    Quick and sensitive determination of gene expression of fatty acid synthase in vitro by using real-time polymerase chain reaction amplification (PCR)

    Obesity results from an imbalance between energy intake and energy expenditure, which leads to a pathological accumulation of adipose tissue, but the underlying mechanism at gene level, is far from being elucidated. The objective of this study was to investigate the correlation between mRNA express from fatty acid synthase (FAS) with a different glucose level in primary adipocytes by real-time polymerase chain reaction amplification (PCR), which can aid in the understanding of the mechanism of obesity in vitro. By using the following formula, this study was able to quantify the mRNA expression of FAS of unknown samples: Y = -3.156X + 41.21 (Y = threshold cycle, X = log starting quantity). The high concentrations of glucose group significantly improved the mRNA expression of FAS (P < 0.01) rather than 0.25 and 0% concentrations of glucose. These results provide significant data that confirm an association between different glucose level and FAS expression in preadipocytes. The glucose concentration of the high group substantially augmented the mRNA expression of FAS.Key words: Expression, fatty acid synthase, lipid deposition, real-time polymerase chain reaction amplification (PCR)

    Prevalence and Risk Factors for Poor Mental Health and Suicidal Ideation in the Nigerian Construction Industry

    Employees' mental health and well-being are a growing concern as countries grapple with heightened mental ill-health and suicide rates, with the construction industry ranking in the top five. Drawing from the context of a developing country, particularly Nigeria, data were collected using mixed methods (i.e., survey and postsurvey interview). The study investigated mental health among 382 construction personnel. The Patient Health Questionnaire (PHQ-9) and Generalized Anxiety Disorder (GAD-2) scales were employed in assessing mental health. The data collected were analyzed using descriptive statistics and logistic regression. The prevalence rates of depression, anxiety, and suicidal ideation were 55.1%, 16%, 9.8% among supervisors, 72%, 36.9%, and 15.3%, respectively, among tradesmen. Univariate logistic regression revealed that years of experience and organizational structure influence mental health. Multiple logistic regression showed that risk factors for mental ill-health include nature of work, little relationship with colleagues, job insecurity, occupational injury, low income, and unsatisfactory living conditions. The findings from this study will assist in intervention building and policy-making within the construction industry of Nigeria and globally

    Fractional Anisotropy in Corpus Callosum Is Associated with Facilitation of Motor Representation during Ipsilateral Hand Movements

    BACKGROUND: Coactivation of primary motor cortex ipsilateral to a unilateral movement (M1(ipsilateral)) has been observed, and the magnitude of activation is influenced by the contracting muscles. It has been suggested that the microstructural integrity of the callosal motor fibers (CMFs) connecting M1 regions may reflect the observed response. However, the association between the structural connectivity of CMFs and functional changes in M1(ipsilateral) remains unclear. The purpose of this study was to investigate the relationship between functional changes within M1(ipsilateral) during unilateral arm or leg movements and the microstructure of the CMFs connecting both homotopic representations (arm or leg). METHODS: Transcranial magnetic stimulation was used to assess changes in motor evoked potentials (MEP) in an arm muscle during unilateral movements compared to rest in fifteen healthy adults. Functional magnetic resonance imaging was then used to identify regions of M1 associated with either arm or leg movements. Diffusion-weighted imaging data was acquired to generate CMFs for arm and leg areas using the areas of activation from the functional imaging as seed masks. Individual values of regional fractional anisotropy (FA) of arm and leg CMFs was then calculated by examining the overlap between CMFs and a standard atlas of corpus callosum. RESULTS: The change in the MEP was significantly larger in the arm movement compared to the leg movement. Additionally, regression analysis revealed that FA in the arm CMFs was positively correlated with the change in MEP during arm movement, whereas a negative correlation was observed during the leg movement. However, there was no significant relationship between FA in the leg CMF and the change in MEP during the movements. CONCLUSIONS: These findings suggest that individual differences in interhemispheric structural connectivity may be used to explain a homologous muscle-dominant effect within M1(ipsilateral) hand representation during unilateral movement with topographical specificity

    Brief Report: A Cross-Sectional Study of Anxiety Levels and Concerns of Chinese Families of Children With Special Educational Needs and Disabilities Post-first-wave of COVID-19

    The COVID-19 pandemic has a multifaceted impact on mental health due to ill health, restrictions and lockdowns, and loss of employment and institutional support. COVID-19 may disproportionally impact families with special educational needs and disabilities (SEND) due to the already higher prevalence of mental health conditions in children with SEND and their parents. Therefore, it is essential to determine the short-term impact of the pandemic on the mental health of families with SEND in order to identify their ongoing health support needs. The current study aims to examine the anxiety level and concerns of children with SEND and their parents living in China. The sample consisted of 271 parents of children with SEND aged between 6 and 17 years (M age = 8.37; SD age = 2.76). Parents completed an online survey between 10 April to 8 June 2020. Both child and parental anxiety levels and various concerns increased after the initial wave of COVID-19 when compared with retrospective pre-COVID-19 levels. Parental anxiety and concern levels were significantly higher for those living in rural areas compared to urban areas. In addition, parental and child anxiety and concern levels were significantly correlated with each other. Parental anxiety at the lowest level made a unique and significant statistical contribution to children's anxiety levels. The implications of the study findings are discussed

    Establishing production service system and information collaboration platform for mold and die products

    This paper investigates how the new concept of product service systems can be used and extended to transform, elevate, and revitalize traditional equipment manufacturing industry such as the Mold and Die (MD) sector. A mold and die production service systems (MPSS) framework is established based on recent developments within our industrial collaborators. Within the MPSS framework, MD manufacturers become more specialized in producing MD products and components while sharing and outsourcing manufacturing-oriented services (MOS) from a service provider. Typical services include collaborative order pooling and release, collaborative project progress status tracking, contractor-managed collaborative outsourcing, collaborative product design, collaborative production planning and scheduling, and after-sales technical supports. MOSs are designed, developed, and deployed as SaaS (software as application services) following the service-oriented architecture. Collectively, they form iMPSS-an Information and Collaboration Platform that enables MPSS.     LNK (SH2B3): paradoxical effects in ovarian cancer.

    LNK (SH2B3) is an adaptor protein studied extensively in normal and malignant hematopoietic cells. In these cells, it downregulates activated tyrosine kinases at the cell surface resulting in an antiproliferative effect. To date, no studies have examined activities of LNK in solid tumors. In this study, we found by in silico analysis and staining tissue arrays that the levels of LNK expression were elevated in high-grade ovarian cancer. To test the functional importance of this observation, LNK was either overexpressed or silenced in several ovarian cancer cell lines. Remarkably, overexpression of LNK rendered the cells resistant to death induced by either serum starvation or nutrient deprivation, and generated larger tumors using a murine xenograft model. In contrast, silencing of LNK decreased ovarian cancer cell growth in vitro and in vivo. Western blot studies indicated that overexpression of LNK upregulated and extended the transduction of the mitogenic signal, whereas silencing of LNK produced the opposite effects. Furthermore, forced expression of LNK reduced cell size, inhibited cell migration and markedly enhanced cell adhesion. Liquid chromatography-mass spectroscopy identified 14-3-3 as one of the LNK-binding partners. Our results suggest that in contrast to the findings in hematologic malignancies, the adaptor protein LNK acts as a positive signal transduction modulator in ovarian cancers

    Waterproofed Photomultiplier Tube Assemblies for the Daya Bay Reactor Neutrino Experiment

    In the Daya Bay Reactor Neutrino Experiment 960 20-cm-diameter waterproof photomultiplier tubes are used to instrument three water pools as Cherenkov detectors for detecting cosmic-ray muons. Of these 960 photomultiplier tubes, 341 are recycled from the MACRO experiment. A systematic program was undertaken to refurbish them as waterproof assemblies. In the context of passing the water leakage check, a success rate better than 97% was achieved. Details of the design, fabrication, testing, operation, and performance of these waterproofed photomultiplier-tube assemblies are presented.Comment: 16 pages, 11 figures. Submitted to Nucl. Instr.
