    The coordinated roles of miR-26a and miR-30c in regulating TGFβ1-induced epithelial-to-mesenchymal transition in diabetic nephropathy

    MicroRNAs (miRNAs) play vital roles in the development of diabetic nephropathy. Here, we compared the protective efficacies of miR-26a and miR-30c in renal tubular epithelial cells (NRK-52E) and determined whether they demonstrated additive effects in the attenuation of renal fibrosis. TGFβ1 suppressed miR-26a and miR-30c expression but up-regulated pro-fibrotic markers in NRK-52E cells, and these changes were also found in the kidney cortex of 40-week-old diabetic Otsuka Long-Evans Tokushima fatty (OLETF) rats. Bioinformatic analyses and luciferase assays further demonstrated that both miR-26a and miR-30c targeted connective tissue growth factor (CTGF); additionally, Snail family zinc finger 1 (Snail1), a potent epithelial-to-mesenchymal transition (EMT) inducer, was targeted by miR-30c. Overexpression of miR-26a and miR-30c coordinately decreased CTGF protein levels and subsequently ameliorated TGFβ1-induced EMT in NRK-52E cells. Co-silencing of miR-26a and miR-30c exhibited the opposite effect. Moreover, miR-26a and miR-30c co-silenced CTGF to decrease ERK1/2 and p38 MAPK activation. Furthermore, miR-26a was up-regulated in urinary extracellular vesicles of diabetic nephropathy patients. Our study provides evidence for the cooperative roles of miR-26a and miR-30c in the pathogenesis of diabetic nephropathy, and the co-targeting of miR-26a and miR-30c could provide a new direction for diabetic nephropathy treatment

    BnaMPK3 Is a Key Regulator of Defense Responses to the Devastating Plant Pathogen Sclerotinia sclerotiorum in Oilseed Rape

    The disease caused by Sclerotinia sclerotiorum has traditionally been difficult to control, resulting in tremendous economic losses in oilseed rape (Brassica napus). Identification of important genes in the defense responses is critical for molecular breeding, an important strategy for controlling the disease. Here, we report that a B. napus mitogen-activated protein kinase gene, BnaMPK3, plays an important role in the defense against S. sclerotiorum in oilseed rape. BnaMPK3 is highly expressed in the stems, flowers and leaves, and its product is localized in the nucleus. Furthermore, BnaMPK3 is highly responsive to infection by S. sclerotiorum and treatment with jasmonic acid (JA) or the biosynthesis precursor of ethylene (ET), but not to treatment with salicylic acid (SA) or abscisic acid. Moreover, overexpression (OE) of BnaMPK3 in B. napus and Nicotiana benthamiana results in significantly enhanced resistance to S. sclerotiorum, whereas resistance is diminished in RNAi transgenic plants. After S. sclerotiorum infection, defense responses associated with ET, JA, and SA signaling are intensified in the BnaMPK3-OE plants but weakened in the BnaMPK3-RNAi plants when compared to those in the wild type plants; by contrast the level of both H2O2 accumulation and cell death exhibits a reverse pattern. The candidate gene association analyses show that the BnaMPK3-encoding BnaA06g18440D locus is a cause of variation in the resistance to S. sclerotiorum in natural B. napus population. These results suggest that BnaMPK3 is a key regulator of multiple defense responses to S. sclerotiorum, which may guide the resistance improvement of oilseed rape and related economic crops

    Assessment of Grassland Ecosystem Service Value in Response to Climate Change in China

    The assessment of ecosystem services provides an intuitive source of information on the benefits humans derive from ecosystems. The equivalent factor method was applied to calculate the ecosystem service value (ESV) in combination with net primary productivity (NPP) calculated by the process-based Carnegie–Ames–Stanford approach (CASA) model. This study evaluated grassland ESV and its spatial evolution characteristics in China from 2001 to 2020 and revealed the impact of climate factors. For 2001–2020, the annual grassland ESV ranged from 1.17 × 1012 to 1.51 × 1012 yuan (renminbi, China yuan—the same below; 0.15×1012–0.15 × 1012–0.20 × 1012, US dollar), with an average of 1.37 × 1012 yuan (0.18×1012).ThespatialpatternofESVperunitareaofgrasslandwasnotablycharacterizedbyanincreasefromnorthwesttosoutheast.However,thevalueofgrasslandecosystemserviceswasrelativelylarge(exceeding10×106yuan;0.18 × 1012). The spatial pattern of ESV per unit area of grassland was notably characterized by an increase from northwest to southeast. However, the value of grassland ecosystem services was relatively large (exceeding 10 × 106 yuan; 1.30 × 106) in northern and western provinces and was the lowest (less than 0.2 × 106 yuan; $0.03 × 106) in eastern and southern provinces. In the last 20 years, grassland ESV has increased in most areas of China and has decreased only in some western and northern areas. Compared with the first 10 years, the average ESV of grassland in most areas increased in the last 10 years, usually by less than 20%. However, it decreased in the western and northern parts of China, mainly concentrated in the alpine meadow and alpine grassland of the Qinghai–Tibet Plateau and the grassland around the Yili region of Xinjiang. Precipitation was the main regulating factor of grassland ESV and had a positive impact in 79% grassland areas, especially in northern China. Evapotranspiration and sunshine hours exhibited a marginal impact on ESV, but temperature and relative humidity had no significant effect. Overall, this study contributes to exploring the spatiotemporal patterns of grassland ecosystem service value and the impact of climate factors in China, thereby providing reliable guidance for grassland ecosystem management

    The Applications of Polymers in Solar Cells: A Review

    The emerging dye-sensitized solar cells, perovskite solar cells, and organic solar cells have been regarded as promising photovoltaic technologies. The device structures and components of these solar cells are imperative to the device’s efficiency and stability. Polymers can be used to adjust the device components and structures of these solar cells purposefully, due to their diversified properties. In dye-sensitized solar cells, polymers can be used as flexible substrates, pore- and film-forming agents of photoanode films, platinum-free counter electrodes, and the frameworks of quasi-solid-state electrolytes. In perovskite solar cells, polymers can be used as the additives to adjust the nucleation and crystallization processes in perovskite films. The polymers can also be used as hole transfer materials, electron transfer materials, and interface layer to enhance the carrier separation efficiency and reduce the recombination. In organic solar cells, polymers are often used as donor layers, buffer layers, and other polymer-based micro/nanostructures in binary or ternary devices to influence device performances. The current achievements about the applications of polymers in solar cells are reviewed and analyzed. In addition, the benefits of polymers for solar cells, the challenges for practical application, and possible solutions are also assessed
