    Development and Validation of the Quality-of-Life Assessment System for Lung Cancer Based on Traditional Chinese Medicine

    Traditional Chinese Medicine (TCM) has many unique features. Thequality-of-life (QoL) instrument for lung cancer based on Traditional Chinese Medicine (QLASTCM-Lu) was the first self-reported instrument specifically developed to assess the quality of life from the perspective of TCM. Structured group methods and theory in developmental rating scale were employed to establish a general and a specific module, respectively. Quantitative and qualitative data from 240 lung cancer patients were collected to assess the psychometric properties. The three identified scales of the QLASTCM-Lu (correspondence between man and universe, unity of the body and spirit, and lung cancer specific module) and the total score demonstrated excellent psychometric properties. Test-retest reliability of all domains ranged from 0.93 to 0.96, and internal consistency α ranged from 0.86 to 0.93. Correlation and factor analysis demonstrated good construct validity. Significant differences in the QLASTCM-Lu scales and total score were found among groups differing in TCM syndrome, supporting the clinical sensitivity of the QLASTCM-Lu. Statistically significant changes were found for each scale and the total score. Responsiveness of the QLASTCM-Lu was greater than that of QLQ-LC43. The QLASTCM-Lu is a psychometrically sound and clinically sensitive measure of quality of life for lung cancer patients, which can be applied to both TCM therapy and Western medicine therapy

    Coseeded Schwann cells myelinate neurites from differentiated neural stem cells in neurotrophin-3-loaded PLGA carriers

    Biomaterials and neurotrophic factors represent promising guidance for neural repair. In this study, we combined poly-(lactic acid-co-glycolic acid) (PLGA) conduits and neurotrophin-3 (NT-3) to generate NT-3-loaded PLGA carriers in vitro. Bioactive NT-3 was released stably and constantly from PLGA conduits for up to 4 weeks. Neural stem cells (NSCs) and Schwann cells (SCs) were coseeded into an NT-releasing scaffold system and cultured for 14 days. Immunoreactivity against Map2 showed that most of the grafted cells (>80%) were differentiated toward neurons. Double-immunostaining for synaptogenesis and myelination revealed the formation of synaptic structures and myelin sheaths in the coculture, which was also observed under electron microscope. Furthermore, under depolarizing conditions, these synapses were excitable and capable of releasing synaptic vesicles labeled with FM1-43 or FM4-64. Taken together, coseeding NSCs and SCs into NT-3-loaded PLGA carriers increased the differentiation of NSCs into neurons, developed synaptic connections, exhibited synaptic activities, and myelination of neurites by the accompanying SCs. These results provide an experimental basis that supports transplantation of functional neural construction in spinal cord injury

    Mechanistic aspects of photo-induced formation of peroxide ions on the surface of cubic Ln(2)O(3) (Ln = Nd, Sm, Gd) under oxygen

    National Basic Research Program of China [2010CB732303]; National Natural Science Foundation of China [21173173, 21033006, 20923004]; Program for Changjiang Scholars and Innovative Research Team in University [IRT1036]The photo-induced formation of peroxide ions on the surface of cubic Ln(2)O(3) (Ln = Nd, Sm, Gd) was studied by in situ microprobe Raman spectroscopy using a 325 nm laser as excitation source. It was found that the Raman bands of peroxide ions at 833-843 cm(-1) began to grow at the expense of the Ln(3+)-O-2 bands at 333-359 cm(-1) when the Ln(2)O(3) samples under O-2 were continuously irradiated with a focused 325 nm laser beam at temperatures between 25-150 degrees C. The intensity of the peroxide Raman band was found to increase with increasing O-2 partial pressure, whereas no peroxide band was detected on the Ln(2)O(3) under N-2 as well as on the samples first irradiated with laser under Ar or N-2 followed by exposure to O-2 in the dark. The experiments using O-18 as a tracer further confirmed that the peroxide ions are generated by a photo-induced reaction between O-2 and the lattice oxygen (O2-) species in Ln(2)O(3). Under the excitation of 325 nm UV light, the transformation of O-2 to peroxide ions on the surface of the above lanthanide sesquioxides can even take place at room temperature. Basicity of the lattice oxygen species on Ln(2)O(3) also has an impact on the peroxide formation. Higher temperature or laser irradiation power is required to initiate the reaction between O-2 and O2- species of weaker basicity

    Disruption of TGF-ÎČ Signaling Improves Ocular Surface Epithelial Disease in Experimental Autoimmune Keratoconjunctivitis Sicca

    TGF-ÎČ is a pleiotropic cytokine that can have pro- or anti-inflammatory effects depending on the context. Elevated levels of bioactive TGF-ÎČ1 in tears and elevated TGF-ÎČ1mRNA transcripts in conjunctiva and minor salivary glands of human Sjögren's Syndrome patients has also been reported. The purpose of this study was to evaluate the response to desiccating stress (DS), an experimental model of dry eye, in dominant-negative TGF-ÎČ type II receptor (CD4-DNTGFÎČRII) mice. These mice have a truncated TGF-ÎČ receptor in CD4(+) T cells, rendering them unresponsive to TGF-ÎČ.DS was induced by subcutaneous injection of scopolamine and exposure to a drafty low humidity environment in CD4-DNTGFÎČRII and wild-type (WT) mice, aged 14 weeks, for 5 days. Nonstressed (NS) mice served as controls. Parameters of ocular surface disease included corneal smoothness, corneal barrier function and conjunctival goblet cell density. NS CD4-DNTGFÎČRII at 14 weeks of age mice exhibited a spontaneous dry eye phenotype; however, DS improved their corneal barrier function and corneal surface irregularity, increased their number of PAS+ GC, and lowered CD4(+) T cell infiltration in conjunctiva. In contrast to WT, CD4-DNTGFÎČRII mice did not generate a Th-17 and Th-1 response, and they failed to upregulate MMP-9, IL-23, IL-17A, RORÎłT, IFN-Îł and T-bet mRNA transcripts in conjunctiva. RAG1KO recipients of adoptively transferred CD4+T cells isolated from DS5 CD4-DNTGFÎČRII showed milder dry eye phenotype and less conjunctival inflammation than recipients of WT control.Our results showed that disruption of TGF-ÎČ signaling in CD4(+) T cells causes paradoxical improvement of dry eye disease in mice subjected to desiccating stress
