    The Important Marine Mammal Area network : a tool for systematic spatial planning in response to the marine mammal habitat conservation crisis

    Major support to the IMMA process was provided by the German Federal Ministry for the Environment, Nature Conservation and Nuclear Safety (BMU) on the basis of a decision adopted by the German Bundestag, through the Global Ocean Biodiversity Initiative (GOBI). The support from the Prince Albert II of Monaco Foundation, OceanCare, NRDC, the Animal Welfare Institute, and the Pacific Life Foundation is also gratefully acknowledged.The Important Marine Mammal Areas (IMMAs) initiative was launched by the Marine Mammal Protected Areas Task Force of the International Union for the Conservation of Nature in 2016, as a response to a conservation crisis in the protection of marine mammals and wider global ocean biodiversity. IMMAs identify discrete portions of habitat that are important for one or more marine mammal species, and that have the potential to be delineated and managed for conservation. They are identified by scientific experts during regional workshops, on the basis of satisfying one or more of eight criteria that capture critical aspects of marine mammal biology, ecology and population structure. Candidate IMMAs undergo independent scientific review prior to being accepted, and then are publicly available via a searchable and downloadable database and a dedicated online e-Atlas. Between 2016 and 2021, eight expert workshops - engaging more than 300 experts - have resulted in the identification of 173 IMMAs located in 90 countries or territories, across a third of the globe. IMMAs identified to date provide important habitats for 58 of the 131 recognized marine mammal species. Around two-thirds of all IMMAs (65%) were identified on the basis of important habitat for a marine mammal species that is threatened on the IUCN Red List. Approximately 61% of IMMA surface areas occur within Exclusive Economic Zone waters, while 39% fall within areas beyond national jurisdiction. The Task Force undertook implementation planning exercises for IMMAs in Palau (Micronesia), the Andaman Islands (India) and the Bazaruto Archipelago and Inhambane Bay (Mozambique), engaging with a range of stakeholders including government and management bodies. IMMAs are increasingly being utilized in environmental impact assessments, marine planning exercises and in international, national and supra-regional conservation, policy and management initiatives, including the Convention on Migratory Species and Convention on Biological Diversity, as well as the design and management of Marine Protected Areas (MPAs) and the extension of MPA networks.     Vulvar sarcomas: Short guideline for histopathological recognition and clinical management. Part 1

    Malignant tumors of the female reproductive system are a serious health and social problem, as they are the second most common cause of death among women, after breast cancer. Their incidence has increased dramatically during recent years, probably due to the different sexual habits and changes in the prevalence of HIV/ AIDS and HPV virus carriers, among other factors. Vulvar tumors represent only 4% of all gynecological neoplasms, and they are fourth in frequency after tumors of the cervix, uterus, and ovary. Ninety eight percent of all vulvar tumors are benign and only 2% are malignant. The overall incidence of tumors with vulvar location is between two and seven cases per 100,000 women, and it increases with age, while the death rate is estimated at 0.7 per 100,000 women. Sarcomas of the vulva comprise approximately 1–3% of all vulvar cancers, with leiomyosarcomas, epithelioid sarcomas, and rhabdomyosarcomas being the most common among them. They are characterized by rapid growth, high metastatic potential, frequent recurrences, aggressive behavior, and high mortality rate. In this paper, we present the most common forms of sarcomas of the vulva (leiomyosarcoma, epithelioid sarcoma, malignant rhabdoid tumor, rhabdomyosarcoma) in order to emphasize the broad differential diagnosis, rare appearance, non-specific clinical picture, aggressive course, and high mortality

    Further characterization of ADAMTS-13 inactivation by thrombin

    Background: The multimeric size and platelet-tethering function of von Willebrand factor (VWF) are modulated by the plasma metalloprotease, a disintegrin and metalloproteinase with a thrombospondin type 1 motif, member 13 (ADAMTS-13). In vitro ADAMTS-13 is susceptible to proteolytic inactivation by thrombin. Objectives: In this study, we aimed to characterize the inactivation of ADAMTS-13 by thrombin and to assess its physiological significance. Methods and results: By N-terminal sequencing of cleavage products, and by mutagenesis, we identified the principal thrombin cleavage sites in ADAMTS-13 as R257 and R1176. Using a library of 76 thrombin mutants, we highlighted the functional importance of exosite I on thrombin in the proteolysis of ADAMTS-13. Proteolysis of ADAMTS-13 by thrombin caused an 8-fold reduction in its affinity for VWF that contributed to its loss of VWF-cleaving function. Intriguingly, thrombin-cleaved ADAMTS-13 both bound and proteolyzed a short recombinant VWF A2 domain substrate (VWF115) normally. Following activation of coagulation in normal plasma, endogenous ADAMTS-13, but not added ADAMTS-13, appeared resistant to coagulation-induced fragmentation. An estimation of the Km for ADAMTS-13 proteolysis by thrombin was appreciably higher than the physiological concentration of ADAMTS-13. This was corroborated by the comparatively low affinity of ADAMTS-13 for thrombin (KD 95 nm). Conclusions: Together, our data suggest that ADAMTS-13 is protected from rapid proteolytic inactivation by thrombin in normal plasma. Whether this remains the case under pathological situations involving elevated/sustained generation of thrombin remains unclear

    A Conformation-Sensitive Monoclonal Antibody against the A2 Domain of von Willebrand Factor Reduces Its Proteolysis by ADAMTS13

    The size of von Willebrand factor (VWF), controlled by ADAMTS13-dependent proteolysis, is associated with its hemostatic activity. Many factors regulate ADAMTS13-dependent VWF proteolysis through their interaction with VWF. These include coagulation factor VIII, platelet glycoprotein 1bα, and heparin sulfate, which accelerate the cleavage of VWF. Conversely, thrombospondin-1 decreases the rate of VWF proteolysis by ADAMTS13 by competing with ADAMTS13 for the A3 domain of VWF. To investigate whether murine monoclonal antibodies (mAbs) against human VWF affect the susceptibility of VWF to proteolysis by ADAMTS13 in vitro, eight mAbs to different domains of human VWF were used to evaluate the effects on VWF cleavage by ADAMTS13 under fluid shear stress and static/denaturing conditions. Additionally, the epitope of anti-VWF mAb (SZ34) was mapped using recombinant proteins in combination with enzyme-linked immunosorbent assay and Western blot analysis. The results indicate that mAb SZ34 inhibited proteolytic cleavage of VWF by ADAMTS13 in a concentration-dependent manner under fluid shear stress, but not under static/denaturing conditions. The binding epitope of SZ34 mAb is located between A1555 and G1595 in the central A2 domain of VWF. These data show that an anti-VWF mAb against the VWF-A2 domain (A1555-G1595) reduces the proteolytic cleavage of VWF by ADAMTS13 under shear stress, suggesting the role of this region in interaction with ADAMTS13

    Monitoring Winter and Summer Abundance of Cetaceans in the Pelagos Sanctuary (Northwestern Mediterranean Sea) Through Aerial Surveys

    Systematic long-term monitoring of abundance is essential to inform conservation measures and evaluate their effectiveness. To instigate such work in the Pelagos Sanctuary in the Mediterranean, two aerial surveys were conducted in winter and summer 2009. A total of 467 (131 in winter, 336 in summer) sightings of 7 species was made. Sample sizes were sufficient to estimate abundance of fin whales in summer (148; 95% CI = 87–254) and striped dolphins in winter (19,462; 95% CI = 12 939–29 273) and in summer (38 488; 95% CI = 27 447–53 968). Numbers of animals within the Sanctuary are significantly higher in summer, when human activities and thus potential population level impacts are highest. Comparisons with data from past shipboard surveys suggest an appreciable decrease in fin whales within the Sanctuary area and an appreciable increase in striped dolphins. Aerial surveys proved to be more efficient than ship surveys, allowing more robust estimates, with smaller CIs and CVs. These results provide essential baseline data for this marine protected area and continued regular surveys will allow the effectiveness of the MPA in terms of cetacean conservation to be evaluated and inform future management measures. The collected data may also be crucial in assessing whether ship strikes, one of the main causes of death for fin whales in the Mediterranean, are affecting the Mediterranean population

    Calcium modulates force sensing by the von Willebrand factor A2 domain

    von Willebrand factor (VWF) multimers mediate primary adhesion and aggregation of platelets. VWF potency critically depends on multimer size, which is regulated by a feedback mechanism involving shear-induced unfolding of the VWF-A2 domain and cleavage by the metalloprotease ADAMTS-13. Here we report crystallographic and single-molecule optical tweezers data on VWF-A2 providing mechanistic insight into calcium-mediated stabilization of the native conformation that protects A2 from cleavage by ADAMTS-13. Unfolding of A2 requires higher forces when calcium is present and primarily proceeds through a mechanically stable intermediate with non-native calcium coordination. Calcium further accelerates refolding markedly, in particular, under applied load. We propose that calcium improves force sensing by allowing reversible force switching under physiologically relevant hydrodynamic conditions.     Well-Being in First Year Medical Students

    Objective: This study explored the well-being, attitudes toward counseling, willingness to seek counseling, and coping strategies of first year medical students. Gender differences in attitudes toward and willingness to seek counseling were also explored. Methods: One hundred five first year medical students (98 % response rate) were administered a 59-item questionnaire about well-being, attitudes toward counseling, willingness to seek counseling, and coping strategies during the first week of medical school. The data were analyzed with hierarchical regression and multivariate analysis of covariance (MANCOVA). Results: Female medical students were less willing to seek counseling and had more negative attitudes toward counseling compared to male medical students. Most students indicated that they chose not to seek counseling because they did not feel a need for it. Three students reported that stigma prevented them from seeking counseling. Unhealthy coping strategies (denial, self-blame, and substance use) were negatively associated with well-being while healthy coping strategies (active coping, emotional support, and instrumental support) did not correlate with well-being. Conclusions: Medical schools should continue efforts to make counseling accessible. Conversations about counseling may help address the more negative attitudes of female students toward counseling, a finding which merits further investigation given that women typically have more positive attitudes toward counseling than men. Use of unhealthy coping strategies can be addressed in classes, clubs, and by advisors and mentors. Limitations of this study include that only first year medical students were surveyed and that it was a cross sectional study
