    RelatiOnship between the impact of invasive largemouth bass and environmental conditions in ponds

The stlldy was conducted at 49 Ponds in Oshu city in lwate prefecture, northeastern Japan・ For determiningthe impact of largemouth bass on fish fauna in ponds, Pearson's correlation wasused between the dominance of largemouth bass (%BASS), total number (S) and diversity index (H') of occurring fish species. Furthermore, multiple regression analysis was used to detemine the relationship between S or HT and environmental factors・ %BASS was negatively correlated with both S and H'. In ponds without bass, both S and H'were`related to maximum depth and S in ponds with bass was also related to maximum depth.     Loss of fibulin-4 disrupts collagen synthesis and maturation: implications for pathology resulting from EFEMP2 mutations

    Homozygous recessive mutations in either EFEMP2 (encoding fibulin-4) or FBLN5 (encoding fibulin-5), critical genes for elastogenesis, lead to autosomal recessive cutis laxa types 1B and 1A, respectively. Previously, fibulin-4 was shown to bind lysyl oxidase (LOX), an elastin/collagen cross-linking enzyme, in vitro. Consistently, reported defects in humans with EFEMP2 mutations are more severe and broad in range than those due to FBLN5 mutations and encompass both elastin-rich and collagen-rich tissues. However, the underlying disease mechanism in EFEMP2 mutations has not been fully addressed. Here, we show that fibulin-4 is important for the integrity of aortic collagen in addition to elastin. Smooth muscle-specific Efemp2 loss in mouse (termed SMKO) resulted in altered fibrillar collagen localization with larger, poorly organized fibrils. LOX activity was decreased in Efemp2-null cells, and collagen cross-linking was diminished in SMKO aortas; however, elastin cross-linking was unaffected and the level of mature LOX was maintained to that of wild-type aortas. Proteomic screening identified multiple proteins involved in procollagen processing and maturation as potential fibulin-4-binding partners. We showed that fibulin-4 binds procollagen C-endopeptidase enhancer 1 (Pcolce), which enhances proteolytic cleavage of the procollagen C-terminal propeptide during procollagen processing. Interestingly, however, procollagen cleavage was not affected by the presence or absence of fibulin-4 in vitro. Thus, our data indicate that fibulin-4 serves as a potential scaffolding protein during collagen maturation in the extracellular space. Analysis of collagen in other tissues affected by fibulin-4 loss should further increase our understanding of underlying pathologic mechanisms in patients with EFEMP2 mutations

    Efficacy of a Novel Narrow Knife with Water Jet Function for Colorectal Endoscopic Submucosal Dissection

    Backgrounds. With respect to the knife’s design in colorectal endoscopic submucosal dissection (ESD), diameter, water jet function, and electric power are important because these relate to efficient dissection. In this study, we analyzed a novel, narrow ball tip-typed ESD knife with water jet function (Flush knife BT-S, diameter: 2.2 mm, length: 2000 mm, Fujifilm Co., Tokyo, Japan) compared to a regular diameter knife (Flush knife BT, diameter: 2.6 mm, length: 1800 mm). Methods. In laboratory and clinical research, electric power, knife insertion time, vacuum/suction amount with knife in the endoscopic channel, and water jet function were analyzed. We used a knife 2.0 mm long for BT-S and BT knives. Results. The BT-S showed faster mean knife insertion time (sec) and better vacuum amount (ml/min) compared to the BT (insertion time: 16.7 versus 21.6, p<0.001, vacuum amount: 38.0 versus 14.0, p<0.01). Additionally, the water jet function of the BT-S was not inferior. In 39 colorectal ESD cases in two institutions, there were mean 4.7 times (range: 1–28) of knife insertion. Suction under knife happened 59% (23/39) and suction of fluid could be done in 100%. Conclusions. Our study showed that the narrow knife allows significantly faster knife insertion, better vacuum function, and effective clinical results

    Magnifying Endoscopy with Blue Laser Imaging Improves the Microstructure Visualization in Early Gastric Cancer: Comparison of Magnifying Endoscopy with Narrow-Band Imaging

    Backgrounds. Magnifying endoscopy with blue laser imaging (ME-BLI) for diagnosis of early gastric cancer (EGC) is as effective as magnifying endoscopy with narrow-band imaging (ME-NBI). However, there are different EGCs in microstructure visualization between ME-BLI and ME-NBI. This study aimed to clarify the pathological features of the EGCs, in which microstructure visualization was different between ME-NBI and ME-BLI. Methods. EGCs were classified into groups A (irregular microsurface pattern (MSP) in ME-BLI and absent MSP in ME-NBI), B (irregular MSP in two modalities), or C (absent MSP in two modalities), according to the vessel plus surface classification. We compared the pathological features of EGCs between the three groups. Results. 17, four, and five lesions could be evaluated in detail in groups A, B and C, respectively. Well-differentiated adenocarcinomas with shallow crypts were more frequent in group A than in group B (58.8 and 0%, resp.). The mean crypt depth of group A was significantly shallower than that of group B (56 ± 20, 265 ± 64 μm, resp., P=0.0002). Conclusions. ME-BLI could better visualize the microstructures of the EGCs with shallow crypts compared with ME-NBI. Therefore, ME-BLI could enable a more accurate diagnosis of EGC with shallow crypts

    Clinical background factors as predictors of the efficacy of 5-aminosalicylic acid suppositories in patients with ulcerative colitis

    Introduction: Although the efficacy of 5-aminosalicylic acid (ASA) suppositories for ulcerative colitis (UC) has been reported in many studies, many studies have also described poor adherence to 5-ASA suppository regimens. We aimed to identify the clinical background factors that influence adherence to 5-ASA suppositories to improve adherence and efficacy of the treatment. Methods: We conducted a retrospective cohort study of 61 patients with active UC who were using 5-ASA suppositories. All patients underwent endoscopy and rectal biopsy for histological diagnosis prior to 5-ASA suppository treatment. The efficacy of 5-ASA suppository treatment was compared in relation to clinical background factors (sex, age, disease duration, disease type, clinical activity, Ulceratve colitis Endoscopic Index of Severity, histological activity, serum C-reactive protein level, concomitant use of immunomodulators, history of steroid use, and dose of oral 5-ASA). Results: The efficacy of 5-ASA suppositories was significantly related to low Lichtiger Colitis Activity Index (LCAI) scores and proctitis type prior to its use. In terms of sex, females tended to show higher efficacy. Multivariate logistic regression analysis using these three factors showed high predictive value for the efficacy of 5-ASA suppositories (AUC, 0.788; sensitivity, 87.2%; and specificity, 63.7%). Discussion/Conclusion: This study is the first to extract clinical background factors for predicting the efficacy of 5-ASA suppositories. The use of 5-ASA suppositories in patients who are expected to show efficacy will be effective in improving patient co-operation

    Determining the relative importance of catchment- and site-scale factors in structuring fish assemblages in small coastal streams

    Coastal streams provide important habitat for many diadromous fish species, which migrate between freshwater and the sea during their life cycle. However, coastal systems are poorly studied in comparison to large, continental river systems. Furthermore, the relative importance of catchment- and site-scale factors in structuring lotic fish communities is largely unknown. In this study, I addressed these issues by surveying the fish fauna of small coastal streams on Sado Island (northwest Japan) and determining the relative importance of catchment- and site-scale factors to the structure of freshwater fish assemblages. In total, 14 freshwater fish species were collected from the 19 streams. All but one of the fish species collected were diadromous and 9 of the species (64%) were amphidromous, primarily represented by the goby group including the genera Rhinogobius and Gymnogobius. Variance partitioning analyses showed that catchment-scale factors (namely, stream discharge) were better predictors of both fish species richness and composition than site scale and spatial factors. These results indicate that discharge, which is directly linked to habitat stability, can have a major role in structuring coastal fish assemblages, likely because small coastal streams experience extreme discharge fluctuations associated with regional weather conditions

    The presence of piscivorous fish has a significant effect on the structure of fish assemblages. Exotic, piscivorous largemouth bass (Micropterus salmoides) have been introduced to a number of Japanese farm ponds that were originally characterized by a high level of biodiversity. We evaluated the effects of exotic bass on the structure of the fish assemblage following their introduction into the farm ponds. We collected fish and measured environmental conditions in 14 ponds of northeastern Japan. The species richness of pond-dwelling fish assemblages was negatively correlated with the relative abundance of exotic bass. In addition, the relative abundance of lentic cyprinid species (e.g., Rhodeus ocellatus ocellatus, Pseudorasbora parva and Carassius sp.) was negatively correlated with the relative abundance of exotic bass. The abundance of these species was also influenced by environmental factors, including transparency and/or coverage of aquatic vegetation, likely related to their influence on the predatory efficiency of exotic bass. Our results suggest that the structure of fish assemblages in farm ponds with exotic piscivorous fish was influenced by both biotic (i.e., predation) and abiotic factors and that some environmental factors mediate the effects of exotic bass on fish species richness and composition
