    Unique incorporation behavior of amino acid-type surfactant into phospholipid vesicle membrane

    金沢大学工学部The incorporation behavior of some anionic surfactants, including amino acid-type surfactants, on phospholipid vesicles was investigated. This was done by measuring the release of a vesicle-entrapped fluorescence probe and the scattered light intensities of vesicle particles in the surfactant solution as a function of surfactant concentration and time. Sodium dodecyl sulfate, sodium dodecanesulfonate, sodium dodecanoyl sarcosinate, and sodium dodecanoyl glutamate were employed in this study. All surfactants ruptured the phospholipid vesicle at around each critical micelle concentration by mixed micelle formation with phospholipid. While leakage of the fluorescence probe took place at a very low concentration in the sulfate- or sulfonate-type surfactant systems, it occurred at the concentration just below the CMC in the amino acid-type surfactant systems. Kinetic analysis of the release of the probe from the vesicles showed that the former surfactants adsorbed independently and homogeneously onto the phospholipid vesicles, while the latter surfactants were cooperatively incorporated

    The duplex stability with target mRNA and the gene silencing potential of a novel bridged nucleic acid analogue are described. The analogue, 2′,4′-BNANC antisense oligonucleotides (AONs) ranging from 10- to 20-nt-long, targeted apolipoprotein B. 2′,4′-BNANC was directly compared to its conventional bridged (or locked) nucleic acid (2′,4′-BNA/LNA)-based counterparts. Melting temperatures of duplexes formed between 2′,4′-BNANC-based antisense oligonucleotides and the target mRNA surpassed those of 2′,4′-BNA/LNA-based counterparts at all lengths. An in vitro transfection study revealed that when compared to the identical length 2′,4′-BNA/LNA-based counterpart, the corresponding 2′,4′-BNANC-based antisense oligonucleotide showed significantly stronger inhibitory activity. This inhibitory activity was more pronounced in shorter (13-, 14-, and 16-mer) oligonucleotides. On the other hand, the 2′,4′-BNANC-based 20-mer AON exhibited the highest affinity but the worst IC50 value, indicating that very high affinity may undermine antisense potency. These results suggest that the potency of AONs requires a balance between reward term and penalty term. Balance of these two parameters would depend on affinity, length, and the specific chemistry of the AON, and fine-tuning of this balance could lead to improved potency. We demonstrate that 2′,4′-BNANC may be a better alternative to conventional 2′,4′-BNA/LNA, even for “short” antisense oligonucleotides, which are attractive in terms of drug-likeness and cost-effective bulk production

    Relaxin-3-Deficient Mice Showed Slight Alteration in Anxiety-Related Behavior

    Relaxin-3 is a neuropeptide belonging to the relaxin/insulin superfamily. Studies using rodents have revealed that relaxin-3 is predominantly expressed in neurons in the nucleus incertus (NI) of the pons, the axons of which project to forebrain regions including the hypothalamus. There is evidence that relaxin-3 is involved in several functions, including food intake and stress responses. In the present study, we generated relaxin-3 gene knockout (KO) mice and examined them using a range of behavioral tests of sensory/motor functions and emotion-related behaviors. The results revealed that relaxin-3 KO mice exhibited normal growth and appearance, and were generally indistinguishable from wild genotype littermates. There was no difference in bodyweight among genotypes until at least 28 weeks after birth. In addition, there were no significant differences between wild-type and KO mice in locomotor activity, social interaction, hot plate test performance, fear conditioning, depression-like behavior, and Y-maze test performance. However, in the elevated plus maze test, KO mice exhibited a robust increase in the tendency to enter open arms, although they exhibited normal performance in a light/dark transition test and showed no difference from wild-type mice in the time spent in central area in the open field test. On the other hand, a significant increase in the acoustic startle response was observed in KO mice. These results indicate that relaxin-3 is slightly involved in the anxiety-related behavior

    Conceptual Design of Electron Density Measurement System for DEMO-Relevant Helical Plasmas

    Electron density measurement remains indispensable to control fueling on a DEMO reactor. For steady-state operation of the DEMO reactor, density measurement should be highly reliable and accurate. A dispersion interferometer and a Faraday polarimeter are free from measurement errors caused by mechanical vibrations. Hence combination of the two diagnostics yields a suitable system for density measurement on future steady-state fusion reactors. A wavelength around 1 ?m is one of the desirable candidates in terms of the fringe shift and the Faraday rotation angle, the variety of optical components, and the efficiency of frequency doubling for the dispersion interferometer. This paper presents a conceptual design for the dispersion interferometer and Faraday polarimeter with a 1 ?m light source

    Effects of Resonant Magnetic Perturbation on Particle Transport in LHD

    In this study, the effects of resonant magnetic perturbation (RMP) on particle transport are investigated in Large Helical device (LHD). The magnetic configuration is selected to be the outwardly shifted configuration, for which the magnetic axis position (Rax) is 3.9 m. At Rax = 3.9 m, the main plasma is surrounded by a thick ergodic layer, with width of about 30% of the plasma minor radius. The perturbation mode m/n = 1/1, where m and n are poloidal and toroidal mode numbers, is applied. The resonant layer is around the last closed flux surface. With RMP, a region in which both the connection and Kolmogorov lengths are finite and the magnetic field is ergodic forms; this region extends inside the main plasma. In the low-collisionality regime, where νh* 1), a clear difference in particle transport is found. A clear difference in turbulence is also observed, suggesting that turbulence plays a significant role in particle transport in the high-collisionality regime both with and without RMP

    Development of impurity seeding and radiation enhancement in the helical divertor of LHD

    Impurity seeding to reduce the divertor heat load was conducted in the large helical device (LHD) using neon (Ne) and krypton (Kr) puffing. Radiation enhancement and reduction of the divertor heat load were observed. In the LHD, the ratio between the total radiated power and the heating power, f rad = Prad/Pheating, is limited up to around 30% in hydrogen plasmas even for high density plasma just below the radiative collapse (ne, bar  >  1   ×   1020 m−3), where ne, bar is the line averaged density. With Ne seeding, the ratio could be raised to 52% at ne, bar ~ 1.3   ×   1019 m−3, albeit with a slight reduction in confinement. f rad ~ 30% could be sustained for 3.4 s using multi-pulse Ne seeding at ne, bar ~ 4   ×   1019 m−3. The localized supplemental radiation was observed along the helical divertor X-points (HDXs) which is similar to the estimated structure by the EMC3-EIRENE code. Kr seeding was also conducted at ne, bar ~ 3.1   ×   1019 m−3. f rad ~ 25% was obtained without a significant change in stored energy. The radiation enhancement had a slower time constant. The supplemental radiation area of the Kr seeded plasma moved from the HDXs to the core plasma. Highly charged states of Kr ions are considered to be the dominant radiators from the plasma core region

    High time resolved electron temperature measurements by using the multi-pass Thomson scattering system in GAMMA 10/PDX

    High time resolved electron temperature measurements are useful for fluctuation study. A multi-pass Thomson scattering (MPTS) system is proposed for the improvement of both increasing the TS signal intensity and time resolution. The MPTS system in GAMMA 10/PDX has been constructed for enhancing the Thomson scattered signals for the improvement of measurement accuracy. The MPTS system has a polarization-based configuration with an image relaying system. We optimized the image relaying optics for improving the multi-pass laser confinement and obtaining the stable MPTS signals over ten passing TS signals. The integrated MPTS signals increased about five times larger than that in the single pass system. Finally, time dependent electron temperatures were obtained in MHz sampling

    Collisionality dependence and ion species effects on heat transport in He and H plasma, and the role of ion scale turbulence in LHD

    Surveys of the ion and electron heat transports of neutral beam (NB) heating plasma were carried out by power balance analysis in He and H rich plasma at LHD. Collisionality was scanned by changing density and heating power. The characteristics of the transport vary depending on collisionality. In low collisionality, with low density and high heating power, an ion internal transport barrier (ITB) was formed. The ion heat conductivity (χi) is lower than electron heat conductivity (χe) in the core region at ρ  <  0.7. On the other hand, in high collisionality, with high density and low heating power, χi is higher than χe across the entire range of plasma. These different confinement regimes are associated with different fluctuation characteristics. In ion ITB, fluctuation has a peak at ρ  =  0.7, and in normal confinement, fluctuation has a peak at ρ  =  1.0. The two confinement modes change gradually depending on the collisionality. Scans of concentration ratio between He and H were also performed. The ion confinement improvements were investigated using gyro-Bohm normalization, taking account of the effective mass and charge. The concentration ratio affected the normalized χi only in the edge region (ρ ~ 1.0). This indicates ion species effects vary depending on collisionality. Turbulence was modulated by the fast ion loss instability. The modulation of turbulence is higher in H rich than in He rich plasma
