    Sesamin catechol glucuronides exert anti-inflammatory effects by suppressing IFN-ÎČ and iNOS expression through the deconjugation in macrophage-like J774.1 cells

    Sesamin, a representative sesame lignan, has health-promoting activities. Sesamin is converted into catechol derivatives and further into their glucuronides or sulfates in vivo, whereas the biological activities of sesamin metabolites remain unclear. We examined the inhibitory effects of sesamin metabolites on the lipopolysaccharide (LPS)-induced NO production in mouse macrophage-like J774.1 cells and found that a mono-catechol derivative SC1, (7α,7'α,8α,8'α)-3,4-dihydroxy-3',4'-methylenedioxy-7,9':7',9-diepoxylignane, has a much higher activity than sesamin and other metabolites. The inhibitory effects of SC1 glucuronides were time-dependently enhanced, associated with the intracellular accumulation of SC1 and the methylated form. SC1 glucuronides and SC1 attenuated the expression of inducible NO synthase (iNOS) and upstream interferon-ÎČ (IFN-ÎČ) in the LPS-stimulated macrophages. The inhibitory effects of SC1 glucuronides against NO production were canceled by the ÎČ-glucuronidase inhibitor and enhanced by the catechol- O-methyltransferase inhibitor. Our results suggest that SC1 glucuronides exert the anti-inflammatory effects by inhibiting the IFN-ÎČ/iNOS signaling through macrophage-mediated deconjugation

    A Retrospective Study of Canine Idiopathic Epilepsy in Referral Centers in Japan

    Idiopathic epilepsy (IE) is one of the most common neurologic disorder in dogs. The aim of this study was to describe the clinical data of dogs with IE in Japan and to search for predisposing factors of IE for further investigation. Medical records and clinical information of dogs diagnosed with IE at 2 referral centers between April 2013 and March 2016 were retrospectively reviewed. It was conducted according to the consensus statement published by International Veterinary Epilepsy Task Force. A total of 70 dogs were used with a median (range) weight of 5.15 (1.85–79.85) kg and age at initial epileptic seizure onset of 4.2 (0.3–11.8) years. Fortyfour dogs were male, and 26 were female. Toy Poodles were over-represented in the present study indicating that this breed may be predisposed to IE and would be a candidate for gene study to elucidate the cause of IE in dogs

    Knowledge, experience, and potential risks of dating violence among Japanese university students: a cross-sectional study

    <p>Abstract</p> <p>Background</p> <p>The Domestic Violence Prevention Act came into effect in Japan in 2001, but covers only marriage partner violence and post-divorce partner violence, and does not recognize intimate partner violence (IPV). The present study was performed to determine the experience of harassment, both toward and from an intimate partner, and recognition of harassment as IPV among Japanese university students.</p> <p>Methods</p> <p>A self-administered questionnaire survey regarding the experience of harassment involving an intimate partner was conducted as a cross-sectional study among freshman students in a prefectural capital city in Japan.</p> <p>Results</p> <p>A total of 274 students participated in the present study. About half of the subjects (both male and female students) had experience of at least one episode of harassment toward or had been the recipient of harassment from an intimate partner. However, the study participants did not recognize verbal harassment, controlling activities of an intimate partner, and unprotected sexual intercourse as violence. Experience of attending a lecture/seminar about domestic violence and dating violence did not contribute to appropriate help-seeking behavior.</p> <p>Conclusions</p> <p>An educational program regarding harassment and violence prevention and appropriate help-seeking behavior should be provided in early adolescence to avoid IPV among youth.</p

    Molecular analysis of the BCR-ABL1 kinase domain in chronic-phase chronic myelogenous leukemia treated with tyrosine kinase inhibitors in practice: Study by the Nagasaki CML Study Group

    An appropriate trigger for BCR-ABL1 mutation analysis has not yet been established in unselected cohorts of chronic-phase chronic myelogenous leukemia patients. We examined 92 patients after 12 months of tyrosine kinase inhibitor (TKI) treatment in Nagasaki Prefecture, Japan. Univariate analysis revealed that significant factors associated with not attaining a major molecular response (MMR) were the presence of the minor BCR-ABL1 fusion gene, a low daily dose of TKI, and the emergence of BCR-ABL1 kinase domain mutations conferring resistance to imatinib. Factors associated with the loss of sustained MMR were a low daily dose of TKI and the emergence of alternatively spliced BCR-ABL1 mRNA with a 35-nucleotide insertion. Taken together, our results suggest that the search for BCR-ABL1 mutations should be initiated if patients have not achieved MMR following 12 months of TKI treatment
