    Enhancement of OVA-induced murine lung eosinophilia by co-exposure to contamination levels of LPS in Asian sand dust and heated dust.

    BackgroundA previous study has shown that the aggravation of Asian sand dust (ASD) on ovalbumin (OVA)-induced lung eosinphilia was more severe in lipopolysaccharide (LPS)-rich ASD than in SiO2-rich ASD. Therefore, the effects of different LPS contamination levels in ASD on the aggravation of OVA-induced lung eosinophilia were investigated in the present study.MethodsBefore beginning the in vivo experiment, we investigated whether the ultra-pure LPS would act only on TLR4 or not using bone marrow-derived macrophages (BMDMs) of wild-type, TLR2-/-, TLR4-/- and MyD88-/- BALB/c mice. ASD collected from the desert was heated to remove toxic organic substances (H-ASD). BALB/c mice were instilled intratracheally with 12 different testing samples prepared with LPS (1 ng and 10 ng), H-ASD, and OVA in a normal saline solution. The lung pathology, cytological profiles in the bronchoalveolar lavage fluid (BALF), the levels of inflammatory cytokines/chemokines in BALF and OVA-specific immunoglobulin in serum were investigated.ResultsThe LPS exhibited no response to the production of TNF-α and IL-6 in BMDMs from TLR4-/-, but did from TLR2-/-. H-ASD aggravated the LPS-induced neutrophilic lung inflammation. In the presence of OVA, LPS increased the level of eosinophils slightly and induced trace levels of Th2 cytokines IL-5 and IL-13 at the levels of 1 ng and 10 ng. In the presence of OVA and H-ASD, LPS induced severe eosinophil infiltration and proliferation of goblet cells in the airways as well as remarkable increases in Th2 cytokines IL-5 and IL-13 in BALF. The mixture containing LPS (1 ng) showed adjuvant activity on OVA-specific IgE and IgG1 production.ConclusionsThe results suggest that H-ASD with naturally-occurring levels of LPS enhances OVA-induced lung eosinophilia via increases in Th2-mediated cytokines and antigen-specific immunoglobulin. These results indicate that LPS is a strong candidate for being a major aggravating substance in ASD

    BackgroundBjerkandera adusta (B. adusta) is one of the most important etiological fungi associated with chronic cough. However, precise details of the inflammatory response to exposure are not well understood yet. B. adusta was recently identified in Asian sand dust (ASD) aerosol. Therefore, in the present study the exacerbating effects of ASD on B. adusta-induced lung inflammation and B. adusta + ASD on ovalbumin (OVA)-induced murine lung eosinophilia were investigated using experimental mice.MethodsIn order to prepare testing samples, B. adusta obtained from ASD aerosol was inactivated by formalin and ASD collected from the atmosphere was heated to remove toxic organic substances (H-ASD). CD-1 mice were instilled intratracheally with 12 different samples prepared with various combinations of B. adusta, H-ASD, and OVA in a normal saline solution. The lung pathology, cytological profiles in bronchoalveolar lavage fluid (BALF), and the levels of inflammatory cytokines/chemokines in BALF were investigated.ResultsH-ASD aggravated the lung eosinophilia induced by B. adusta alone, which also aggravated the lung eosinophilia induced by OVA. The mixture of OVA, H-ASD, and B. adusta caused serious fibrous thickening of the subepithelial layer, eosinophil infiltration, and proliferation of goblet cells in the airways along with remarkable increases of IL-13, eotaxin, IL-5, and MCP-3 in BALF.ConclusionsThe results of the present study demonstrated that B. adusta isolated from ASD aerosol induces allergic lung diseases. H-ASD enhanced allergic reactions caused by OVA or B. adusta. A mixture of B. adusta, H-ASD, and OVA caused the most remarkable exacerbation to the allergic airway inflammation via remarkable increases of pro-inflammatory mediators

    Matter Distribution around Galaxies

    We explore the mass distribution of material associated with galaxies from the observation of gravitational weak lensing for the galaxy mass correlation function with the aid of NN-body simulations of dark matter. The latter is employed to unfold various contributions that contribute to the integrated line of sight mass density. We conclude that galaxies have no definite edges of the matter distribution, extending to the middle to neighbouring galaxies with the density profile roughly r2.4r^{-2.4} beyond the virial radius. The mass distributed beyond the virial radius (gravitationally bound radius) explains the gap seen in the mass density estimates, the global value Ωm0.27\Omega_m\sim 0.27 and typically Ωgal0.15\Omega_{\rm gal} \sim 0.15 from the luminosity density multiplied by the mass to light ratio. We suggest to use a physical method of gravitational lensing to characterise galaxy samples rather than characterise them with photometric means.Comment: 16 pages, 5 figures. ApJ accepte

    Diffuser total efficiency using generalized actuator disc model and its maximization method

    The diffuser total efficiency was formulated and defined based on the generalized actuator disc model for the index of the efficiency of the diffuser-alone of the diffuser-augmented wind turbines. An optimization method to maximize the diffuser total efficiency was developed using a genetic algorithm and axisymmetric computational fluid dynamics. A case study was conducted for a 10% chord-to-diameter ratio, 2% thickness-to-chord plate, and the crest position at 50% chord of the diffuser. The optimal result showed a diffuser total efficiency of 1.087. Furthermore, 1392 (=48 population × 29 generations) simulation cases of the optimization process showed that high diffuser total efficiency appears at a low-drag coefficient, high-lift coefficient, and 15–25% low diffuser height-to-chord ratio

    Total synthesis of palau’amine

    Palau’amine has received a great deal of attention in the past two decades as an attractive synthetic target by virtue of its intriguing molecular architecture and significant immunosuppressive activity. Here we report the total synthesis of palau’amine characterized by the construction of an ABDE tetracyclic ring core including a trans-bicylo[3.3.0]octane skeleton at a middle stage of total synthesis. The ABDE tetracyclic ring core is constructed by a cascade reaction of a cleavage of the N–N bond, including simultaneous formation of imine, the addition of amide anion to the resulting imine (D-ring formation) and the condensation of pyrrole with methyl ester (B-ring formation) in a single step. The synthetic palau’amine is confirmed to exhibit excellent immunosuppressive activity. The present synthetic route has the potential to help elucidate a pharmacophore as well as the mechanistic details of immunosuppressive activity

    Flower visitor fauna of the narrow endemic lily Lilium rubellum Baker in a lowland habitat in Yamagata, northern Japan

    Floral visitor fauna of the narrow endemic lily Lilium rubellum was examined in a lowland habitat in Kaminoyama City, Yamagata Prefecture, northern Japan. Flowers of L. rubellum bloomed from early to late June. During 23 h of observing floral visitors, 64 insects were detected on L. rubellum flowers. Although coleopteran insects were most frequently found on L. rubellum flowers, they did not seem to be effective pollinators because of their body size. Bees were less frequently observed than coleopteran insects, but all individuals contacted sexual organs in L. rubellum flowers. Syrphid flies were seen less frequently, but they were also considered effective pollinators. From our observations, L. rubellum is a bee-pollinated species of the genus Lilium. Bee pollination has also been recorded in a species of Lilium sect. Archelirion, L. japonicum var. abeanum. The floral characteristics of L. japonicum var. abeanum (e.g., pinkish color, relatively small and tubular corolla, and lateral insertion of anthers into the corolla) were similar to those of L. rubellum and the character combination may be related to the bee-pollination mode in Lilium. key words: bee pollination, Bombus, Ceratina, flower visitor fauna, Lilium japonicum var. abeanum, Lilium rubellum, nitidulid beetle, syrphid fl

    Electrical cardiometry for hemodynamics

    Few reports have focused on hemodynamics around delivery in pregnant women because of the difficulty of continuous and noninvasive measurement. Electrical cardiometry allows noninvasive continuous monitoring of hemodynamics and has recently been used in non-pregnant subjects. We compared the use of electrical cardiometry versus transthoracic echocardiography in healthy pregnant women and evaluated hemodynamics immediately after vaginal delivery. In Study 1, electrical cardiometry and transthoracic echocardiography were used to measure cardiac output in 20 pregnant women with threatened premature delivery. A significant correlation was found between the two methods, with electrical cardiometry showing the higher cardiac output. In Study 2, heart rate, stroke volume, and cardiac output were continuously measured in 15 women during vaginal delivery up to 2 h postpartum. Cardiac output increased markedly because of an increased heart rate and stroke volume at the time of newborn delivery. The heart rate then immediately returned to baseline, while cardiac output remained elevated for at least 2 h after delivery because of a sustained high stroke volume. Electrical cardiometry was as readily available as transthoracic echocardiography for evaluating hemodynamics and allowed for continuous measurement during labor.
