    Comparison of net ecosystem carbon exchange estimation in a mixed temperate forest using field eddy covariance and MODIS data

    Quantification of net ecosystem carbon exchange (NEE) between the atmosphere and vegetation is of great importance for regional and global studies of carbon balance. The eddy covariance technique can quantify carbon budgets and the effects of environmental controls for many forest types across the continent but it only provides integrated CO2 flux measurements within tower footprints and need to be scaled up to large areas in combination with remote sensing observations. In this study we compare a multiple-linear regression (MR) model which relates enhanced vegetation index and land surface temperature derived from the moderate resolution imaging spectroradiometer (MODIS), and photosynthetically active radiation with the site-level NEE, for estimating carbon flux exchange between the ecosystem and the environment at the deciduous-dominated Harvard Forest to three other methods proposed in the literature. Six years (2001–2006) of eddy covariance and MODIS data are used and results show that the MR model has the best performance for both training (2001–2004, R2 = 0.84, RMSE = 1.33 g Cm−2 day−1) and validation (2005–2006, R2 = 0.76, RMSE = 1.54 g Cm−2 day−1) datasets comparing to the other ones. It provides the potential to estimate carbon flux exchange across different ecosystems at various time intervals for scaling up plot-level NEE of CO2 to large spatial areas

    Biological roles of RNA m5C modification and its implications in Cancer immunotherapy

    Epigenetics including DNA and RNA modifications have always been the hotspot field of life sciences in the post-genome era. Since the first mapping of N6-methyladenosine (m6A) and the discovery of its widespread presence in mRNA, there are at least 160-170 RNA modifications have been discovered. These methylations occur in different RNA types, and their distribution is species-specific. 5-methylcytosine (m5C) has been found in mRNA, rRNA and tRNA of representative organisms from all kinds of species. As reversible epigenetic modifications, m5C modifications of RNA affect the fate of the modified RNA molecules and play important roles in various biological processes including RNA stability control, protein synthesis, and transcriptional regulation. Furthermore, accumulative evidence also implicates the role of RNA m5C in tumorigenesis. Here, we review the latest progresses in the biological roles of m5C modifications and how it is regulated by corresponding “writers”, “readers” and “erasers” proteins, as well as the potential molecular mechanism in tumorigenesis and cancer immunotherapy

    Integrated metabolomics and metagenomics analysis of plasma and urine identified microbial metabolites associated with coronary heart disease

    Coronary heart disease (CHD) is top risk factor for health in modern society, causing high mortality rate each year. However, there is no reliable way for early diagnosis and prevention of CHD so far. So study the mechanism of CHD and development of novel biomarkers is urgently needed. In this study, metabolomics and metagenomics technology are applied to discover new biomarkers from plasma and urine of 59 CHD patients and 43 healthy controls and trace their origin. We identify GlcNAc-6-P which has good diagnostic capability and can be used as potential biomarkers for CHD, together with mannitol and 15 plasma cholines. These identified metabolites show significant correlations with clinical biochemical indexes. Meanwhile, GlcNAc-6-P and mannitol are potential metabolites originated from intestinal microbiota. Association analysis on species and function levels between intestinal microbes and metabolites suggest a close correlation between Clostridium sp. HGF2 and GlcNAc-6-P, Clostridium sp. HGF2, Streptococcus sp. M143, Streptococcus sp. M334 and mannitol. These suggest the metabolic abnormality is significant and gut microbiota dysbiosis happens in CHD patients

    Identification of Drugs that Interact with Herbs in Drug Development

    To date, several clinically important drugs have been identified that interact with commonly used herbs. These drugs include (among others) warfarin, midazolam, digoxin, amitriptyline, indinavir, cyclosporine, tacrolimus and irinotecan. Importantly, many of these drugs have very narrow therapeutic indices. Most of them are substrates for cytochrome P450s (CYPs) and/or P-glycoprotein (P-gp). Because drug–herb interactions can significantly affect circulating levels of drug and, hence, alter the clinical outcome, the identification of drugs that interact with commonly used herbal medicines has important implications in drug development. In silico, in vitro, animal and human studies are often used to identify drug interactions with herbs. We propose that drug–herb and herb–CYP interaction studies should be incorporated into drug development. Because of the clinical significance of drug interactions with herbs, there is a strong necessity to identify drugs that may interact with herbal medicines in drug development

    Spatiotemporal variation and freeze-thaw asymmetry of Arctic sea ice in multiple dimensions during 1979 to 2020

    Arctic sea ice is broadly regarded as an indicator and amplifier of global climate change. The rapid changes in Arctic sea ice have been widely concerned. However, the spatiotemporal changes in the horizontal and vertical dimensions of Arctic sea ice and its asymmetry during the melt and freeze seasons are rarely quantified simultaneously based on multiple sources of the same long time series. In this study, the spatiotemporal variation and freeze-thaw asymmetry of Arctic sea ice were investigated from both the horizontal and vertical dimensions during 1979-2020 based on remote sensing and assimilation data. The results indicated that Arctic sea ice was declining at a remarkably high rate of -5.4 x 104 km2/a in sea ice area (SIA) and -2.2 cm/a in sea ice thickness (SIT) during 1979 to 2020, and the reduction of SIA and SIT was the largest in summer and the smallest in winter. Spatially, compared with other sub-regions, SIA showed a sharper declining trend in the Barents Sea, Kara Sea, and East Siberian Sea, while SIT presented a larger downward trend in the northern Canadian Archipelago, northern Greenland, and the East Siberian Sea. Regarding to the seasonal trend of sea ice on sub-region scale, the reduction rate of SIA exhibited an apparent spatial heterogeneity among seasons, especially in summer and winter, i.e., the sub-regions linked to the open ocean exhibited a higher decline rate in winter; however, the other sub-regions blocked by the coastlines presented a greater decline rate in summer. For SIT, the sub-regions such as the Beaufort Sea, East Siberian Sea, Chukchi Sea, Central Arctic, and Canadian Archipelago always showed a higher downward rate in all seasons. Furthermore, a striking freeze-thaw asymmetry of Arctic sea ice was also detected. Comparing sea ice changes in different dimensions, sea ice over most regions in the Arctic showed an early retreat and rapid advance in the horizontal dimension but late melting and gradual freezing in the vertical dimension. The amount of sea ice melting and freezing was disequilibrium in the Arctic during the considered period, and the rate of sea ice melting was 0.3 x 104 km2/a and 0.01 cm/a higher than that of freezing in the horizontal and vertical dimensions, respectively.     Assessment of coastal zone sustainable development: A case study of Yantai, China

    The assessment of sustainable development is a challenging task as its measuring is rather complex without a mature framework. In this paper, as a case study, a coastal city of China-Yantai was assessed for sustainable development in the period from 1998 to 2007. We used a methodological framework based on 36 indicators and three composite indices from the dimensions of environment, economy and society subsystems. The assessment results indicated that Yantai was almost in the potentially unsustainable development or intermediate sustainable development, except in 1998 and in 2007. Accordingly, the progress of sustainable development was divided into two stages in the light of the relative changes of three subsystems. Some relevant issues, such as natural capital, GPI vs. GDP in sustainable development assessment were discussed. Finally, an uncertainty analysis was also given in the assessment. In conclusion, the sustainable development in Yantai had experienced a shift from environment-based to social-economic-based in the past 10 years. (C) 2010 Elsevier Ltd.     How important are the wetlands in the middle-lower Yangtze River region: An ecosystem service valuation approach

    The middle-lower Yangtze River region, with its lake groups and river systems, is one of the important wetland regions in the world. In this study, physical dimension measurement and monetary evaluation were conducted to estimate the value of wetland services in this region. Results revealed that the total value of the wetland ecosystem services in the middle-lower Yangtze River region is US 162.5billionperyear,whichreflectstheirreplaceableimportanceofwetlandsinthisregion.Thewetlandecosystemservicevaluesrevealedconsiderablespatialvariability.PoyangLake,HongLake,andShijiuLakehavethemostecosystemservicevalue.Thevalueofhuman−maclewetlands(2.62x10(4)US162.5 billion per year, which reflects the irreplaceable importance of wetlands in this region. The wetland ecosystem service values revealed considerable spatial variability. Poyang Lake, Hong Lake, and Shijiu Lake have the most ecosystem service value. The value of human-macle wetlands (2.62 x 10(4) US /ha/yr) is 48% lower than that of natural wetlands (5.04 x 10(4) US $/hafyr), indicating that the conversion of natural wetlands for aquaculture from the sustainability perspective is not significant. The direct and indirect values of wetland ecosystems are 46.17% and 53.83% of the total values, respectively. Recognizing that the value of wetlands in this region is mainly embodied as indirect use values, conservation of the regulating and supporting services should be prioritized in the design of future wetland ecosystem management plans.     Annual normalized difference vegetation index (NDVI) and chlorophyll-a (Chl-a) concentration are the most important large-scale indicators of terrestrial and oceanic ecosystem net primary productivity. In this paper, the Sea-viewing Wide Field-of-view Sensor level 3 standard mapped image annual products from 1998 to 2009 are used to study the spatial-temporal characters of terrestrial NDVI and oceanic Chl-a concentration on two sides of the coastline of China by using the methods of mean value (M), coefficient of variation (CV), the slope of unary linear regression model (Slope), and the Hurst index (H). In detail, we researched and analyzed the spatial-temporal dynamics, the longitudinal zonality and latitudinal zonality, the direction, intensity, and persistency of historical changes. The results showed that: (1) spatial patterns of M and CV between NDVI and Chl-a concentration from 1998 to 2009 were very different. The dynamic variation of terrestrial NDVI was much mild, while the variation of oceanic Chl-a concentration was relatively much larger; (2) distinct longitudinal zonality was found for Chl-a concentration and NDVI due to their hypersensitivity to the distance to shoreline, and strong latitudinal zonality existed for Chl-a concentration while terrestrial NDVI had a very weak latitudinal zonality; (3) overall, the NDVI showed a slight decreasing trend while the Chl-a concentration showed a significant increasing trend in the past 12 years, and both of them exhibit strong self-similarity and long-range dependence which indicates opposite future trends between land and ocean.Annual normalized difference vegetation index (NDVI) and chlorophyll-a (Chl-a) concentration are the most important large-scale indicators of terrestrial and oceanic ecosystem net primary productivity.     Influencing factors of care dependence in patients with coronary heart disease after percutaneous coronary intervention—A cross‐sectional study

    Abstract Aim Care dependence has been scarcely investigated in coronary heart disease patients after percutaneous coronary intervention. This study aimed to investigate the association between frailty, self‐efficacy, combined effects of frailty and self‐efficacy, mental health, and care dependence in coronary heart disease patients after percutaneous coronary intervention. Design Cross‐sectional study. Methods Data from 400 patients after percutaneous coronary intervention were collected from 2017–2020. Logistic regression model and mediating analysis were used to identify the association between frailty, self‐efficacy, combined effects of frailty and self‐efficacy, and care dependence. Results Patients with frailty and self‐efficacy tended to have severe care dependence symptoms. There was no correlation between frailty symptoms, self‐efficacy, and care dependence in patients without symptoms of anxiety or depression. But in patients with anxiety or depression symptoms, there is a strong correlation between frailty symptoms, lower self‐efficacy, and care dependence. Mental health played an inhibitory effect on frailty and care dependence

    Artemisinin Confers Cytoprotection toward Hydrogen Peroxide-Induced Cell Apoptosis in Retinal Pigment Epithelial Cells in Correlation with the Increased Acetylation of Histone H4 at Lysine 8

    Increased oxidative stress is one of the critical pathologies inducing age-related macular degeneration (AMD), characterized by retinal pigment epithelial (RPE) cell damage and death. The unbalanced acetylation and deacetylation of histones have been implicated in AMD pathogenesis or hydrogen peroxide (H2O2)-induced cell damage. Therefore, strategies aimed at controlling the balance between acetylation and deacetylation may effectively protect RPE cells from oxidative damage. Artemisinin is an antimalarial lactone drug derived from Artemisia annua, with antioxidant activity known to modulate histone acetylation in the brain, but its effect on the retina is unknown. In this study, we aimed to investigate whether Artemisinin exerts a cytoprotective effect on oxidative stress-induced apoptosis in RPE cells by regulating histone acetylation. We hypothesized that Artemisinin confers cytoprotection toward H2O2-induced apoptosis in RPE cells through this mechanism. In the present study, we found that Artemisinin at a sub-clinic dosage of 20 ÎŒM inhibited the H2O2-induced cell viability decrease and B-cell lymphoma 2 (Bcl-2) protein level decrease and attenuated the H2O2-induced decrease in the histone H4 lysine (Lys) 8 acetylation [Acetyl-H4 (Lys 8)] level in the retinal RPE cell line D407. As expected, histone deacetylase inhibitor Trichostatin A at the concentration of 250 nM increased the Acetyl-H4 (Lys 8) level in D407 cells and attenuated the H2O2-induced cell viability decrease and apoptosis. Similar findings were obtained using adult RPE (ARPE)19 cells, another human RPE cell line, and primary human RPE cell cultures. In conclusion, these results confirmed our hypothesis and indicated that Artemisinin attenuated H2O2-induced apoptosis in apparent correlation with the increase in the Acetyl-H4 (Lys 8) level, which is associated with gene transcription and cell survival. By modulating histone acetylation, Artemisinin may restore the balance between acetylation and deacetylation and enhance the resistance and survival of RPE cells under oxidative stress. Our study provides novel mechanistic insights into the effect of Artemisinin on histone acetylation and apoptosis in RPE cells and supports the potential application of Artemisinin in the prevention and/or treatment of AMD
