    FLT3L and Plerixafor Combination Increases Hematopoietic Stem Cell Mobilization and Leads to Improved Transplantation Outcome

    AbstractHematopoietic stem cell (HSC) transplantation has curative potential for patients with hematological malignancies. Clinically, HSCs derived from mobilized peripheral blood are used more frequently than bone marrow. However, current standard mobilizing agents yield grafts that may not contain sufficient HSCs. Here, using murine models, we discovered that FLT3L synergized with plerixafor to mobilize phenotypically defined HSCs and their combination (FP) was superior to granulocyte colony-stimulating factor (G-CSF) alone or in combination with plerixafor (GP). Additionally, FP mobilized more regulatory T cells, natural killer cells, and plasmacytoid dendritic cells compared with G-CSF alone or GP. Both syngeneic and allogeneic grafts mobilized by FP led to long-term survival in transplanted mice. Collectively, FP represents a promising novel and potent mobilization regimen with potential clinical application in both the autologous and allogeneic transplantation settings

    Experimental Study and Application of Inorganic Solidified Foam Filling Material for Coal Mines

    Spontaneous combustion of residual coal in a gob due to air leakage poses a major risk to mining safety. Building an airtight wall is an effective measure for controlling air leakage. A new type of inorganic solidified foam-filled material was developed and its physical and chemical properties were analyzed experimentally. The compressive strength of this material increased with the amount of sulphoaluminate cement. With an increasing water–cement ratio, the initial setting time was gradually extended while the final setting time firstly shortened and then extended. The change in compressive strength had the opposite tendency. Additionally, as the foam expansion ratio increased, the solidification time tended to decrease but the compressive strength remained approximately constant. With an increase in foam production, the solidification time increased and the compressive strength decreased exponentially. The results can be used to determine the optimal material ratios of inorganic solidified foam-filled material for coal mines, and filling technology for an airtight wall was designed. A field application of the new material demonstrated that it seals crossheadings tightly, leaves no fissures, suppresses air leakage to the gob, and narrows the width of the spontaneous combustion and heat accumulation zone

    Dynamic neural network of insight: a functional magnetic resonance imaging study on solving Chinese 'chengyu' riddles.

    The key components of insight include breaking mental sets and forming the novel, task-related associations. The majority of researchers have agreed that the anterior cingulate cortex may mediate processes of breaking one's mental set, while the exact neural correlates of forming novel associations are still debatable. In the present study, we used a paradigm of answer selection to explore brain activations of insight by using event-related functional magnetic resonance imaging during solving Chinese 'chengyu' (in Chinese pinyin) riddles. Based on the participant's choice, the trials were classified into the insight and non-insight conditions. Both stimulus-locked and response-locked analyses are conducted to detect the neural activity corresponding to the early and late periods of insight solution, respectively. Our data indicate that the early period of insight solution shows more activation in the middle temporal gyrus, the middle frontal gyrus and the anterior cingulate cortex. These activities might be associated to the extensive semantic processing, as well as detecting and resolving cognitive conflicts. In contrast, the late period of insight solution produced increased activities in the hippocampus and the amygdala, possibly reflecting the forming of novel association and the concomitant "Aha" feeling. Our study supports the key role of hippocampus in forming novel associations, and indicates a dynamic neural network during insight solution
