    Inhibitory mechanism of 3-hydroxypropionaldehyde accumulation in 1,3-propanediol synthesis with Klebsiella pneumoniae

    3-Hydroxypropionaldehyde accumulation may cause the cessation of 1,3-propanediol sustained production with glycerol by Klebsiella pneumoniae. The impeller tip speed shift from higher to lower speed at glycerol excess or the pulsed glycerol feeding could lead to an abrupt increase of the 3- hydroxypropionaldehyde concentration (up to 10 mmol/l) in 10 min. The intracellular consequence of the 3-hydroxypropionaldehyde accumulation has not yet been elucidated. The rapid accumulation of 3- hydroxypropionaldehyde relying on the impeller tip speed shift was employed to investigate the influences of 3-hydroxypropionaldehyde to the activities of nine key enzymes related to glycerol metabolism, CO2 and O2 levels in off-gas, cell growth and 1,3-propanediol synthesis. Compared with that at 1.19 mmol/l 3-hydroxypropionaldehyde in broth, the residual enzymatic activities of the nine key enzymes ranged from 9.44 to 74.68% in the cultures at 7.5 mmol/l 3-hydroxypropionaldehyde in broth. The inhibitions of cell growth and the 1,3-propanediol synthesis were unnoticeable at the low level of 3- hydroxypropionaldehyde. By contrast, the CO2 and O2 levels changes in off-gas response to the 3- hydroxypropionaldehyde accumulation were less than 15 min. These results suggest that 3- hydroxypropionaldehyde inhibited the growth and metabolism of K. pneumoniae in a more complicated manner.Keywords: Fermentation, glycerol, 3-hydroxypropionaldehyde, Klebsiella pneumoniae, 1,3-propanediol

    Design and fabrication of silicon nanowires towards efficient solar cells

    The recent rise of semiconductor nanowires opens new opportunities for realizing high efficiency photovoltaic devices at low cost due to the unique one-dimensional structure with remarkable electrical and optical properties. Particularly, silicon nanowires (SiNWs), as one of the most earth-abundant materials, have been investigated worldwide to develop cost-effective solar cells. Great efforts have been devoted to fabricating ordered/disordered SiNWs using cost-effective approaches and achieving optimized structural parameters, such as array periodicity, nanowire morphology, length and diameter. Systematic theoretical investigations along with experimental studies on optical and electrical properties of SiNWs have been carried out. These efforts have led to obtaining remarkable improvement of the power conversion efficiency of SiNW solar cells from 10% in the last few years. However, till now, the power conversion efficiency of these SiNW solar cells is far from satisfactory for any commercial applications compared with the traditional bulk silicon solar cells. Further development of SiNW solar cells requires better understanding of the optical and electrical properties of the nanowire solar cells. Improvement in fabrication of high quality nanowires in a controlled fashion also plays a significant role in nanowire solar cell design and fabrication. To guide future development of SiNW solar cells, the recent work on SiNWs is reviewed. Following that, various techniques aiming to achieve high quality nanowires at low cost are introduced. Both bottom-up and top-down techniques are discussed. Then, electrical properties and various types of solar cells based on SiNWs are discussed. Finally challenges and prospects of SiNW solar cells are presented

    Nuclear quadrupole resonance as a non-destructive testing tool

    Nuclear pure quadrupole resonance (NQR) is a resonance technique that provides distinctly different information from that provided by nuclear magnetic resonance (NMR). In NMR the splitting of the energy levels, and therefore the frequency observed, occurs because of the interaction of the nuclear magnetic moment with an external magnetic field. Information about the system under study comes from perturbations on this magnetic interaction. These perturbations lead to a broadening of the line, or to relaxation effects on the interchange of energy between the spins and the lattice, and among the spins. In NQR the primary interaction is between the electric quadrupole moment of a nucleus and the electric field gradient at that nucleus. The field gradient is provided by internal interactions in the sample itself, arising from the chemical bonds, rather than by an external field. Anything that changes the bonding environment, such as tensile stress, will cause shifts in the quadrupole resonance frequency. All nuclei with spin greater than 1/2 have a nuclear quadrupole moment, in addition to their magnetic moment. The nucleus used as an example in this paper is 75As, which has spin 3/2

    Phylogenetic position of the freshwater fish trypanosome, Trypanosoma ophiocephali (Kinetoplastida) inferred from the complete small subunit ribosomal RNA gene sequence

    The complete small subunit rRNA (SSrRNA) gene sequence (2,142 nucleotides) of the freshwater fish trypanosome Trypanosoma ophiocephali Chen (1964) was determined. The phylogenetic analysis deduced using neighbor-joining, maximum parsimony, and Bayesian methods demonstrated the existence of an “aquatic clade”. T. ophiocephali was revealed to be a member of the freshwater fish trypanosomes and form the sister species with Trypanosoma siniperca and Trypanosoma sp. Carpio with high bootstrap values (98% MP, 100% NJ, 100% Bay). The high similarity of SSrRNA gene sequences and morphometric characters showed that T. ophiocephali, T. siniperca and T. sp. Carpio probably were the same species. The phylogenetic trees further suggested that Chinese freshwater fish trypanosome might be paraphyletic, and fish trypanosomes should have low host specificity

    Measured and calculated seismic velocities and densities for granulites from xenolith occurrences and adjacent exposed lower crustal sections: A comparative study from the North China craton

    Granulites from the Neogene xenolith-bearing Hannuoba alkaline basalt and from the Manjinggou-Wayaokou exposed lower crustal section in the Archean Huai'an terrain, which occurs within and surrounds the Hannuoba basalt, provide a unique opportunity for a comparative study on petrophysical properties and composition of the lower crust represented by these two types of samples. P and S wave velocities and densities of 12 Hannuoba lower crustal xenoliths and one associated spinel lherzolite xenolith as well as nine granulites and granulite-facies metasedimentary rocks from the Archean Huai'an terrain were measured in laboratory at pressures up to 600 MPa and temperatures up to 600°C. Calculations of P and S wave velocities were also made for the same suite of samples based on modal mineralogy and single-crystal velocities whose variations with composition are considered by using microprobe analyses and velocities of end members. The measured and calculated Vp at room temperature and 600 MPa, where the microcrack effect is considered to be almost eliminated, agree within 4% for rocks from the Manjinggou-Wayaokou section and the adjacent Wutai-Jining upper crustal to upper lower crustal section. In contrast, the xenoliths show systematically lower measured Vp by up to 15% relative to calculated velocities, even if decompression-induced products of kelyphite and glass are taken into account. The lower measured velocities for xenoliths are attributed to grain boundary alteration and residual porosity. This implies that although granulite xenoliths provide direct information about lower crustal constitution and chemical composition, they are not faithful samples for studying in situ seismic properties of the lower crust in terms of measured velocities due to alterations during their entrainment to the surface, which changes their physical properties significantly. In this respect, granulites from high-grade terrains are better samples because they are not subjected to significant changes during their slow transport to the surface and because physical properties depend primarily on mineralogy in addition to pressure and temperature. On the other hand, calculated velocities for granulite xenoliths are consistent with velocities for granulites from terrains, suggesting that they can be also used to infer lower crust composition by correlating with results from seismic refraction studies.
