    The involvement of type IV pili and the phytochrome CphA in gliding motility, lateral motility and photophobotaxis of the cyanobacterium Phormidium lacuna

    Phormidium lacuna is a naturally competent, filamentous cyanobacterium that belongs to the order Oscillatoriales. The filaments are motile on agar and other surfaces and display rapid lateral movements in liquid culture. Furthermore, they exhibit a photophobotactic response, a phototactic response towards light that is projected vertically onto the area covered by the culture. However, the molecular mechanisms underlying these phenomena are unclear. We performed the first molecular studies on the motility of an Oscillatoriales member. We generated mutants in which a kanamycin resistance cassette (KanR) was integrated in the phytochrome gene cphA and in various genes of the type IV pilin apparatus. pilM, pilN, pilQ and pilT mutants were defective in gliding motility, lateral movements and photophobotaxis, indicating that type IV pili are involved in all three kinds of motility. pilB mutants were only partially blocked in terms of their responses. pilB is the proposed ATPase for expelling of the filament in type IV pili. The genome reveals proteins sharing weak pilB homology in the ATPase region, these might explain the incomplete phenotype. The cphA mutant revealed a significantly reduced photophobotactic response towards red light. Therefore, our results imply that CphA acts as one of several photophobotaxis photoreceptors or that it could modulate the photophobotaxis response

    The Iraq War has been the costliest U.S. conflict since the Vietnam War. To date, few official studies have been conducted to review what happened, why it happened, and what lessons should be drawn. The U.S. Army in the Iraq War is the Army's initial operational level analysis of this conflict, written in narrative format, with assessments and lessons embedded throughout the work. This study reviews the conflict from a Landpower perspective and includes the contributions of coalition allies, the U.S. Marine Corps, and special operations forces. Presented principally from the point of view of the commanders in Baghdad, the narrative examines the interaction of the operational and strategic levels, as well as the creation of theater level strategy and its implementation at the tactical level. Volume 1 begins in the truce tent at Safwan Airfield in southern Iraq at the end of Operation DESERT STORM and briefly examines actions by U.S. and Iraqi forces during the interwar years. The narrative continues by examining the road to war, the initially successful invasion, and the rise of Iraqi insurgent groups before exploring the country's slide toward civil war. This volume concludes with a review of the decision by the George W. Bush administration to "surge" additional forces to Iraq, placing the conduct of the "surge" and its aftermath in the second volume

    Validation of the Mayo Dysphagia Questionnaire

    While multiple instruments characterize upper gastrointestinal symptoms, a validated instrument devoted to the measurement of a spectrum of esophageal dysphagia attributes is not available. Therefore, we constructed and validated the Mayo Dysphagia Questionnaire (MDQ). The 27 items of the MDQ underwent content validity, feasibility, concurrent validity, reproducibility, internal consistency, and construct validity testing. To assess content validity, five esophageal subspecialty gastroenterologists reviewed the items to ensure inclusion of pertinent domains. Feasibility testing was done with eight outpatients who refined problematic items. To assess concurrent validity, 70 patient responses on the MDQ were compared to responses gathered in a structured patient-physician interview. A separate group of 70 outpatients completed the MDQ twice to assess the reproducibility of each item. A total of 148 patients participated in the validation process (78 [53%] men; mean age 62). On average, the MDQ took 6 minutes to complete. A single item (odynophagia) tested poorly with a kappa value of < 0.4. Otherwise, the majority of concurrent validity kappa values were in the good to excellent range with a mean of 0.63 (95% CI 0.22–0.89). The majority of reproducibility kappa values were also in the good to excellent range with a median kappa value of 0.76 (interquartile range: 0.67–0.81). Cronbach's alpha values were excellent in the range of 0.86–0.88. Spearman rank correlation coefficients to assess construct validity were also excellent in the range of 0.87–0.98. Thus, the MDQ is a concise instrument that demonstrates overall excellent concurrent validity, reproducibility, internal consistency, and construct validity for the features of esophageal dysphagia.Peer Reviewedhttp://deepblue.lib.umich.edu/bitstream/2027.42/71625/1/j.1442-2050.2007.00670.x.pd
