    Summary: This study aimed to investigate the effect of cardiopulmonary bypass (CPB) on Annexin A1 expression in the peripheral blood mononuclear cells (PBMCs) of children with congenital heart disease (CHD). A total of 30 children receiving CPB for interventricular septal defect were included. Peripheral blood was collected before and after CPB. PBMCs were collected by density gradient centrifugation. Protein extra ction was performed by lysis and subjected to 2D-QUANT for protein quantitation. Isoelectric focusing electrophoresis (IEF) was carried out followed by gel image analysis. Protein spots with a difference in expression of >1.5 fold were collected as candidate proteins which were subjected to mass spectrometry for the identification of diffe rentially expressed proteins. Western blot assay was emplo yed to confirm the expressions of target proteins. Peripheral blood collected at two time points was subjected to two-dimensional electrophoresis, and a total of 12 differentially expressed proteins were identified. Of them, 5 proteins had decreased expression before CPB (T0) but their expressions increased after CPB (T1); the remaining 7 proteins had in cre ased expres sions before CPB but their expressions re du ced after CPB. One of these differentially expres sed proteins was Annexin A1. Western blot assay confirmed that Annexin A1 expression began to increase at 0.5 h after CPB, and the increase of Annexin A1 was more obvious after CPB. Our findings primarily indicate the potential mechanism underlying the role of PBMC in inflammatory response following CPB, and provide a target for the prevention and control of post-CPB systemic inflammatory response syndrome (SIRS). Keywords: annexin A1, cardiopulmonary bypass, peripheral blood mononuclear cells, congenital heart disease Kratak sadr`aj: Svrha studije bila je da se istra`i uticaj kardiopulmonarnog bajpasa (KPB) na ekspresiju aneksina A1 u mononuklearnim }elijama periferne krvi (M]PK) dece sa uro|enim sr~anim oboljenjima (SO). Istra`ivanjem je obuhva}eno ukupno 30 dece kod koje je zbog interventrikularnog septalnog defekta izveden kardiopulmonarni bajpas. Uzorci periferne krvi uzeti su pre i posle zahvata. M]PK su sakupljene centrifugiranjem u gradijentu gustine. Proteini su ekstra hovani lizom a kvantitacija proteina obavljena je metodom 2D-QUANT. Izvr{ena je elektroforeza izoelektri~nim fokusiranjem (EIF) a potom analiza gel image. Proteinske ta~ke sa razlikom u ekspresiji >1,5 puta uzete su kao proteini kandidati, koji su podvrgnuti masenoj spektrometriji radi identifikovanja razli~ite ekspresije proteina. Za potvr|ivanje ekspresija ciljnih proteina upotrebljen je test Western blot. Uzorci periferne krvi uzeti su dva puta u odre|enom vremenskom razmaku i podvrgnuti dvodimenzionalnoj elektroforezi te je identifikovano ukupno 12 diferencijalno izra`enih proteina. Od toga je 5 proteina imalo smanjenu ekspresiju pre KPB (T0), ali se njihova ekspresija posle KPB pove}ala (T1); preostalih 7 proteina imalo je pove}anu ekspresiju pre KPB, ali se njihova ekspresija smanjila posle KPB. Jedan od ovih diferencijalno izra`enih proteina bio je aneksin A1. Testom Western blot potvr|eno je da je ekspresija aneksina A1 poela da raste 0,5 h posle KPB, dok je porast aneksina A1 posle KPB bio upadljiviji. Na{i nalazi prevashodno ukazuju na potencijalni mehanizam uloge M]PK u inflamatornom odgo voru posle KPB i pru`aju metu za prevenciju i kontrolu sindroma post-KPB sistemskog inflamatornog odgovora (SSIO).     A Prospective Study of the Incidence of Retinopathy of Prematurity in China: Evaluation of Different Screening Criteria

    To investigate the incidence of Retinopathy of Prematurity (ROP) in Beijing, North China, and to evaluate the effectiveness of different ROP screening criteria, we conducted a prospective cohort study in a single-neonatal intensive care unit (NICU). A total of 2997 premature infants with birth weight (BW) ≤ 2000 g and/or gestational age (GA) ≤ 34 weeks had completed ROP screening. ROP was diagnosed in 356 (11.9%) infants. The mean GA was 30.46 ± 1.98 weeks and the mean BW was 1477.35 ± 371.29 g. Of the 59 (2.0%) infants receiving treatment, the mean GA was 29.37 ± 2.10 weeks, and the mean BW was 1240.80 ± 330.71 g. The incidence of ROP declined from 14.7% in 2009 and 11.1% in 2010 to 9.5% in 2011. The United Kingdom (UK) criteria could reduce the screening number by 40.8%, and 3 infants with type I ROP needing treatment were missed, but none in 2011. The United States (US) criteria could reduce the screening number by 66.5%, and 10 infants with type I ROP needing treatment were missed, including one in 2011. So the UK criteria may be appropriate for screening of ROP in our NICU in 2011. Future multisite epidemiologic studies are required to establish suitable ROP screening criteria in China

    Izraženost surfaktantnog proteina B u bronhoalveolarnom ispirku terminske novorođenčadi sa sindromom respiracijskog distresa

    The aim was to investigate the surfactant protein B (SP-B) expression in the bronchoalveolar lavage fluid (BALF ) of full-term neonates with respiratory distress syndrome (RD S). The enzyme-linked immunosorbent assay was performed to assess SP-B expression in BALF of 60 full-term neonates with RD S and 23 healthy neonates and correlation of SP-B level with RD S classification according to chest x-ray findings and PaO2/FiO2 before mechanical ventilation in neonates with RD S. The SP-B level was significantly lower in the RD S group (17.63±6.80 ng/mL) than in healthy neonates (103.95±6.38 ng/mL) (P<0.001). The SP-B level correlated positively with PaO2/ FiO2 before mechanical ventilation (r=0.838, P<0.001). Moreover, the lower the SP-B level, the more severe was the RD S as determined by chest x-ray (P<0.001). In conclusion, full-term neonates with RD S had reduced SP-B in BALF , which was related to the severity of RD S, suggesting that SP-B supplement may be an effective strategy in the treatment of RD S in full-term neonates.Cilj studije bio je ispitati izraženost surfaktantnog proteina B (SP-B) u bronhoalveolarnom ispirku (BALF ) terminske novorođenčadi sa sindromom respiracijskog distresa (SRD ). Izraženost SP-B određena je testom ELI SA u BALF 60 terminske novorođenčadi sa SRD i 23 zdrave novorođenčadi. Utvrđena je korelacija razine SP-B s klasifikacijom SRD prema rendgenskoj snimci prsišta i vrijednosti PaO2/FiO2 prije mehaničke ventilacije u novorođenčadi sa SRD . U skupini novorođenčadi sa SRD razina SP-B bila je značajno niža (17,63±6,80 ng/mL) od one u zdrave novorođenčadi (103,95±6,38 ng/ mL) (P<0,001). Utvrđena je pozitivna korelacija razine SP-B i PaO2/FiO2 prije mehaničke ventilacije (r=0,838, P<0,001). Štoviše, što je bila niža razina SP-B, to je teži bio SRD procijenjen prema rendgenskoj snimci prsišta (P<0,001). Zaključuje se da terminska novorođenčad sa SRD ima sniženu razinu SP-B u BALF i to je povezano s težinom SRD . Ovi nalazi ukazuju na to da bi dodatak SP-B mogla biti učinkovita strategija u liječenju SRD kod terminske novorođenčadi

    Association of thyroid-stimulating hormone and lipid levels with suicide attempts among adolescents with major depressive disorder in China

    ObjectivesEvidence suggested that thyroid-stimulating hormone (TSH) and lipid levels were associated with major depressive disorder (MDD). However, its role in suicide attempts in adolescents with MDD was unclear. This investigation was to probe into the relationship between TSH, lipid levels, and attempted suicide in adolescents with MDD.MethodsA total of 179 adolescents with MDD were included from January 2021 to January 2022. Socio-demographic data and clinical data were obtained through self-made questionnaires. TSH and lipid levels were analyzed by a recognized laboratory. The Epidemiological Studies Depression Scale (CES-D) was used to assess the severity of depression. Questions about whether there was a behavior of suicide attempts were completed through conversation interviews.ResultsResults showed that the CES-D total score, TSH, and low-density lipoprotein cholesterol (LDL-C) of suicide attempters were significantly higher than those of non-suicide attempters. Ordinary family relationships were more likely to attempt suicide than good family relationships. The CES-D total score, ordinary family relationships, TSH and LDL-C were still significant in binary logistic regression, with an adjusted odds ratio (OR) of 1.04, 3.42, 5.14, and 1.76, respectively. The area under the receiver operating characteristic (ROC) curve showed that the area under the ROC curve (AUC) ranged from 0.60 to 0.74.ConclusionSuicide attempts are common among adolescents with MDD and were associated with CES-D total score, ordinary family relationships, TSH, and LDL-C. Given the association between suicide attempts and TSH and LDL-C, the dynamic changes in TSH and LDL-C levels should be detected regularly

    Molecular Cloning and Characterization of Two Genes Encoding Dihydroflavonol-4-Reductase from Populus trichocarpa

    Dihydroflavonol 4-reductase (DFR, EC 1.1.1.219) is a rate-limited enzyme in the biosynthesis of anthocyanins and condensed tannins (proanthocyanidins) that catalyzes the reduction of dihydroflavonols to leucoanthocyanins. In this study, two full-length transcripts encoding for PtrDFR1 and PtrDFR2 were isolated from Populus trichocarpa. Sequence alignment of the two PtrDFRs with other known DFRs reveals the homology of these genes. The expression profile of PtrDFRs was investigated in various tissues of P. trichocarpa. To determine their functions, two PtrDFRs were overexpressed in tobacco (Nicotiana tabacum) via Agrobacterium-mediated transformation. The associated color change in the flowers was observed in all 35S:PtrDFR1 lines, but not in 35S:PtrDFR2 lines. Compared to the wild-type control, a significantly higher accumulation of anthocyanins was detected in transgenic plants harboring the PtrDFR1. Furthermore, overexpressing PtrDFR1 in Chinese white poplar (P. tomentosa Carr.) resulted in a higher accumulation of both anthocyanins and condensed tannins, whereas constitutively expressing PtrDFR2 only improved condensed tannin accumulation, indicating the potential regulation of condensed tannins by PtrDFR2 in the biosynthetic pathway in poplars

    Summary Background: The aim of this study was to investigate the role of pulmonary surfactant-associated protein B (SP-B) expression in the pathogenesis of neonatal respiratory distress syndrome (RDS) via detecting the protein and mRNA expression of SP-B. Methods: A total of 60 unrelated neonates who died of RDS were chosen as the RDS group and then subgrouped into ≤32 weeks group, 32∼37 weeks group and ≥37 weeks group (n=20). Sixty neonates who died of other diseases were enrolled as controls and subdivided into 3 matched groups based on the gestational age. Western blot assay and RT-PCR were employed. Results: In the RDS group, SP-B protein expression was reduced or deficient in 8 neonates of which 6 had no SP-B protein expression. In the control group, only 1 had reduced SP-B protein expression. The reduced or deficient SP-B protein expression in 9 neonates of both groups was noted in the ≥37 weeks group. In the RDS group, the SP-B mRNA expression was significantly lower than that in the control group. In the ≤37 weeks group, SP-B mRNA expression was comparable between the RDS group and control group. In the 32∼37 weeks group, the SP-B mRNA expression in the RDS group was significantly reduced when compared with the control group. In the ≥37 weeks group, the SP-B mRNA expression in the RDS group was dramatically lower than that in the control group. Conclusions: Alteration of SP-B expression is present at transcriptional and translational levels. Reduction of SP-B mRNA and protein expression is involved in the pathogenesis of RDS

    This study was to investigate the effect of intra-amniotic administration of pulmonary surfactant (PS) on lung maturation in conditions of experimentally induced premature rupture of membranes (PROM) and intrauterine infection of rabbits. To establish animal (rabbit) models of intrauterine infection caused by PROM, E. coli was intrauterinely injected in 24-and 26-day pregnant animals. Twenty healthy pregnancy adult Japanese white rabbits were divided into three groups: the infection group (8 rabbits), infection group with intra-amniotic PS administration (8 rabbits) and healthy controls (4 rabbits). Ultrastructure changes in the lung were observed under an electron microscope 19.5 hours after intervention. The results showed that the lung infection levels of fetal rabbits in the infection group and the infection group with PS had no significant difference. Bacillus bodies and infiltrated inflammatory cells can be occasionally seen in the alveoli, bronchial lumen, and cytoplasm. The type II alveolar epithelial cells (AT-II) were decreased in the infection group than that in control group. Lamellar bodies showed vacuolation changes and different levels of apoptosis. In the infection group with PS, the count of AT-II did not show an obvious decrease. Lamellar bodies increased, and different secretion forms appeared. Also, there was little apoptosis and no obvious collagen fiber hyperplasia in antenatal administration of PS group. We believe that once PROM has happened, intrauterine infection and inflammation stimulated a cascade reaction on the fetal lung, leading to abnormal changes in the alveolar ultrastructure.     A 15-day-old neonate with complete transposition of the great arteries/intact ventricular septum was admitted with life-threatening hypoxemia and heart arrest. After successful resuscitation, heart beat recovered but blood lactate began to arise and maintained above 15 mmol/L 6 hours later. Emergency arterial switch operation was done at 20 hours after resuscitation. Planned extracorporeal membrane oxygenation support was employed postoperatively. The baby experienced severe pulmonary hemorrhage and severe hypoxemia after weaning from cardiopulmonary bypass, which were treated with extracorporeal membrane oxygenation support

    Candida utilis candidaemia in premature infants: a retrospective single-centre study

    This retrospective study was conducted in a neonatal intensive care unit in Beijing. Patients whose blood culture yielded Candida utilis during hospitalisation from January 2009 to December 2017 were enrolled. Thirteen preterm infants of median gestational age 29.85 weeks were included. Laboratory tests on the day of onset showed thrombocytopaenia in 11 patients, granulocytopaenia in eight and elevated C-reactive protein in seven. No fungal endophthalmitis, renal infection, carditis or involvement of other end organs was observed in any of the cases. All 13 patients were cured after fluconazole therapy
