    Canopy Multilayering and Woody Species Diversity of a Subtropical Evergreen Broadleaf Forest, Okinawa Island.

    v. ill. 23 cm.QuarterlyWoody species diversity and the spatial distribution of trees in a subtropical evergreen broadleaf forest on a silicate substrate, Okinawa Island, were investigated to determine the forest’s architectural stratification. The forest stand consisted of four architectural layers. The values of Shannon’s index H0 and Pielou’s index J 0 tended to increase from the top layer downward, except for the bottom layer. The lower layers contained many species relative to their smaller height ranges. High woody species diversity of the forest depended on small trees. This trend of species diversity was different from that of forest on a limestone substrate on Okinawa Island, where high woody species diversity depended on large trees. Conservation of small trees in the lower layers, especially the bottom layer, is indispensable to maintain diversity in Okinawan evergreen broadleaf forests. Castanopsis sieboldii (Mak.) Hatusima had the highest importance value in all layers, indicating that it is typically a facultative shade species as well as a climax species. The spatial distribution patterns of trees were found to be random in the lower three layers, but in the top layer clumping seemed to occur at three spatial scales. A high degree of overlapping in spatial distributions of trees among the layers suggested that light cannot penetrate easily into the lower layers. As a result, most species in the lower layers must be shade-tolerant. Mean weight index decreased from the top toward the bottom layer, and tree density increased from the top downward. This trend resembled the mean weight-density trajectory of self-thinning plant populations

    High-Throughput Identification and Screening of Novel Methylobacterium Species Using Whole-Cell MALDI-TOF/MS Analysis

    Methylobacterium species are ubiquitous α-proteobacteria that reside in the phyllosphere and are fed by methanol that is emitted from plants. In this study, we applied whole-cell matrix-assisted laser desorption/ionization time-of-flight mass spectrometry analysis (WC-MS) to evaluate the diversity of Methylobacterium species collected from a variety of plants. The WC-MS spectrum was reproducible through two weeks of cultivation on different media. WC-MS spectrum peaks of M. extorquens strain AM1 cells were attributed to ribosomal proteins, but those were not were also found. We developed a simple method for rapid identification based on spectra similarity. Using all available type strains of Methylobacterium species, the method provided a certain threshold similarity value for species-level discrimination, although the genus contains some type strains that could not be easily discriminated solely by 16S rRNA gene sequence similarity. Next, we evaluated the WC-MS data of approximately 200 methylotrophs isolated from various plants with MALDI Biotyper software (Bruker Daltonics). Isolates representing each cluster were further identified by 16S rRNA gene sequencing. In most cases, the identification by WC-MS matched that by sequencing, and isolates with unique spectra represented possible novel species. The strains belonging to M. extorquens, M. adhaesivum, M. marchantiae, M. komagatae, M. brachiatum, M. radiotolerans, and novel lineages close to M. adhaesivum, many of which were isolated from bryophytes, were found to be the most frequent phyllospheric colonizers. The WC-MS technique provides emerging high-throughputness in the identification of known/novel species of bacteria, enabling the selection of novel species in a library and identification without 16S rRNA gene sequencing

    The Yuu-yuu Health Circle is a place for students to learn and deepen their understanding and interest by putting into practice the knowledge and skills they have gained in class at the university. This chapter introduces its maintenance and promotion activities for older adults in the community that were participated by students of the Departments of Physical Therapy and Occupational Therapy at Kinjo University before and during the coronavirus pandemic. The activity method and contents changed significantly during the pandemic. However, as a medical and welfare university, its job to develop human resources that deal with people has not changed. As an educational method toward such purpose, the activities helped students, who need to actively communicate with people of different ages and improve their interpersonal communication skills even during the pandemic

    Weak Values with Decoherence

    The weak value of an observable is experimentally accessible by weak measurements as theoretically analyzed by Aharonov et al. and recently experimentally demonstrated. We introduce a weak operator associated with the weak values and give a general framework of quantum operations to the W operator in parallel with the Kraus representation of the completely positive map for the density operator. The decoherence effect is also investigated in terms of the weak measurement by a shift of a probe wave function of continuous variable.     Daily 1,000 IU vitamin D was supplemented to 87 years old female(Case 1)and 68 years old male(Case 2)subjects with sarcopenia and vitamin D insufficiency/deficiency for 2 years and 5 months and 2 years and 7 months, respectively. Before, 1 year and 9 months, and 2 years and 5 months after vitamin D supplementation in Case 1, serum 25‐hydroxyvitamin D[25(OH)D]level was 9 ng/ml, 23.8 ng/ml and 23.1 ng/ml, skeletal muscle mass was 8.95 kg, 10.2 kg and 10.0 kg, handgrip strength was 7.3 kg, 8.9 kg and 9.9 kg, and Barthel index was 40, 85 and 90, respectively. Before, and 2 years and 7 months after vitamin D supplementation in Case 2, serum 25(OH)D level was 26 ng/ml and 34.4 ng/ml, skeletal muscle mass was 24.6 kg and 25.1 kg, handgrip strength was 37.2 kg and 38.3 kg, and Barthel index was 100 and 100, respectively. Thus, activity of daily living(ADL)markedly improved in Case 1 and keep high in Case 2. Therefore, it is suggested that daily 1,000 IU vitamin D supplementation is effective to keep QOL of aged persons higher

    Antifibrotic effects of CXCR4 antagonist in bleomycin-induced pulmonary fibrosis in mice

    Circulating fibrocytes had been reported to migrate into the injured lungs, and contribute to fibrogenesis via chemokine-chemokine receptor systems including CXCL12-CXCR4 axis. Here we hypothesized that blockade of CXCR4 might inhibit the migration of fibrocytes to the injured lungs and the subsequent pulmonary fibrosis. To explore the antifibrotic effects of blockade of CXCR4, we used a specific antagonist for CXCR4, AMD3100, in bleomycin-induced pulmonary fibrosis model in mice. Administration of AMD3100 significantly improved the loss of body weight of mice treated with bleomycin, and inhibited the fibrotic lesion in subpleural areas of the lungs. The quantitative analysis demonstrated that treatment with AMD3100 reduced the collagen content and fibrotic score (Aschcroft score) in the lungs. Although AMD3100 did not affect cell classification in bronchoalveolar lavage fluid on day 7, the percentage of lymphocytes was reduced by AMD3100 on day 14. AMD3100 directly inhibited the migration of human fibrocytes in response to CXCL12 in vitro, and reduced the trafficking of fibrocytes into the lungs treated with bleocmycin in vivo. These results suggest that the blockade of CXCR4 might be useful strategy for therapy of patients with pulmonary fibrosis via inhibiting the migration of circulating fibrocytes

    Significant association of RNF213 p.R4810K, a moyamoya susceptibility variant, with coronary artery disease

    Background The genetic architecture of coronary artery disease has not been fully elucidated, especially in Asian countries. Moyamoya disease is a progressive cerebrovascular disease that is reported to be complicated by coronary artery disease. Because most Japanese patients with moyamoya disease carry the p.R4810K variant of the ring finger 213 gene (RNF213), this may also be a risk factor for coronary artery disease; however, this possibility has never been tested. Methods and results We genotyped the RNF213 p.R4810K variant in 956 coronary artery disease patients and 716 controls and tested the association between p.R4810K and coronary artery disease. We also validated the association in an independent population of 311 coronary artery disease patients and 494 controls. In the replication study, the p.R4810K genotypes were imputed from genome-wide genotyping data based on the 1000 Genomes Project. We used multivariate logistic regression analyses to adjust for well-known risk factors such as dyslipidemia and smoking habits. In the primary study population, the frequency of the minor variant allele was significantly higher in patients with coronary artery disease than in controls (2.04% vs. 0.98%), with an odds ratio of 2.11 (p = 0.017). Under a dominant model, after adjustment for risk factors, the association remained significant, with an odds ratio of 2.90 (95% confidence interval: 1.37-6.61; p = 0.005). In the replication study, the association was significant after adjustment for age and sex (odds ratio = 4.99; 95% confidence interval: 1.16-21.53; p = 0.031), although it did not reach statistical significance when further adjusted for risk factors (odds ratio = 3.82; 95% confidence interval: 0.87-16.77; p = 0.076). Conclusions The RNF213 p.R4810K variant appears to be significantly associated with coronary artery disease in the Japanese population

    RNA-Binding Protein Musashi1 Modulates Glioma Cell Growth through the Post-Transcriptional Regulation of Notch and PI3 Kinase/Akt Signaling Pathways

    Musashi1 (MSI1) is an RNA-binding protein that plays critical roles in nervous-system development and stem-cell self-renewal. Here, we examined its role in the progression of glioma. Short hairpin RNA (shRNA)-based MSI1-knock down (KD) in glioblastoma and medulloblastoma cells resulted in a significantly lower number of self renewing colony on day 30 (a 65% reduction), compared with non-silencing shRNA-treated control cells, indicative of an inhibitory effect of MSI1-KD on tumor cell growth and survival. Immunocytochemical staining of the MSI1-KD glioblastoma cells indicated that they ectopically expressed metaphase markers. In addition, a 2.2-fold increase in the number of MSI1-KD cells in the G2/M phase was observed. Thus, MSI1-KD caused the prolongation of mitosis and reduced the cell survival, although the expression of activated Caspase-3 was unaltered. We further showed that MSI1-KD glioblastoma cells xenografted into the brains of NOD/SCID mice formed tumors that were 96.6% smaller, as measured by a bioluminescence imaging system (BLI), than non-KD cells, and the host survival was longer (49.3±6.1 days vs. 33.6±3.6 days; P<0.01). These findings and other cell biological analyses suggested that the reduction of MSI1 in glioma cells prolonged the cell cycle by inducing the accumulation of Cyclin B1. Furthermore, MSI1-KD reduced the activities of the Notch and PI3 kinase-Akt signaling pathways, through the up-regulation of Numb and PTEN, respectively. Exposure of glioma cells to chemical inhibitors of these pathways reduced the number of spheres and living cells, as did MSI1-KD. These results suggest that MSI1 increases the growth and/or survival of certain types of glioma cells by promoting the activation of both Notch and PI3 kinase/Akt signaling
