    Comportamento de Biomphalaria glabrata variantes albina e melânica frente à infecção pelo Schistosoma mansoni

    The behaviour of the albino and melanic variants of Biomphalaria glabrata of Belo Horizonte (MG. Brazil) was studied comparatively, in terms of their respective susceptibilities to infection by Schistosoma mansoni of the same origin, through observation of the elimination of cercariae for a three-month period and the calculation of mortality and infection rates, in control and in infected snails. The number of amoebocytes, granulocytes and hyalinocytes in the circulating hemolymph during different periods of infection was analyzed. The evolution of the infection in the tissues was observed by means of histological cross-sections. The melanic variant showed greater susceptibility to infection and a higher mortality rate. The albino variant showed a higher number of circulating amoebocytes, both granulocytes and hyalinocytes. A higher number of degenerated sporocysts were seen in the histological cross-sections of the albino variant. The results suggest that the melanic variant of B. glabrata was more susceptible to infection by S. mansoni than was the albino variant.Foi estudado comparativamente o comportamento das variantes albina e melânica de Biomphalaria glabrata de Belo Horizonte (MG. Brasil), em relação à susceptibilidade à infecção pelo Schistosoma mansoni de mesma origem, através da observação da eliminação de cercárias por um período de três meses e a verificação das taxas de mortalidade e infecção nos moluscos controles e infectados. Analisou-se também, o número de amebócitos granulócitos e hialinócitos na hemolinfa circulante em diferentes períodos de infecção. A evolução da infecção nos tecidos foi observada através de cortes histológicos. A variante melânica apresentou maior susceptibilidade à infecção e maior taxa de mortalidade. A variante albina apresentou maior número de amebócitos circulantes, tanto granulócitos quanto hialinócitos. Nos cortes histológicos foi verificado maior número de esporocistos degenerados na variedade albina. Por estes resultados sugere-se que nas populações estudadas a variante melânica de B. glabrata, foi mais susceptível à infecção pelo S. mansoni.21722

    The attraction exerted by Biomphalaria glabrata infected with Angiostrongylus costaricensis on Schistosoma mansoni miracidia of the BH strain was tested, using a glass apparatus composed by two circular chambers connected to a channel. One mollusk or a sample of a snail conditioning water (SCW) was placed in one of the chambers, randomly selected, and the remaining of the apparatus was filled with chlorine-free water. Ten miracidia were placed in the centre of the channel and their behaviour was observed for 15 minutes. Ten replicates were made for each treatment (snail or SCW), using different specimens of mollusks or different samples of SCW. Mollusks infected with A. costaricensis attracted significantly less S. mansoni miracidia than non-infected B. glabrata. In addition, miracidia were also significantly more attracted to SCW from mollusks infected with A. costaricensis

    Gerações de Biomphalaria glabrata e Biomphalaria tenagophila selecionadas geneticamente para resistência e suscetibilidade ao Schistosoma mansoni das linhagens BH e SJ foram utilizadas no estudo da adaptação do trematódeo ao hospedeiro intermediário. As gerações dos planorbídeos foram obtidas por autofecundação dos moluscos que se apresentaram suscetíveis ou resistentes após a exposição aos miracídios de Schistosoma mansoni. Para Biomphalaria glabrata foram obtidas as gerações: Parental, F1S (Suscetível), F1R (Resistente), F2S e F2R. Para a Biomphalaria tenagophila foram estudadas as gerações: Parental, F1S, F1R e F50S. A comparação das taxas de infecção apresentadas pelas diferentes gerações mostrou que, em ambas as espécies, o aumento da suscetibilidade foi mais facilmente obtido do que o aumento da resistência. A dificuldade em aumentar a resistência do molusco ao S. mansoni tem fortes implicações epidemiológicas.<br>Generations of Biomphalaria glabrata and Biomphalaria tenagophila selected genetically for resistance and susceptibility to Schistosoma mansoni of strains BH and SJ were used in a study of the trematode adaptation to the intermediate host. Descendants of the planorbids were obtained by self-fertilization of the mollusks that became susceptible or resistant after exposure to the miracidia of Schistosoma mansoni. For Biomphalaria glabrata they were obtained from the following generations: Parental, F1S (Susceptible), F1R (Resistant), F2S and F2R. For Biomphalaria tenagophila the studied generations were: Parental, F1S, F1R and F50S. The comparison of the infection rates presented by the different generations showed that the increase in susceptibility was more easily obtained in both species. The difficulty in increasing the resistance of the mollusks to Schistosoma mansoni has important epidemiologic implications

    Twenty mice were exposed to cercariae from mollusks treated with hydrocortisone and another 20 mice received cercariae from non-treated mollusks. The behavior of the parasites from the two groups of mollusks was compared based on the ability of cercariae to penetrate mice, on the total number of worms recovered after eight weeks of infection, on the relationship between the number of penetrating cercariae and the number of recovered worms and on the number of eggs in the feces. Treating the mollusks with hydrocortisone did not alter the ability of cercariae to penetrate mice nor did it affect the total number of worms recovered. The number of female worms, the number of coupled worms and the number of eggs in the feces were greater in mice infected by cercariae from mollusks treated with hydrocortisone.<br>Vinte camundongos foram expostos a cercárias oriundas de moluscos tratados com hidrocortisona e outros vinte receberam cercárias de moluscos não tratados. O comportamento dos parasitas dos dois grupos foi comparado com base na habilidade das cercárias em penetrar nos camundongos, no número total de vermes recuperados, após oito semanas de infecção, na relação entre o número de cercárias penetrantes e o número de vermes recuperados e o número de ovos nas fezes. O tratamento dos moluscos com hidrocortisona não alterou a habilidade das cercárias em penetrar nos camundongos nem afetou o número total de vermes recuperados. O número de vermes fêmeas, o número de vermes acasalados e o número de ovos nas fezes aumentaram em camundongos infectados por cercárias de moluscos tratados com hidrocortisona

    Genetic markers between Biomphalaria glabrata snails susceptible and resistant to Schistosoma mansoni infection

    The analysis of the genetic variability related to susceptibility to Schistosoma mansoni infection in the vector of the genus Biomphalaria is important in terms of a better understanding of the epidemiology of schistosomiasis itself, the possible pathological implications of this interaction in vertebrate hosts, and the formulation of new strategies and approaches for disease control. In the present study, the genetic variability of B. glabrata strains found to be resistant or susceptible to S. mansoni infection was investigated using DNA amplification by random amplified polymorphic DNA-polymerase chain reaction (RAPD-PCR). The amplification products were analyzed on 8% polyacrylamide gel and stained with silver. We selected 10 primers, since they have previously been useful to detect polymorphism among B. glabrata and/or B. tenagophila. The results showed polymorphisms with 5 primers. Polymorphic bands observed only in the susceptible strain. The RAPD-PCR methodology represents an adequate approach for the analysis of genetic polymorphisms. The understanding of the genetic polymorphisms associated to resistance may contribute to the future identification of genomic sequences related to the resistance/susceptibility of Biomphalaria to the larval forms of S. mansoni and to the development of new strategies for the control of schistosomiasis
