    Loss of Pikfyve causes Transdifferentiation of Dictyostelium Spores into Basal Disc Cells

    The 1-phosphatidylinositol-3-phosphate 5-kinase PIKfyve generates PtdIns3,5P2 on late phagolysosomes, which by recruiting the scission protein Atg18, results in their fragmentation in the normal course of endosome processing. Loss of PIKfyve function causes cellular hypervacuolization in eukaryotes and organ failure in humans. We identified pikfyve as the defective gene in a Dictyostelium mutant that failed to form spores. The amoebas normally differentiated into prespore cells and initiated spore coat protein synthesis in Golgi-derived prespore vesicles. However, instead of exocytosing, the prespore vesicles fused into the single vacuole that typifies the stalk and basal disc cells that support the spores. This process was accompanied by stalk wall biosynthesis, loss of spore gene expression and overexpression of ecmB, a basal disc and stalk-specific gene, but not of the stalk-specific genes DDB_G0278745 and DDB_G0277757. Transdifferentiation of prespore into stalk-like cells was previously observed in mutants that lack early autophagy genes, like atg5, atg7, and atg9. However, while autophagy mutants specifically lacked cAMP induction of prespore gene expression, pikfyve(−) showed normal early autophagy and prespore induction, but increased in vitro induction of ecmB. Combined, the data suggest that the Dictyostelium endosomal system influences cell fate by acting on cell type specific gene expression

    Conditions Regional Development Bank (BPD) in terms of Business Growth, Markets and Competitiveness within the last three years shows an alarming condition. The study aims to assess the impact of market structure and efficiency of the performance of the BPD in Indonesia. The data used is secondary data in the form of financial statements of banks BPD in 2004-2013. The analytical tool used in this research is panel data regression. The results found that the banking industry in particular BPD in Indonesia supports the traditional hypothesis that assumes that the concentration is a proxy of market power (market power). This study has used the narrative of an elderly man to learn about what has given his life meaning.Method: Fifteen key concepts were extracted from interviews using the narrative approach.Results and Conclusions: Mr. A, now near the end of his life, has found meaning in living his life, not only for himself, but also for those around him. The elderly support their own physical and mental health,and adjust to day-to-day life. Nursing care for the elderly should watch for and support these self-supporting efforts

    Biochemical analyses of lipids deposited on silicone hydrogel lenses

    AbstractPurposeThis study was performed to determine the levels of lipids deposited on in vivo worn silicone hydrogel lenses.MethodsThree silicone hydrogel materials, galyfilcon A, senofilcon A, and asmofilcon A, were worn for 2 weeks by 35 normal subjects. Total lipid deposition was determined by the sulfo-phospho-vanillin reaction. Cholesterol was estimated by a colorimetric probe through enzymatic oxidation. Phospholipid level was estimated by determining phosphorus with ammonium molybdate through enzymatic digestion.ResultsThe total lipid content recovered from galyfilcon A, senofilcon A, and asmofilcon A was 32.9 ± 33.8, 42.1 ± 14.0, and 36.6 ± 31.9 μg/lens, respectively. The cholesterol content recovered from galyfilcon A, senofilcon A, and asmofilcon A was 26.2 ± 26.9, 28.6 ± 19.4, and 31.1 ± 21.1 μg/lens, respectively. There were no statistically significant differences in total lipids and cholesterol among the contact lens types. However, the quantity of phospholipid recovered from the asmofilcon A (7.0 ± 5.5 μg/lens) lenses was significantly higher than from galyfilcon A (1.1 ± 0.8 μg/lens) and senofilcon A (2.4 ± 0.8 mg/lens) lenses (p < 0.05, Mann-Whitney test).ConclusionsThe quantity of total lipid and cholesterol deposited on the 3 silicone hydrogel lenses tested did not differ. However, there were significant differences in the amounts of phospholipid deposited among the 3 silicone hydrogel lenses, of which clinical significance should be explored in the future study
