    Arylsulfonyl indoline-enzamide exhibits inhibitory effect on nasopharyngeal carcinoma

    Purpose: To investigate the effect of arylsulfonyl indoline-benzamide (ASIB) on the viability of nasopharyngeal carcinoma (NPC) cells, and the underlying mechanism of action. Methods: The viability of C666 and NPC 039 cells was determined using 3-(4, 5-dimethylthiazol-2-yl) 2, 5-diphenyltetrazolium bromide (MTT) assay. Cell migration was analysed by wound healing assay, while protein expression levels of matrix metalloproteinases (MMPs), p50, p65 and NF κB were assayed using western blotting. Results: MTT assay results showed that ASIB treatment led to significant and dose-dependent reductions in the viability of C666 and NPC 039 (p ˂ 0.05). The migration and invasive potential of C666 cells were decreased on incubation with ASIB for 48 h. Western blotting data showed significant decrease in MMP 2/9 expressions in C666 cells on treatment with ASIB (p ˂ 0.05). The levels of p65 and p50 in the nuclear fraction of C666 cells were markedly lower than those in the negative control group. Arylsulfonyl indoline-benzamide (ASIB) treatment for 48 h decreased the level of NF κB expression in C666 cells (p ˂ 0.05). The volume of tumor excised from ASIB-treated NPC mice was lower than that of the untreated group. Conclusion: Arylsulfonyl indoline-benzamide (ASIB) exhibits inhibitory effects on the viability and metastasis potential of NPC cells. Thus, it may be beneficial in the treatment of nasopharyngeal carcinoma but this has to be further investigated

    Strain Rate Effects on Head-on Quenching of Laminar Premixed Methane-air flames

    Head-on quenching is a canonical configuration for flame-wall interaction. In the present study, the transient process of a laminar premixed flame impinging on a wall is investigated for different strain rates, while previous studies with detailed chemistry and transport focused only on unstrained conditions. Increasing strain rate leads to a reduction in the normalized quenching distance, and an increase in the normalized wall heat flux, both are considered as global flame quantities. Looking more into the local microstructure of the quenching process, CO formation and oxidation near the wall are shifted to higher temperatures under higher strain rates. Further, the local flame structure and the thermochemical state are affected by differential diffusion driven by differences in species’ gradients and diffusivities. Quenching leads to increased species’ gradients and consequently differential diffusion is amplified near the wall compared to propagating flames. However, this effect is suppressed for increasing strain rates, which is explained in more detail by a source term analysis of the transport equation for the differential diffusion parameter ZHC. Results for the global quantities and the local flame structure show that the impact of the strain rate weakens for higher wall temperatures. Finally, the analyses of the thermo-chemical quantities in the composition space shows that H2 can be a good parameter to characterize the strain rate both for propagating and quenching flamelet

    ACI-BENCH: a Novel Ambient Clinical Intelligence Dataset for Benchmarking Automatic Visit Note Generation

    Recent immense breakthroughs in generative models such as in GPT4 have precipitated re-imagined ubiquitous usage of these models in all applications. One area that can benefit by improvements in artificial intelligence (AI) is healthcare. The note generation task from doctor-patient encounters, and its associated electronic medical record documentation, is one of the most arduous time-consuming tasks for physicians. It is also a natural prime potential beneficiary to advances in generative models. However with such advances, benchmarking is more critical than ever. Whether studying model weaknesses or developing new evaluation metrics, shared open datasets are an imperative part of understanding the current state-of-the-art. Unfortunately as clinic encounter conversations are not routinely recorded and are difficult to ethically share due to patient confidentiality, there are no sufficiently large clinic dialogue-note datasets to benchmark this task. Here we present the Ambient Clinical Intelligence Benchmark (ACI-BENCH) corpus, the largest dataset to date tackling the problem of AI-assisted note generation from visit dialogue.     Altered Gut Microbiota Composition in Subjects Infected With Clonorchis sinensis

    Clonorchiasis is an infectious disease caused by helminths of Clonorchis sinensis (C. sinensis). The adult parasite mainly inhabits the bile duct and gall bladder, and results in various complications to the hepatobiliary system. The amount of bile secreted into the intestine is reduced in cases of C. sinensis infection, which may alter the pH of the gut and decrease the amount of surfactant protein D released from the gallbladder. However, the impact of parasitic infection on the human gut microbiome remains unclear. To this end, we examined the gut microbiota composition in 47 modified Kato–Katz thick smear-positive (egg-positive) volunteers and 42 healthy controls from five rural communities. Subjects were grouped into four sub-populations based on age and infection status. High-throughput 16S rRNA gene sequencing revealed significant changes in alpha diversity between EP1 and EN1. The beta diversity showed alterations between C. sinensis-infected subjects and healthy controls. In C. sinensis infected patients, we found the significant reduction of certain taxa, such as Bacteroides and anti-inflammatory Bifidobacterium (P < 0.05). Bacteroides, a predominant gut bacteria in healthy populations, was negatively correlated with the number of C. sinensis eggs per gram (EPG, r = −0.37, P adjust < 0.01 in 20–60 years old group; r = −0.64, P adjust = 0.04 in the 60+ years old group). What’s more, the reduction in the abundance of Bifidobacterium, a common probiotic, was decreased particularly in the 60 + years old group (r = −0.50, P = 0.04). The abundance of Dorea, a potentially pro-inflammatory microbe, was higher in infected subjects than in healthy individuals (P < 0.05). Variovorax was a unique bacteria that was only detected in infected subjects. These results clearly demonstrate the significant influence of C. sinensis infection on the human gut microbiota and provided new insights into the control, prevention, diagnosis, and clinical study of clonorchiasis through the human gut microbiota

    Development and validation of a nomogram model for predicting unfavorable functional outcomes in ischemic stroke patients after acute phase

    IntroductionPrediction of post-stroke functional outcome is important for personalized rehabilitation treatment, we aimed to develop an effective nomogram for predicting long-term unfavorable functional outcomes in ischemic stroke patients after acute phase.MethodsWe retrospectively analyzed clinical data, rehabilitation data, and longitudinal follow-up data from ischemic stroke patients who underwent early rehabilitation at multiple centers in China. An unfavorable functional outcome was defined as a modified Rankin Scale (mRS) score of 3–6 at 90 days after onset. Patients were randomly allocated to either a training or test cohort in a ratio of 4:1. Univariate and multivariate logistic regression analyses were used to identify the predictors for the development of a predictive nomogram. The area under the receiver operating characteristic curve (AUC) was used to evaluate predictive ability in both the training and test cohorts.ResultsA total of 856 patients (training cohort: n = 684; test cohort: n = 172) were included in this study. Among them, 518 patients experienced unfavorable outcomes 90 days after ischemic stroke. Trial of ORG 10172 in Acute Stroke Treatment classification (p = 0.024), antihypertensive agents use [odds ratio (OR) = 1.86; p = 0.041], 15-day Barthel Index score (OR = 0.930; p < 0.001) and 15-day mRS score (OR = 13.494; p < 0.001) were selected as predictors for the unfavorable outcome nomogram. The nomogram model showed good predictive performance in both the training (AUC = 0.950) and test cohorts (AUC = 0.942).ConclusionThe constructed nomogram model could be a practical tool for predicting unfavorable functional outcomes in ischemic stroke patients underwent early rehabilitation after acute phase

    Seizing the window of opportunity to mitigate the impact of climate change on the health of Chinese residents

    The health threats posed by climate change in China are increasing rapidly. Each province faces different health risks. Without a timely and adequate response, climate change will impact lives and livelihoods at an accelerated rate and even prevent the achievement of the Healthy and Beautiful China initiatives. The 2021 China Report of the Lancet Countdown on Health and Climate Change is the first annual update of China’s Report of the Lancet Countdown. It comprehensively assesses the impact of climate change on the health of Chinese households and the measures China has taken. Invited by the Lancet committee, Tsinghua University led the writing of the report and cooperated with 25 relevant institutions in and outside of China. The report includes 25 indicators within five major areas (climate change impacts, exposures, and vulnerability; adaptation, planning, and resilience for health; mitigation actions and health co-benefits; economics and finance; and public and political engagement) and a policy brief. This 2021 China policy brief contains the most urgent and relevant indicators focusing on provincial data: The increasing health risks of climate change in China; mixed progress in responding to climate change. In 2020, the heatwave exposures per person in China increased by 4.51 d compared with the 1986–2005 average, resulting in an estimated 92% increase in heatwave-related deaths. The resulting economic cost of the estimated 14500 heatwave-related deaths in 2020 is US$176 million. Increased temperatures also caused a potential 31.5 billion h in lost work time in 2020, which is equivalent to 1.3% of the work hours of the total national workforce, with resulting economic losses estimated at 1.4% of China’s annual gross domestic product. For adaptation efforts, there has been steady progress in local adaptation planning and assessment in 2020, urban green space growth in 2020, and health emergency management in 2019. 12 of 30 provinces reported that they have completed, or were developing, provincial health adaptation plans. Urban green space, which is an important heat adaptation measure, has increased in 18 of 31 provinces in the past decade, and the capacity of China’s health emergency management increased in almost all provinces from 2018 to 2019. As a result of China’s persistent efforts to clean its energy structure and control air pollution, the premature deaths due to exposure to ambient particulate matter of 2.5 μm or less (PM2.5) and the resulting costs continue to decline. However, 98% of China’s cities still have annual average PM2.5 concentrations that are more than the WHO guideline standard of 10 μg/m3. It provides policymakers and the public with up-to-date information on China’s response to climate change and improvements in health outcomes and makes the following policy recommendations. (1) Promote systematic thinking in the related departments and strengthen multi-departmental cooperation. Sectors related to climate and development in China should incorporate health perspectives into their policymaking and actions, demonstrating WHO’s and President Xi Jinping’s so-called health-in-all-policies principle. (2) Include clear goals and timelines for climate-related health impact assessments and health adaptation plans at both the national and the regional levels in the National Climate Change Adaptation Strategy for 2035. (3) Strengthen China’s climate mitigation actions and ensure that health is included in China’s pathway to carbon neutrality. By promoting investments in zero-carbon technologies and reducing fossil fuel subsidies, the current rebounding trend in carbon emissions will be reversed and lead to a healthy, low-carbon future. (4) Increase awareness of the linkages between climate change and health at all levels. Health professionals, the academic community, and traditional and new media should raise the awareness of the public and policymakers on the important linkages between climate change and health.</p

    Exploring the Adequacy of Massive Constructed Public Housing in China

    Public housing programs are an effective strategy to provide adequate housing, not only in developed countries, but also developing countries. This study holistically investigates the sitting occupants&#8217; perception of adequate housing towards their public housing units using Chongqing, a typical city in western China, as a case study. Results showed that generally, the public rental housing (PRH) programs were perceived to be adequate by their residents in the estates sampled. The components of neighborhood environment, housing unit, and affordability were the top three factors affecting the overall housing adequacy. The importance of physical aspects as well as the nonphysical aspects of adequate housing is likely to change according to their residential purpose. Therefore, the residential purpose of residents should be taken into consideration when planning the physical and nonphysical elements of public housing programs. Meanwhile, socioeconomic characteristics of age, family income, family members, residence length, and housing types have significant effects on overall housing adequacy and its components. These findings shed some useful insights on the sustainable development of public housing in China and provide a useful reference for future public housing developments in developing countries. The provision of adequate housing also helps to attract and retain talent, which consequently improves the competitiveness of the city

    Prunus mume polyphenols maintains the quality and nutrient values of winter jujube (Zizyphus jujuba Mill.) fruits during storage

    This study evaluated the effect of Prunus mume polyphenols (PMPs) treatment on qualities and nutrient in winter jujube fruits during storage (4°C). Winter jujube fruits were treated with 5, 10, and 15 mg L−1 PMP, respectively. Compared with the control, PMP treatments effectively preserved the quality and nutrient values of winter jujube fruits. They also enhanced the activities of antioxidant enzymes, inhibited the growth of microorganisms, and delayed postharvest metabolism in the fruits. These findings suggest that PMP, as a green, safe agent, can be potentially developed as a postharvest fruit preservative to substantially extend the shelf life
