    A datamining approach to identifying spatial patterns of phosphorus forms in the Stormwater Treatment Areas in the Everglades

    The Everglades ecosystem in Florida, USA, is naturally phosphorus (P) limited, and faces threats of ecosystem change and associated losses to habitat, biodiversity, and ecosystem function if subjected to high inflows of P and other nutrients. In addition to changes in historic hydropattern, upstream agriculture (sugar cane, vegetable, citrus) and urbanization has placed the Everglades at risk due to nutrient-rich runoff. In response to this threat, the Stormwater Treatment Areas (STAs) were constructed along the northern boundary of the Everglades as engineered ecological systems designed to retain P from water flowing into the Everglades. This research investigated data collected over a period from 2002 to 2014 from the interior of the STAs using data mining and analysis techniques including (a) exploratory methods such as Principal Component Analysis to test for patterns and groupings in the data, and (b) modelling approaches to test for predictive relationships between environmental variables. The purpose of this research was to reveal and compare spatial trends and relationships between environmental variables across the various treatment cells, flow-ways, and STAs. Common spatial patterns and their drivers indicated that the flow-ways do not function along simple linear gradients; instead forming zonal patterns of P distribution that may increasingly align with the predominant flow path over time. Findings also indicate that the primary drivers of the spatial distribution of P in many of these systems relate to soil characteristics. The results suggest that coupled cycles may be a key component of these systems; i.e. the movement and transformation of P is coupled to that of nitrogen (N)

    Harmonised soil data across Europe with a 1:250 000 geo-referenced soil database will allow for exchange of data across member states and the provide the information needed by the European Commission and European Environment Agency for reporting on issues relating to soil quality under a fu-ture Soil Framework Directive. Within this context, the Environmental Protection Agency of the Republic of Ireland commissioned a project run by Teagasc to produce a 1:250 000 soil map of the Republic of Ire-land.     Interaction patterns of methoprene-tolerant and germ cell-expressed Drosophila JH receptors suggest significant differences in their functioning

    Methoprene-tolerant (Met) and germ cell-expressed (Gce) proteins were shown to be juvenile hormone (JH) receptors of Drosophila melanogaster with partially redundant functions. We raised the question of where the functional differentiation of paralogs comes from. Therefore, we tested Met and Gce interaction patterns with selected partners. In this study, we showed the ability of Gce and its C-terminus (GceC) to interact with 14-3-3 in the absence of JH. In contrast, Met or Met C-terminus (MetC) interactions with 14-3-3 were not observed. We also performed a detailed structural analysis of Met/Gce interactions with the nuclear receptor fushi tarazu factor-1 (Ftz-F1) ligand-binding domain. We showed that GceC comprising an Ftz-F1-binding site and full-length protein interacts with Ftz-F1. In contrast to Gce, only MetC (not full-length Met) can interact with Ftz-F1 in the absence of JH. We propose that the described differences result from the distinct tertiary structure and accessibility of binding sites in the full-length Met/Gce. Moreover, we hypothesize that each interacting partner can force disordered MetC and GceC to change the structure in a partner-specific manner. The observed interactions seem to determine the subcellular localization of Met/Gce by forcing their translocation between the nucleus and the cytoplasm, which may affect the activity of the proteins. The presented differences between Met and Gce can be crucial for their functional differentiation during D. melanogaster development and indicate Gce as a more universal and more active paralog.     Damage Detection in Tensegrity using Interacting Particle-Ensemble Kalman Filter

    The 10th EWSHM, originally scheduled for the first week of July 2020, is planned to be held the first week of July 2022 in Palermo.International audienceTensegrity structures form a special class of truss with dedicated cables and bars, that take tension and compression, respectively. To ensure equilibrium, the tensegrity members are required to be prestressed. Over prolonged usage, the cables may lose their prestress while bars may buckle, affecting the structural stiffness as well as its dynamic properties. The stiffness of tensegrities also vary with the load even in the absence of damage. This can potentially mask the effect of damage leading to a false impression of tensegrity health. This poses a major challenge in tensegrity health monitoring especially when the load is stochastic and unknown. Present study develops a vibration based output-only time-domain approach for monitoring the health of any tensegrity in the presence of uncertainties due to ambient force and measurement noise. An Interacting Particle Ensemble Kalman Filter (IPEnKF) has been used that can efficiently monitor tensegrity health from contaminated response data. IPEnKF combines a bank of Ensemble Kalman Filters to estimate response states while running within a Particle Filter envelop that estimates a set of location based health parameters. Further to make damage detection cheaper, strain responses are used as measurements.     SILAC Analysis Reveals Increased Secretion of Hemostasis-Related Factors by Senescent Cells

    Cellular senescence irreversibly arrests cell proliferation, accompanied by a multi-component senescence-associated secretory phenotype (SASP) that participates in several age-related diseases. Using stable isotope labeling with amino acids (SILACs) and cultured cells, we identify 343 SASP proteins that senescent human fibroblasts secrete at 2-fold or higher levels compared with quiescent cell counterparts. Bioinformatic analysis reveals that 44 of these proteins participate in hemostasis, a process not previously linked with cellular senescence. We validated the expression of some of these SASP factors in cultured cells and in vivo. Mice treated with the chemotherapeutic agent doxorubicin, which induces widespread cellular senescence in vivo, show increased blood clotting. Conversely, selective removal of senescent cells using transgenic p16-3MR mice showed that clearing senescent cells attenuates the increased clotting caused by doxorubicin. Our study provides an in-depth, unbiased analysis of the SASP and unveils a function for cellular senescence in hemostasis

    The serum zinc concentration as a potential biological marker in patients with major depressive disorder

    Despite many clinical trials assessing the role of zinc in major depressive disorder (MDD), the conclusions still remain ambiguous. The aim of the present clinical study was to determine and comparison the zinc concentration in the blood of MDD patients (active stage or remission) and healthy volunteers (controls), as well as to discuss its potential clinical usefulness as a biomarker of the disease. In this study 69 patients with current depressive episode, 45 patients in remission and 50 controls were enrolled. The zinc concentration was measured by electrothermal atomic absorption spectrometry (ET AAS). The obtained results revealed, that the zinc concentration in depressed phase were statistically lower than in the healthy volunteers [0.89 vs. 1.06 mg/L, respectively], while the zinc level in patients achieve remission was not significantly different from the controls [1.07 vs. 1.06 mg/L, respectively]. Additionally, among the patients achieve remission a significant differences in zinc concentration between group with and without presence of drug-resistance in the previous episode of depression were observed. Also, patients in remission demonstrated correlation between zinc level and the average number of depressive episodes in the last year. Serum zinc concentration was not dependent on atypical features of depression, presence of psychotic symptoms or melancholic syndrome, age, age of onset or duration of disease, number of episodes in the life time, duration of the episode/remission and severity of depression measured by the Hamilton Rating Scale for Depression (HDRS), and the Montgomery-Asberg Depression Rating Scale (MADRS). Concluding, our findings confirm the correlation between zinc deficit present in the depressive episode, and are consistent with the majority of previous studies. These results may also indicate that serum zinc concentration might be considered as a potential biological marker of MDD

    Transcriptional Profiling of Bacillus anthracis Sterne (34F2) during Iron Starvation

    Lack of available iron is one of many environmental challenges that a bacterium encounters during infection and adaptation to iron starvation is important for the pathogen to efficiently replicate within the host. Here we define the transcriptional response of B. anthracis Sterne (34F2) to iron depleted conditions. Genome-wide transcript analysis showed that B. anthracis undergoes considerable changes in gene expression during growth in iron-depleted media, including the regulation of known and candidate virulence factors. Two genes encoding putative internalin proteins were chosen for further study. Deletion of either gene (GBAA0552 or GBAA1340) resulted in attenuation in a murine model of infection. This attenuation was amplified in a double mutant strain. These data define the transcriptional changes induced during growth in low iron conditions and illustrate the potential of this dataset in the identification of putative virulence determinants for future study
