Cyclodextrin-responsive nanogel as an artificial chaperone for horseradish peroxidase
The thermal stabilization and refolding of horseradish peroxidase (HRP) upon heating were investigated using an artificial molecular chaperone consisting of cholesterol-bearing pullulan (CHP) nanogels. The CHP nanogels inhibited the aggregation of HRP under heating by complexation with the denatured HRP. The enzyme activity of HRP complexed with CHP nanogels was not detected. However, the enzyme activity recovered up to 80% of native HRP after the addition of cyclodextrin (CD) to the complex. The dissociation of CHP nanogels was induced by the formation of an inclusion complex of cholesterol groups of CHP with CD. The enzyme activity of HRP was only significantly recovered by the addition of β-CD or its derivatives. Natural molecular chaperones, such as GroEL/ES, trap, fold, and release the nonnative proteins by changing the hydrophobicity of the specific sites of the molecular chaperone that interact with the nonnative protein. The functional mechanism of the nanogel chaperon system is similar to that of natural molecular chaperones. The nanogel chaperone system is a useful tool to aid the refolding and thermal stabilization of unstable proteins for post-genome research, and in medical and biological applications

Network of two-Chinese-character compound words in Japanese language
Some statistical properties of a network of two-Chinese-character compound
words in Japanese language are reported. In this network, a node represents a
Chinese character and an edge represents a two-Chinese-character compound word.
It is found that this network has properties of "small-world" and "scale-free."
A network formed by only Chinese characters for common use ({\it joyo-kanji} in
Japanese), which is regarded as a subclass of the original network, also has
small-world property. However, a degree distribution of the network exhibits no
clear power law. In order to reproduce disappearance of the power-law property,
a model for a selecting process of the Chinese characters for common use is
proposed
Effect of acidity on the physicochemical properties of α- and β-chitin nanofibers
Published online 11 Apr 2017We have investigated whether acidity can be used to control the physicochemical properties of chitin nanofibers (ChNFs). In this study, we define acidity as the molar ratio of dissociated protons from the acid to the amino groups in the raw chitin powder. The effect of acidity on the physicochemical properties of α- and β-ChNFs was compared. The transmittance and viscosity of the β-ChNFs drastically and continuously increased with increasing acidity, while those of the α-ChNFs were not affected by acidity. These differences are because of the higher ability for cationization based on the more flexible crystal structure of β-chitin than α-chitin. In addition, the effect of the acid species on the transmittance of β-ChNFs was investigated. The transmittance of β-ChNFs can be expressed by the acidity regardless of the acid species, such as hydrochloric acid, phosphoric acid, and acetic acid. How do differences in native language affect out-of-body experiences?
Out-of-body experiences are scientifically inducible cognitive phenomena attracting global attention due to their application in the Metaverse and medical care. Despite previous studies suggesting that one’s native language influences one’s cognition, the out-of-body experiences of humans with different native languages have not been investigated separately. This study replicated an experiment from a 2007 study to investigate whether differences in native language affect the ability to have scientifically induced out-of-body experiences. A total of 19 age-matched native English and Japanese speakers completed the experiment in two blocks. Thereafter, their experiences were evaluated using questionnaires, and their responses were compared. Importantly, no significant differences between the English and Japanese native-speaker conditions were found. The results showed that out-of-body experiences were induced similarly in both groups, suggesting that people can have out-of-body experiences as a response to similar stimuli, regardless of their native language. However, differences in participants’ introspective reports suggested that their experiences may differ qualitatively, possibly, due to the different linguistic backgrounds. The elucidation of the mechanisms of science-assisted out-of-body experiences that consider different cultural and cognitive characteristics, such as native language, could lead to the investigation of their applications in the borderless Metaverse and medicine
The treatment effect of endovascular therapy for chronic limb‐threatening ischemia with systemic sclerosis
Systemic sclerosis (SSc) is a collagen disease with immune abnormalities, vasculopathy, and fibrosis. Ca blockers and prostaglandins are used to treat peripheral circulatory disturbances. Chronic limb-threatening ischemia (CLTI) is a disease characterized by extremity ulcers, necrosis, and pain due to limb ischemia. Since only a few patients present with coexistence of CLTI and SSc, the treatment outcomes of revascularization in these cases are unknown. In this study, we evaluated the clinical characteristics and treatment outcomes of seven patients with CLTI and SSc, and 35 patients with uncomplicated CLTI who were hospitalized from 2012 to 2022. A higher proportion of patients with uncomplicated CLTI had diabetes and male. There were no significant differences in the age at which ischemic ulceration occurred, other comorbidities, or in treatments, including antimicrobial agents, revascularization and amputation, improvement of pain, and the survival time from ulcer onset between the two subgroups. EVT or amputation was performed in six or two of the seven patients with CLTI and SSc, respectively. Among those who underwent EVT, 33% (2/6) achieved epithelialization and 67% (4/6) experienced pain relief. These results suggest that the revascularization in cases with CLTI and SSc should consider factors such as infection and general condition, since revascularization improve the pain of these patients
A Study on the Incidence and Comorbidities of Autism Spectrum Disorders Accompanied by Intellectual Disabilities in Yonago City, Japan
[Background] Autism spectrum disorders (ASD) with intellectual disabilities may be associated with many factors. This study focused on patients with ASD with intellectual disabilities, defined by a threshold intelligence quotient (IQ) or development quotient (DQ) of 70. We also discuss comorbidities and other factors related to ASD. [Methods] We extracted case records of patients born between April 1995 and March 2001 who lived in Yonago City, as of January 2011, and had visited the two specialist institutions for consultation regarding developmental issues. The list was further narrowed down to patients identified, as having ASD by pediatric neurologists based on Diagnostic and Statistical Manual of Mental Disorders (Fifth Edition) (DSM-5). We selected patients with < IQ/DQ 70 using the most recent intelligence/development test to determine comorbidities and other factors related to ASD. [Results] The data of 81 patients (59 males and 22 females) were extracted, corresponding to an incidence of 76.2 patients out of every 10,000 births. The male-to-female ratio was 2.7:1. Comorbidities and related factors of ASD were observed in 25 cases (30.9%). Eleven cases had perinatal abnormalities. Other abnormalities were observed in 17 cases, including epilepsies in 7, chromosomal abnormalities in 4, familial mental retardation in 1, and acquired brain injury in 1. [Conclusion] It is important to treat and support individuals with ASD and intellectual disabilities taking into account the characteristics and prognosis of the comorbidities and related factors
