    Anticancer effect of a combination of cisplatin and matrine on cervical cancer U14 cells and U14 tumor-bearing mice, and possible mechanism of action involved

    Purpose: To investigate the anticancer effects of cisplatin (DDP) in combination with matrine on cervical cancer U14 cell tumor-bearing mice. Methods: The cell proliferation of cervical cancer U14 cells treated with DDP (25, 20, 15, 10 and 5 μg/mL); matrine (2.5, 2.0, 1.5, 1.0 and 0.5 mg/mL); or DDP (15 μg/mL) + matrine (2.5, 2.0, 1.5, 1.0 and 0.5 mg/mL) was determined with MTT assay. The anticancer effect of DDP + matrine in U14 tumor-bearing mice was also investigated, based on expression of tumor suppressor lung cancer 1 (TSLC1) using quantitative real time-polymerase chain reaction (qRT-PCR) and immunohistochemistry. Results: The inhibition of proliferation of U14 cells ranged from 26.68–70.25, 10.20–61.73, and 51.89–89.75 % for DDP, matrine and DDP + matrine, respectively. In mice with U14 solid tumors, the DDP group had 12.3 % weight loss (p < 0.05). Treatment with DDP, matrine, and DDP + matrine reduced tumor growth by 64.56, 42.22–56.67, and 67.78–81.11 %, respectively (p < 0.01). Results from RT-qPCR and immunohistochemistry showed corresponding increases in expression levels of TSLC1. Conclusion: These results indicate that the anticancer activity of DDP + matrine is higher than that of a single treatment with either DPP or matrine. The likely mechanism of action might be related to promotion of TSLC1 expression. This finding provides a potential strategy for the management of cervical cancer

    Disinfection efficacy of green synthesized gold nanoparticles for medical disinfection applications

    Background: In recent times, biosyntheses of metal nanoparticles were used for several life rescue applications. In this study, Dillenia indica leaf aqueous extract was utilized for the synthesis of gold nanoparticles.Objective: To test anti-microbial properties of biologically fabricated gold nanoparticles.Methods: Gold nanoparticles were efficiently prepared by making use of aqueous leaf extract of Dillenia indica. The excitation of formed AuNPs was confirmed using UV–Vis spectrophotometer. In particular, absorption spectra of AuNPs exhibited a well-defined SPR band centered at around 530 nm.Results: The high-resolution Scanning Electron Microscope (SEM) results of the obtained AuNPs confirmed the formation of particles with a size range of 5–50 nm. The ultra-high resolution TEM (UHRTEM) images displayed clear lattice fringes on the particle surfaces. Single crystalline nature of the biosynthesized AuNPs was represented by means of selected-area electron diffraction pattern.Conclusion: The antibacterial activity of AuNPs revealed significant activity towards both gram negative and gram positive bacteria signifying their potential disinfection related applications in medicine and biology.Keywords: Dillenia indica leaves, disinfection, AuNPs

    Electric power network fractal and its relationship with power system fault

    Mreža elektro-energetskog sustava ima karakteristike fraktala. Ima osnovnu značajku složene mreže: sličnost samoj sebi. Autori su najprije izračunali vrijednosti fraktalne dimenzije za nekoliko elektro-energetskih rešetki, uključujući sustav 9 sabirnica WSCC (Western Systems Coordinating Council), sustav 14 sabirnica IEEE, sustav 30 sabirnica IEEE, sustav 39 sabirnica IEEE, sustav 118 sabirnica IEEE i sustav 300 sabirnica IEEE, kao i nekoliko stvarnih energetskih rešetki kao što su 500 kV glavne China Southern Power Grid (CSPG), 500 kV glavne energetske rešetke provincije Guangdong te 500 kV i 220 kV miješane energetske rešetke provincije Guangdong, itd. Na temelju fraktalne vrijednosti energetske rešetke analizirana je usporedba i odnos između fraktalne vrijednosti i intenziteta kvarenja energetskog sustava. Osnovni je zaključak da će za isti nivo napona, što je veća skala, energetska rešetka vjerojatno imati veću fraktalnu vrijednost i veći intenzitet kvarenja. Kod iste skale, što je gušća energetska rešetka to će vjerojatno biti veća fraktalna vrijednost i veći intenzitet kvarenja rešetke. Zaključci pružaju novi uvid u prosudbu statusa osjetljivosti energetskog sustava s interdisciplinarnog gledišta te voditi do novih smjerova u istraživanju.Electric power system network is with fractal characteristic. It has the basic feature of a complex network: self-similarity. The authors first calculated the fractal dimension values for several electric power grids, including WSCC (Western Systems Coordinating Council) 9 bus system, IEEE14 bus system, IEEE 30 bus system, IEEE 39 bus system, IEEE 118 bus system and IEEE 300 bus system; as well as some real power grids such as China Southern Power Grid (CSPG) 500 kV main power grid, Guangdong province 500 kV main power grid, and Guangdong province 500 kV and 220 kV mixed power grid, etc. Based on the power grid fractal value, a comparison and relationship between the fractal value and power system failure rate is analysed. The basic conclusion is that for the same voltage level, the larger the scale is, the larger fractal value and higher failure rate the power grid will possibly have. For the same scale, the denser the power grid, the larger fractal value and higher failure rate the power grid will probably have. The conclusions provide a new vision on the power system vulnerability status judgment from an interdisciplinary view and lead to a new research direction

    Plant-Mediated RNAi for Controlling Apolygus lucorum

    The polyphagous mirid bug Apolygus lucorum (Heteroptera: Miridae) is a serious pest of agricultural crops in China, with more than 200 species of host plants including two very important crops, maize and soybean. Currently, prevention and control of A. lucorum rely mainly on chemical pesticides that cause environmental as well as health related problems. Plant-mediated RNAi has proven to offer great potential for pest control in the past decade. In this study, we screened and obtained seven candidate genes (Alucβ-actin, AlucV-ATPase-A/D/E, AlucEif5A, AlucEcR-A, AlucIAP) by injection-based RNAi which produced A. lucorum mortality rates of 46.01–82.32% at day 7 after injection. Among them, the plant-mediated RNAi of AlucV-ATPase-E was successfully introduced into transgenic maize and soybean, and the populations of A. lucorum were significantly decreased following feeding on the transgenic maize and soybean. These results are intended to provide helpful insight into the generation of bug-resistant plants through a plant-mediated RNAi strategy

    Incorporation and solidification mechanism of manganese doped cement clinker

    Using municipal and industrial solid waste as a substitute raw material and fuel in cement rotary kiln co-processing is considered an economic and environmentally friendly alternative to the use of traditional fuels. However, the presence of heavy metals in solid waste is a growing concern in the cement rotary kiln co-processing technique. The solidification mechanism of heavy metals in cement clinker is directly related to their stabilization. Cement clinkers doped with manganese oxide (MnO2: 0.0%–5.0% wt%) were prepared in a laboratory to investigate the impacts of extrinsic Mn on cement clinker calcination. The insignificant changes in X-ray diffractometer patterns indicated that the fixed Mn had little influence on the mineral lattice structure. Raman spectra and X-ray photoelectron spectroscopy revealed the transformation of the silicate phase when the Mn dose was increased. Moreover, the satisfactory solidification ratio confirmed the incorporation of Mn in the cement clinker. These results provided evidence of the influence rule of Mn in the cement clinker calcination process. Furthermore, Raman spectroscopy showed great potential for the qualitative and semi-quantitative analysis of the cementitious materials derived from cement rotary kiln co-processing. These results will be important for the further development of green cement manufacturing technology

    New insight into the phylogeographic pattern of Liriodendron chinense (Magnoliaceae) revealed by chloroplast DNA: east–west lineage split and genetic mixture within western subtropical China

    Background Subtropical China is a global center of biodiversity and one of the most important refugia worldwide. Mountains play an important role in conserving the genetic resources of species. Liriodendron chinense is a Tertiary relict tree largely endemic to subtropical China. In this study, we aimed to achieve a better understanding of the phylogeographical pattern of L. chinense and to explore the role of mountains in the conservation of L. chinense genetic resources. Methods Three chloroplast regions (psbJ-petA, rpl32-ndhF, and trnK5’-matK) were sequenced in 40 populations of L. chinense for phylogeographical analyses. Relationships among chloroplast DNA (cpDNA) haplotypes were determined using median-joining networks, and genetic structure was examined by spatial analysis of molecular variance (SAMOVA). The ancestral area of the species was reconstructed using the Bayesian binary Markov Chain Monte Carlo (BBM) method according to its geographic distribution and a maximum parsimony (MP) tree based on Bayesian methods. Results Obvious phylogeographic structure was found in L. chinense. SAMOVA revealed seven groups matching the major landscape features of the L. chinense distribution area. The haplotype network showed three clades distributed in the eastern, southwestern, and northwestern regions. Separate northern and southern refugia were found in the Wu Mountains and Yungui Plateau, with genetic admixture in the Dalou Mountains and Wuling Mountains. BBM revealed a more ancient origin of L. chinense in the eastern region, with a west–east split most likely having occurred during the Mindel glacial stage. Discussion The clear geographical distributions of haplotypes suggested multiple mountainous refugia of L. chinense. The east–west lineage split was most likely a process of gradual genetic isolation and allopatric lineage divergence when the Nanling corridor was frequently occupied by evergreen or coniferous forest during Late Quaternary oscillations. Hotspots of haplotype diversity in the Dalou Mountains and Wuling Mountains likely benefited from gene flow from the Wu Mountains and Yungui Plateau. Collectively, these results indicate that mountain regions should be the main units for conserving and collecting genetic resources of L. chinense and other similar species in subtropical China

    Detection and differentiation of Borrelia burgdorferi sensu lato in ticks collected from sheep and cattle in China

    <p>Abstract</p> <p>Background</p> <p>Lyme disease caused by <it>Borrelia burgdorferi </it>sensu lato complex is an important endemic zoonosis whose distribution is closely related to the main ixodid tick vectors. In China, isolated cases of Lyme disease infection of humans have been reported in 29 provinces. Ticks, especially ixodid ticks are abundant and a wide arrange of <it>Borrelia </it>natural reservoirs are present. In this study, we developed a reverse line blot (RLB) to identify <it>Borrelia </it>spp. in ticks collected from sheep and cattle in 7 Provinces covering the main extensive livestock regions in China.</p> <p>Results</p> <p>Four species-specific RLB oligonucleotide probes were deduced from the spacer region between the 5S-23S rRNA gene, along with an oligonucleotide probe which was common to all. The species specific probes were shown to discriminate between four genomic groups of <it>B. burgdorferi </it>sensu lato i.e. <it>B. burgdorferi </it>sensu stricto, <it>B. garinii, B. afzelii</it>, and <it>B. valaisiana</it>, and to bind only to their respective target sequences, with no cross reaction to non target DNA. Furthermore, the RLB could detect between 0.1 pg and 1 pg of <it>Borrelia </it>DNA.</p> <p>A total of 723 tick samples (<it>Haemaphysalis, Boophilus, Rhipicephalus </it>and <it>Dermacentor</it>) from sheep and cattle were examined with RLB, and a subset of 667 corresponding samples were examined with PCR as a comparison. The overall infection rate detected with RLB was higher than that of the PCR test.</p> <p>The infection rate of <it>B. burgdoreri </it>sensu stricto was 40% in south areas; while the <it>B. garinii infection rate </it>was 40% in north areas. The highest detection rates of <it>B. afzelii </it>and <it>B. valaisiana </it>were 28% and 22%, respectively. Mixed infections were also found in 7% of the ticks analyzed, mainly in the North. The proportion of <it>B. garinii </it>genotype in ticks was overall highest at 34% in the whole investigation area.</p> <p>Conclusion</p> <p>In this study, the RLB assay was used to detect <it>B. burgdorferi </it>sensu lato in ticks collected from sheep and cattle in China. The results showed that <it>B. burdorferi senso stricto </it>and <it>B. afzelii </it>were mainly distributed in the South; while <it>B. garinii </it>and <it>B. valaisiana </it>were dominant in the North. <it>Borrelia </it>spirochaetes were detected in <it>Rhipicephalus </it>spp for the first time. It is suggested that the <it>Rhipicephalus </it>spps might play a role in transmitting <it>Borrelia </it>spirochaetes.</p
