    Lesions of the nucleus accumbens core modulate development of matching behavior

    BACKGROUND: The development of choice is a crucial determinant in the performance of appetitive responses. Given two options with different reinforcement rates, animals match their relative rate of responding to the relative rates of reinforcement (i.e., matching behavior). A previous study has shown that the nucleus accumbens core (AcbC) is involved in the performance of matching behavior in trained animals. However, the role of the AcbC in the acquisition of matching behavior has not been addressed. RESULTS: We conducted a series of experimental sessions to examine the role of the AcbC on the development of matching behavior. Instrumental responding was measured in rats with excitotoxic lesions of the AcbC. Rats were given two options that differed in the relative rate of reinforcement under concurrent variable-interval schedules. The locations of the more frequently reinforced option and the alternative option were randomly switched between sessions. Lesions of the AcbC accelerated the development of matching behavior compared to the sham-operated group. The AcbC-lesioned rats exhibited closer conformity to the matching law than shams when the options were in the same positions as in the previous session (the same condition), but not when the option locations had been switched (the different condition). The AcbC rats showed smaller probabilities of switching behavior between alternatives than shams. Post-reinforcement pausing was not affected by the AcbC lesion. Neither numbers of rewards obtained nor number of lever presses were different between the AcbC-lesioned rats and shams over session blocks. CONCLUSIONS: Our results suggest that the AcbC plays a regulatory role in the development of matching behavior through switching probabilities rather than perception of reward magnitude. The differential effect of AcbC lesions on the matching behavior between the same and different conditions suggests influence of the spontaneous recovery, that is, reversion to a previously reinforced choice at the beginning of the next session, on the development of matching behavior in the AcbC-lesioned rats

    Resveratrol promotes expression of SIRT1 and StAR in rat ovarian granulosa cells: an implicative role of SIRT1 in the ovary

    <p>Abstract</p> <p>Background</p> <p>Resveratrol is a natural polyphenolic compound known for its beneficial effects on energy homeostasis, and it also has multiple properties, including anti-oxidant, anti-inflammatory, and anti-tumor activities. Recently, silent information regulator genes (Sirtuins) have been identified as targets of resveratrol. Sirtuin 1 (SIRT1), originally found as an NAD<sup>+</sup>-dependent histone deacetylase, is a principal modulator of pathways downstream of calorie restriction, and the activation of SIRT1 ameliorates glucose homeostasis and insulin sensitivity. To date, the presence and physiological role of SIRT1 in the ovary are not known. Here we found that SIRT1 was localized in granulosa cells of the human ovary.</p> <p>Methods</p> <p>The physiological roles of resveratrol and SIRT1 in the ovary were analyzed. Immunohistochemistry was performed to localize the SIRT1 expression. SIRT1 protein expression of cultured cells and luteinized human granulosa cells was investigated by Western blot. Rat granulosa cells were obtained from diethylstilbestrol treated rats. The cells were treated with increasing doses of resveratrol, and subsequently harvested to determine mRNA levels and protein levels. Cell viability was tested by MTS assay. Cellular apoptosis was analyzed by caspase 3/7 activity test and Hoechst 33342 staining.</p> <p>Results</p> <p>SIRT1 protein was expressed in the human ovarian tissues and human luteinized granulosa cells. We demonstrated that resveratrol exhibited a potent concentration-dependent inhibition of rat granulosa cells viability. However, resveratrol-induced inhibition of rat granulosa cells viability is independent of apoptosis signal. Resveratrol increased mRNA levels of SIRT1, LH receptor, StAR, and P450 aromatase, while mRNA levels of FSH receptor remained unchanged. Western blot analysis was consistent with the results of quantitative real-time RT-PCR assay. In addition, progesterone secretion was induced by the treatment of resveratrol.</p> <p>Conclusions</p> <p>These results suggest a novel mechanism that resveratrol could enhance progesterone secretion and expression of luteinization-related genes in the ovary, and thus provide important implications to understand the mechanism of luteal phase deficiency.</p

    Gender-specific associations of vision and hearing impairments with adverse health outcomes in older Japanese: a population-based cohort study

    BACKGROUND: Several epidemiological studies have shown that self-reported vision and hearing impairments are associated with adverse health outcomes (AHOs) in older populations; however, few studies have used objective sensory measurements or investigated the role of gender in this association. Therefore, we examined the association of vision and hearing impairments (as measured by objective methods) with AHOs (dependence in activities of daily living or death), and whether this association differed by gender. METHODS: From 2005 to 2006, a total of 801 residents (337 men and 464 women) aged 65 years or older of Kurabuchi Town, Gunma, Japan, participated in a baseline examination that included vision and hearing assessments; they were followed up through September 2008. Vision impairment was defined as a corrected visual acuity of worse than 0.5 (logMAR = 0.3) in the better eye, and hearing impairment was defined as a failure to hear a 30 dB hearing level signal at 1 kHz in the better ear. Information on outcomes was obtained from the town hall and through face-to-face home visit interviews. We calculated the risk ratios (RRs) of AHOs for vision and hearing impairments according to gender. RESULTS: During a mean follow-up period of 3 years, 34 men (10.1%) and 52 women (11.3%) had AHOs. In both genders, vision impairment was related to an elevated risk of AHOs (multi-adjusted RR for men and women together = 1.60, 95% CI = 1.05-2.44), with no statistically significant interaction between the genders. In contrast, a significant association between hearing impairment and AHOs (multi-adjusted RR = 3.10, 95% CI = 1.43-6.72) was found only in the men. CONCLUSION: In this older Japanese population, sensory impairments were clearly associated with AHOs, and the association appeared to vary according to gender. Gender-specific associations between sensory impairments and AHOs warrant further investigation

    Global Norms, Local Activism, and Social Movement Outcomes: Global Human Rights and Resident Koreans in Japan

    The authors integrate social movement outcomes research and the world society approach to build a theoretical model to examine the impact of global and local factors on movement outcomes. Challenging the current research on policy change, which rarely examines the effects of global norms and local activism in one analysis, they argue (1) that global regimes empower and embolden local social movements and increase pressure on target governments from below, and (2) that local activists appeal to international forums with help from international activists to pressure the governments from above. When the pressures from the top and the bottom converge, social movements are more likely to succeed. Furthermore, these pressures are stronger in countries integrated into global society and on issues with strong global norms. The empirical analysis of social movements by resident Koreans in Japan advocating for four types of human rights—civil, political, social/economic, and cultural—demonstrates that the movements produced more successes as Japan\u27s involvement in the international human rights regime expanded since the late 1970s, and that activism on issues with strong global norms achieved greater successes. The analysis also shows that lack of cohesive domestic activism can undercut the chances of social movements\u27 success even with strong global norms on the issue

    Constrained paths based on Farey sequence in learning to juggle

    In this paper, we report on the results of a study conducted to describe the learning dynamics of three-ball juggling from the perspective of frequency locking. The theoretical prediction about coordination patterns that could appear in learning process of three-ball juggling from the principle of frequency locking based on Farey sequence showed the existence of several stable coordination patterns denoted by dwell ratios of 0.83, 0.75, 0.67, and 0.50. We observed the changes in the coordination patterns during actual learning processes based on task performance, and compared them with the predictive coordination patterns. Consequently, we discovered that individuals acquired their own coordination patterns in the early stage of learning, and those coordination patterns did not change in subsequent learning processes. The observed coordination patterns corresponded with theoretical prediction, and imply that these observed patterns have a stable coordination structure that shows strong frequency locking among the temporal variables that comprise juggling. This implies that the learning dynamics of three-ball juggling can be described as a process in which a learner acquires one of several stable coordination patterns based on the principle of frequency locking during the exploratory process in the early stage of learning. In other words, there may be several paths through the learning process that utilize different attractors
