. Accordingly, the estimated net community production was 566ā€“1131 mg C māˆ’2 dāˆ’1 within the euphotic zone. The offshore transport of a low-salinity, high-DIN water patch during flooding could probably have a significant influence on biogeochemical cycles in the broad shelf, and even the adjacent Japan Sea

    Discussion on the Waterproof and Drainage System of the Coastal Tunnel and Analysis of Water Pressure Law outside Lining: A Case Study of the Gongbei Tunnel

    With the orderly promotion of the infrastructure construction in China, the number of coastal tunnels is increasing, but the coastal environment is accompanied by a large amount of groundwater and the designs of the waterproof and drainage system of coastal tunnels have always been a hot topic in the industry. If the waterproof and drainage system designs of the coastal tunnels are improper, the tunnel structure will easily be damaged and the stability and service life of the whole project will be affected. Based on the Gongbei tunnel project of the Hong Kong-Zhuhai-Macao Bridge, this paper discusses the designs of the waterproof and drainage system, and the waterproof detail structures of the Gongbei tunnel. Indoor similarity tests were carried out to study the law of the free water discharge of the tunnel under variable water heads, the relationship between the water pressure outside the lining and the water discharge, the relationship between the water pressure outside the pipe roof and the water discharge, and the distribution of water pressure around the tunnel. The reliability of the indoor similarity test was verified by the field test. The results show that the whole waterproof system should be adopted in the Gongbei tunnel, that is, grouting ringā€‰+ā€‰pipe roof freezing ringā€‰+ā€‰initial liningā€‰+ā€‰waterproof boardā€‰+ā€‰tertiary lining. In a certain range, the greater the water head, the greater the free water discharge of the tunnel. When the water head is large, the free drainage tends to a fixed value. When the tunnel is completely plugged, the water pressure outside the tunnel is distributed from the hydrostatic pressure. When the tunnel adopts drainage measures, the water pressure outside the lining will be reduced to a certain extent even the water discharge is very small. The above research can provide a reference for the design of the waterproof and drainage system similar to the coastal tunnel in the future
