    Identification of cyclic AMP-phosphodiesterase variants from the PDE4D gene expressed in human peripheral mononuclear cells

    AbstractTo determine whether the expression of different PDE4D variants is unique to the rat or conserved through evolution, we have characterized the different PDE4D mRNAs expressed in human peripheral blood mononuclear cells. RT-PCR was performed using primers based on rat sequences and mRNAs from mononuclear cells. The specifically amplified fragments had a size identical to that predicted for rat PDE4D1, PDE4D2 and PDE4D3. Sequencing confirmed that these fragments are derived from the human PDE4D gene. Their sequence was highly homologous to that reported for the rat variants. cDNAs corresponding to the entire ORF of human PDE4D2 and PDE4D3 were expressed in mammalian cells, causing a large increase in PDE activity. Western blot analysis of human peripheral blood mononuclear cell extracts demonstrated the presence of proteins corresponding to the recombinant PDE4D1 and PDE4D2. The pattern of splicing and different promoter usage of the PDE4D gene is therefore conserved during evolution, which indicates an important physiological role

    A chromosomelevel genome assembly of the Asian arowana, Scleropages formosus

    Asian arowana (Scleropages formosus), an ancient teleost belonging to the Order Osteoglossomorpha, has been a valuable ornamental fish with some varieties. However, its biological studies and breeding germplasm have been remarkably limited by the lack of a reference genome. To solve these problems, here we report high-quality genome sequences of three common varieties of Asian arowana (the golden, red and green arowana). We firstly generated a chromosome-level genome assembly of the golden arowana, on basis of the genetic linkage map constructed with the restriction site-associated DNA sequencing (RAD-seq). In addition, we obtained draft genome assemblies of the red and green varieties. Finally, we annotated 22,016, 21,256 and 21,524 protein-coding genes in the genome assemblies of golden, red and green varieties respectively. Our data were deposited in publicly accessible repositories to promote biological research and molecular breeding of Asian arowana

    Series AC Arc Fault Detection Method Based on High-Frequency Coupling Sensor and Convolution Neural Network

    Arc faults induced by residential low-voltage distribution network lines are still one of the main causes of residential fires. When a series arc fault occurs on the line, the value of the fault current in the circuit is limited by the load. Traditional circuit protection devices cannot detect series arcs and generate a trip signal to implement protection. This paper proposes a novel high-frequency coupling sensor for extracting the features of low-voltage series arc faults. This sensor is used to collect the high-frequency feature signals of various loads in series arc state and normal working state. The signal will be transformed into two-dimensional feature gray images according to the temporal-domain sequence. A neural network with a three-layer structure based on convolution neural network is designed, trained and tested using the various typical loads’ arc states and normal states data sets composed of these images. This detection method can simultaneously accurately identify series arc, as well as the load type. Seven different domestic appliances were selected for experimental verification, including a desktop computer, vacuum cleaner, induction cooker, fluorescent lamp, dimmer, heater and electric drill. Then, the stability and universality of the detection algorithm is also verified by using electronic load with adjustable power factor and peak factor. The experimental results show that the designed sensor has the advantages of simple structure and wide frequency response range. The detection algorithm comparison confirms that the classification accuracy of the seven domestic appliances’ work states in the fourteen categories could reach 98.36%. The adjustable load in the two categories could reach above 99%. The feasibility of hardware implementation based on FPGA of this method is also evaluated

    The energy use behavior analysis method can dig out the user’s energy use behavior rules from the energy use big data, thereby improving the quality of the grid-side management service in the integrated energy system. Firstly, it summarizes the characteristics of the integrated energy system and constructs the integrated energy system service system; secondly, it summarizes the data-driven electricity consumption behavior analysis research model. Then, it elaborates on the collection and aggregation of electricity consumption information, and refined user classification. Next, the comprehensive application of energy consumption behavior analysis in load forecasting, demand response modeling and other typical scenarios is deeply analyzed. Finally, the challenges that may be encountered in further research are clarified and the follow-up work is prospected

    Asymmetric Unwrapping of Nucleosomes under Tension Directed by DNA Local Flexibility

    SummaryDynamics of the nucleosome and exposure of nucleosomal DNA play key roles in many nuclear processes, but local dynamics of the nucleosome and its modulation by DNA sequence are poorly understood. Using single-molecule assays, we observed that the nucleosome can unwrap asymmetrically and directionally under force. The relative DNA flexibility of the inner quarters of nucleosomal DNA controls the unwrapping direction such that the nucleosome unwraps from the stiffer side. If the DNA flexibility is similar on two sides, it stochastically unwraps from either side. The two ends of the nucleosome are orchestrated such that the opening of one end helps to stabilize the other end, providing a mechanism to amplify even small differences in flexibility to a large asymmetry in nucleosome stability. Our discovery of DNA flexibility as a critical factor for nucleosome dynamics and mechanical stability suggests a novel mechanism of gene regulation by DNA sequence and modifications

    Periodic DNA patrolling underlies diverse functions of Pif1 on R-loops and G-rich DNA

    Pif1 family helicases are conserved from bacteria to humans. Here, we report a novel DNA patrolling activity which may underlie Pif1’s diverse functions: a Pif1 monomer preferentially anchors itself to a 3′-tailed DNA junction and periodically reel in the 3′ tail with a step size of one nucleotide, extruding a loop. This periodic patrolling activity is used to unfold an intramolecular G-quadruplex (G4) structure on every encounter, and is sufficient to unwind RNA-DNA heteroduplex but not duplex DNA. Instead of leaving after G4 unwinding, allowing it to refold, or going beyond to unwind duplex DNA, Pif1 repeatedly unwinds G4 DNA, keeping it unfolded. Pif1-induced unfolding of G4 occurs in three discrete steps, one strand at a time, and is powerful enough to overcome G4-stabilizing drugs. The periodic patrolling activity may keep Pif1 at its site of in vivo action in displacing telomerase, resolving R-loops, and keeping G4 unfolded during replication, recombination and repair
