    Super Denoise Net: Speech Super Resolution with Noise Cancellation in Low Sampling Rate Noisy Environments

    Speech super-resolution (SSR) aims to predict a high resolution (HR) speech signal from its low resolution (LR) corresponding part. Most neural SSR models focus on producing the final result in a noise-free environment by recovering the spectrogram of high-frequency part of the signal and concatenating it with the original low-frequency part. Although these methods achieve high accuracy, they become less effective when facing the real-world scenario, where unavoidable noise is present. To address this problem, we propose a Super Denoise Net (SDNet), a neural network for a joint task of super-resolution and noise reduction from a low sampling rate signal. To that end, we design gated convolution and lattice convolution blocks to enhance the repair capability and capture information in the time-frequency axis, respectively. The experiments show our method outperforms baseline speech denoising and SSR models on DNS 2020 no-reverb test set with higher objective and subjective scores

    A Novel Two Mode-Acting Inhibitor of ABCG2-Mediated Multidrug Transport and Resistance in Cancer Chemotherapy

    Multidrug resistance (MDR) is a major problem in successful treatment of cancers. Human ABCG2, a member of the ATP-binding cassette transporter superfamily, plays a key role in MDR and an important role in protecting cancer stem cells. Knockout of ABCG2 had no apparent adverse effect on the mice. Thus, ABCG2 is an ideal target for development of chemo-sensitizing agents for better treatment of drug resistant cancers and helping eradicate cancer stem cells.Using rational screening of representatives from a chemical compound library, we found a novel inhibitor of ABCG2, PZ-39 (N-(4-chlorophenyl)-2-[(6-{[4,6-di(4-morpholinyl)-1,3,5-triazin-2-yl]amino}-1,3-benzothiazol-2-yl)sulfanyl]acetamide), that has two modes of actions by inhibiting ABCG2 activity and by accelerating its lysosome-dependent degradation. PZ-39 has no effect on ABCB1 and ABCC1-mediated drug efflux, resistance, and their expression, indicating that it may be specific to ABCG2. Analyses of its analogue compounds showed that the pharmacophore of PZ-39 is benzothiazole linked to a triazine ring backbone.Unlike any previously known ABCG2 transporter inhibitors, PZ-39 has a novel two-mode action by inhibiting ABCG2 activity, an acute effect, and by accelerating lysosome-dependent degradation, a chronic effect. PZ-39 is potentially a valuable probe for structure-function studies of ABCG2 and a lead compound for developing therapeutics targeting ABCG2-mediated MDR in combinational cancer chemotherapy

    Design and optimization analysis of a new double-layer tube type heat exchanger for lead-bismuth reactors

    Double-layer heat tubes have been designed to effectively reduce the occurrence of heat pipe rupture accidents. However, inter-tube thermal contact resistance can decrease heat transfer efficiency, thus hampering the heat dissipation in the primary loop system of lead-bismuth reactors. Therefore, optimizing the design of double-layer heat tubes is necessary. This work focuses on the double-layer heat exchanger of a lead-bismuth reactor and utilizes gallium-based graphene nanofluids as a thermal interface material to fill the gap between the heat tubes. Furthermore, the impact of the length, wall thickness, outer diameter, and spacing of heat tubes on the heat transfer performance of the double-layer heat exchanger with and without the nanofluids has been analyzed. The study aims to optimize the JF factor and cost-effectiveness ratio (CER). Genetic algorithms are employed to optimize and evaluate the heat transfer performance of the main heat exchanger based on the four aforementioned parameters. Consequently, a new design scheme is obtained for the double-layer heat exchanger, which increases the optimized overall heat transfer coefficient of the main heat exchanger by 5.79%, pressure drop in the primary loop by 2.32%, JF factor by 5%, and CER by 24.62%. These results demonstrate that the gallium-based graphene nanofluids can effectively enhance the heat transfer performance of the double-layer heat exchanger while reducing the likelihood of steam generator tube rupture accidents

    A role for the arginine methylation of Rad9 in checkpoint control and cellular sensitivity to DNA damage

    The genome stability is maintained by coordinated action of DNA repairs and checkpoints, which delay progression through the cell cycle in response to DNA damage. Rad9 is conserved from yeast to human and functions in cell cycle checkpoint controls. Here, a regulatory mechanism for Rad9 function is reported. In this study Rad9 has been found to interact with and be methylated by protein arginine methyltransferase 5 (PRMT5). Arginine methylation of Rad9 plays a critical role in S/M and G2/M cell cycle checkpoints. The activation of the Rad9 downstream checkpoint effector Chk1 is impaired in cells only expressing a mutant Rad9 that cannot be methylated. Additionally, Rad9 methylation is also required for cellular resistance to DNA damaging stresses. In summary, we uncovered that arginine methylation is important for regulation of Rad9 function, and thus is a major element for maintaining genome integrity

    Reliability Analysis of Hydraulic System of a Tunnel-Erecting Machine Based on Dynamic Fault Tree and Bayesian Network

    Prefabricated utility tunnels play an important role in modern urban infrastructure construction. However, prefabricated utility tunnel segments are heavy, and the hoisting conditions are complicated, resulting in increased requirements in terms of the reliability of the equipment used for the erection and paving of utility tunnels, especially the hydraulic system of tunnel-erecting machines. Therefore, in this study, we performed reliability analysis of the hydraulic system of a tunnel-erecting machine. First, the working principle of the tunnel-erecting machine and its hydraulic system is analyzed, and a Takagi-Sugeno (T-S) dynamic fault tree model is constructed using the T-S dynamic fault tree analysis method, which is further transformed into a Bayesian network (BN) model. Secondly, according to the failure probability of the root node, combined with the BN conditional probability table (CPT), the failure probability of the leaf nodes of the hydraulic system of the tunnel-erecting machine in each time period and task time is forwardly inferred. Then, through the quantitative analysis of the sensitivity parameters in the BN analysis method, the importance of the components in the system can be reflected. Finally, the posterior probability of failure of the root node of the hydraulic system is calculated through the reverse reasoning of the BN analysis method, and the sensitive components of the system are identified. The results show that the proposed method can determine the main factors affecting the hydraulic system of a tunnel-erecting machine and provide reference for the safe operation of such equipment, as well as system maintenance
