    Inovação na diplomacia cultural: o caso da China

    This study focuses on the innovation of China’s cultural diplomacy (CCD) by means of the Confucius Institute (CI). The main contents revolve around the following research goals: 1) to understand the strategic framework and practical path of CCD, and to clarify the context of its inheritance and innovation; 2) to analyze whether the CI, epitomized as a crucial innovation of CCD, has improved China’s national image in Portuguese-speaking countries (PSCs) and enhanced the attraction and international competitiveness of Chinese culture; and 3) to explore how China can better formulate its CD strategy in line with the exigencies of the modern era. The study combines the methods of literature review, case study, and questionnaire research to explore the topics from different perspectives to strengthen the scientific nature of the research results. In addition to the introduction and conclusion, the thesis is divided into five chapters. Chapter 1 discusses the connotation and value of CD. Chapter 2 expounds the development and innovation of CCD. Chapter 3 systematically summarizes China’s cultural interaction in its diplomatic process with PSCs. Chapter 4 elaborates on the CI in terms of its operation mode and diplomatic means. Chapter 5 forms the core of the study and involves empirical analysis of case-study and questionnaire data. It aims to investigate the functions, public image, influence, and practical means of CIs in the process of CD. Major findings indicate that CIs in PSCs have achieved ideal social feedback and play a positive role in shaping the image of China. However, according to the different continents where CIs are located, the survey results show distinct characteristics which are closely related to China’s different foreign policies towards Latin America, Europe, and Africa and are determined by the historical experiences and national conditions of the various countries. The future task for CCD is to clarify China’s institutional roots and the cultural genes behind its development by using cultural exchanges and China’s fluid culture to convey a message of China’s pursuit of peace, development, and cooperation.Este estudo tem como foco a inovação da Diplomacia Cultural da China (DCC) através do Instituto Confúcio (IC). Os principais conteúdos giram em torno dos seguintes objetivos de investigação: 1) compreender o enquadramento estratégico e o percurso prático da Diplomacia Cultural da China e clarificar o contexto da sua herança e inovação; 2) analisar se o IC, exemplo de inovação crucial da Diplomacia Cultural da China, melhorou a imagem nacional da China nos Países de Língua Oficial Portuguesa (PALOP) e aumentou a atração e competitividade internacional da cultura chinesa; e 3) explorar como a China pode formular melhor a sua estratégia de Diplomacia Cultural, de acordo com as exigências da era moderna. O estudo combina os métodos de revisão de literatura, estudo de caso e pesquisa de questionário para explorar os tópicos de diferentes perspetivas com o objetivo de fortalecer a natureza científica dos resultados de investigação. Além da introdução e da conclusão, a tese está dividida em cinco capítulos. O Capítulo 1 discute a conotação e o valor de Diplomacia Cultural (DC). O Capítulo 2 expõe o desenvolvimento e a inovação da Diplomacia Cultural da China (DCC). O Capítulo 3 resume sistematicamente a interação cultural da China no seu processo diplomático com os Países de Língua Oficial Portuguesa (PALOP). O Capítulo 4 discorre sobre o papel do IC em termos do seu modo de operar e dos seus meios diplomáticos. O Capítulo 5 constitui o núcleo da tese e envolve a análise empírica dos dados do estudo de caso e do questionário. Tem como objetivo investigar as funções, a imagem pública, a influência e os meios práticos dos IC no processo de Diplomacia Cultural (DC). As principais descobertas indicam que os IC nos PALOP alcançaram o feedback social ideal e desempenham um papel positivo na formação da imagem da China. No entanto, de acordo com os diferentes continentes onde os IC estão situados, os resultados da pesquisa apresentam caraterísticas distintas que estão intimamente relacionadas com as diferentes políticas externas da China para a América Latina, a Europa e a África e são determinadas pelas experiências históricas e pelas condições nacionais dos vários países. A futura tarefa da Diplomacia Cultural da China (DCC) será a de esclarecer as raízes institucionais da China e os genes culturais por trás do seu desenvolvimento, usando intercâmbios culturais e a cultura fluida da China para transmitir uma mensagem de busca de paz, de desenvolvimento e de cooperação por parte da China.Programa Doutoral em Políticas Pública

    Fine mapping of a QTL for ear size on porcine chromosome 5 and identification of high mobility group AT-hook 2 (HMGA2) as a positional candidate gene

    <p>Abstract</p> <p>Background</p> <p>Ear size and shape are distinct conformation characteristics of pig breeds. Previously, we identified a significant quantitative trait locus (QTL) influencing ear surface on pig chromosome 5 in a White Duroc × Erhualian F<sub>2 </sub>resource population. This QTL explained more than 17% of the phenotypic variance.</p> <p>Methods</p> <p>Four new markers on pig chromosome 5 were genotyped across this F<sub>2 </sub>population. RT-PCR was performed to obtain expression profiles of different candidate genes in ear tissue. Standard association test, marker-assisted association test and F-drop test were applied to determine the effects of single nucleotide polymorphisms (SNP) on ear size. Three synthetic commercial lines were also used for the association test.</p> <p>Results</p> <p>We refined the QTL to an 8.7-cM interval and identified three positional candidate genes i.e. <it>HMGA2</it>, <it>SOX5 </it>and <it>PTHLH </it>that are expressed in ear tissue. Seven SNP within these three candidate genes were selected and genotyped in the F<sub>2 </sub>population. Of the seven SNP, <it>HMGA2 </it>SNP (JF748727: g.2836 A > G) showed the strongest association with ear size in the standard association test and marker-assisted association test. With the F-drop test, F value decreased by more than 97% only when the genotypes of <it>HMGA2 </it>g.2836 A > G were included as a fixed effect. Furthermore, the significant association between g.2836 A > G and ear size was also demonstrated in the synthetic commercial Sutai pig line. The haplotype-based association test showed that the phenotypic variance explained by <it>HMGA2 </it>was similar to that explained by the QTL and at a much higher level than by <it>SOX5</it>. More interestingly, <it>HMGA2 </it>is also located within the dog orthologous chromosome region, which has been shown to be associated with ear type and size.</p> <p>Conclusions</p> <p><it>HMGA2 </it>was the closest gene with a potential functional effect to the QTL or marker for ear size on chromosome 5. This study will contribute to identify the causative gene and mutation underlying this QTL.</p

    Imputation-Based Whole-Genome Sequence Association Study Reveals Constant and Novel Loci for Hematological Traits in a Large-Scale Swine F2 Resource Population

    The whole-genome sequences of progenies with low-density single-nucleotide polymorphism (SNP) genotypes can be imputed with high accuracy based on the deep-coverage sequences of key ancestors. With this imputation technology, a more powerful genome-wide association study (GWAS) can be carried out using imputed whole-genome variants and the phenotypes of interest to overcome the shortcomings of low-power detection and the large confidence interval derived from low-density SNP markers in classic association studies. In this study, 19 ancestors of a large-scale swine F2 White Duroc × Erhualian population were deeply sequenced for their genome with an average coverage of 25×. Considering 98 pigs from 10 different breeds with high-quality deep sequenced genomes, we imputed the whole genomic variants of 1020 F2 pigs genotyped by the PorcineSNP60 BeadChip with high accuracy and obtained 14,851,440 sequence variants after quality control. Based on this, 87 novel quantitative traits loci (QTLs) for 18 hematological traits at three different physiological stages of the F2 pigs were identified, among which most of the novel QTLs have been repeated in two of the three stages. Literature mining pinpointed that the FGF14 and LCLAT1 genes at SSC11 and SSC3 may affect the MCH at day 240 and MCV at day 18, respectively. The present study shows that combining high-quality imputed genomic variants and correlated phenomic traits into GWAS can improve the capability to detect QTL considerably. The large number of different QTLs for hematological traits identified at multiple growth stages implies the complexity and time specificity of these traits

    Evolution of Sequence Type 4821 Clonal Complex Hyperinvasive and Quinolone-Resistant Meningococci

    Expansion of quinolone-resistant Neisseria meningitidis clone ChinaCC4821-R1-C/B from sequence type (ST) 4821 clonal complex (CC4821) caused a serogroup shift from serogroup A to serogroup C invasive meningococcal disease (IMD) in China. To determine the relationship among globally distributed CC4821 meningococci, we analyzed whole-genome sequence data from 173 CC4821 meningococci isolated from 4 continents during 1972-2019. These meningococci clustered into 4 sublineages (1-4); sublineage 1 primarily comprised of IMD isolates (41/50, 82%). Most isolates from outside China (40/49, 81.6%) formed a distinct sublineage, the Europe-USA cluster, with the typical strain designation B:P1.17-6,23:F3-36:ST-3200(CC4821), harboring mutations in penicillin-binding protein 2. These data show that the quinolone-resistant clone ChinaCC4821-R1-C/B has expanded to other countries. The increasing distribution worldwide of serogroup B CC4821 raises the concern that CC4821 has the potential to cause a pandemic that would be challenging to control, despite indirect evidence that the Trumenba vaccine might afford some protection

    Identification of loci affecting teat number by genome-wide association studies on three pig populations

    Objective Three genome-wide association studies (GWAS) and a meta-analysis of GWAS were conducted to explore the genetic mechanisms underlying variation in pig teat number. Methods We performed three GWAS and a meta-analysis for teat number on three pig populations, including a White Duroc×Erhualian F2 resource population (n = 1,743), a Chinese Erhualian pig population (n = 320) and a Chinese Sutai pig population (n = 383). Results We detected 24 single nucleotide polymorphisms (SNPs) that surpassed the genome-wide significant level on Sus Scrofa chromosomes (SSC) 1, 7, and 12 in the F2 resource population, corresponding to four loci for pig teat number. We highlighted vertnin (VRTN) and lysine demethylase 6B (KDM6B) as two interesting candidate genes at the loci on SSC7 and SSC12. No significant associated SNPs were identified in the meta-analysis of GWAS. Conclusion The results verified the complex genetic architecture of pig teat number. The causative variants for teat number may be different in the three population

    A Missense Mutation in PPARD Causes a Major QTL Effect on Ear Size in Pigs

    Chinese Erhualian is the most prolific pig breed in the world. The breed exhibits exceptionally large and floppy ears. To identify genes underlying this typical feature, we previously performed a genome scan in a large scale White Duroc × Erhualian cross and mapped a major QTL for ear size to a 2-cM region on chromosome 7. We herein performed an identical-by-descent analysis that defined the QTL within a 750-kb region. Historically, the large-ear feature has been selected for the ancient sacrificial culture in Erhualian pigs. By using a selective sweep analysis, we then refined the critical region to a 630-kb interval containing 9 annotated genes. Four of the 9 genes are expressed in ear tissues of piglets. Of the 4 genes, PPARD stood out as the strongest candidate gene for its established role in skin homeostasis, cartilage development, and fat metabolism. No differential expression of PPARD was found in ear tissues at different growth stages between large-eared Erhualian and small-eared Duroc pigs. We further screened coding sequence variants in the PPARD gene and identified only one missense mutation (G32E) in a conserved functionally important domain. The protein-altering mutation showed perfect concordance (100%) with the QTL genotypes of all 19 founder animals segregating in the White Duroc × Erhualian cross and occurred at high frequencies exclusively in Chinese large-eared breeds. Moreover, the mutation is of functional significance; it mediates down-regulation of β-catenin and its target gene expression that is crucial for fat deposition in skin. Furthermore, the mutation was significantly associated with ear size across the experimental cross and diverse outbred populations. A worldwide survey of haplotype diversity revealed that the mutation event is of Chinese origin, likely after domestication. Taken together, we provide evidence that PPARD G32E is the variation underlying this major QTL
