    Cell autonomous and non-autonomous functions of plant intracellular immune receptors in stomatal defense and apoplastic defense.

    Stomatal closure defense and apoplastic defense are two major immunity mechanisms restricting the entry and propagation of microbe pathogens in plants. Surprisingly, activation of plant intracellular immune receptor NLR genes, while enhancing whole plant disease resistance, was sometimes linked to a defective stomatal defense in autoimmune mutants. Here we report the use of high temperature and genetic chimera to investigate the inter-dependence of stomatal and apoplastic defenses in autoimmunity. High temperature inhibits both stomatal and apoplastic defenses in the wild type, suppresses constitutive apoplastic defense responses and rescues the deficiency of stomatal closure response in autoimmune mutants. Chimeric plants have been generated to activate NLR only in guard cells or the non-guard cells. NLR activation in guard cells inhibits stomatal closure defense response in a cell autonomous manner likely through repressing ABA responses. At the same time, it leads to increased whole plant resistance accompanied by a slight increase in apoplastic defense. In addition, NLR activation in both guard and non-guard cells affects stomatal aperture and water potential. This study thus reveals that NLR activation has a differential effect on immunity in a cell type specific matter, which adds another layer of immune regulation with spatial information

    Stigma in Elderly Females with Stress Urinary Incontinence: A Latent Profile Analysis

    Background: Stress urinary incontinence (SUI) is a commonly occurring urological disorder in females, particularly among the elderly population. Females with SUI often experience significant stigma associated with their condition. This study aimed to investigate the current status of stigma among elderly females with SUI and analyze its heterogeneous subtypes. Methods: The Stigma Scale for Chronic Illness (SSCI) was used to survey 245 participants in two tertiary hospitals in Guangdong from November 2021 to September 2022. Latent profile analysis was employed to create a classification model, and variance and correlation analyses were conducted to assess the influencing factors. Results: A total of 245 elderly females with SUI participated in the survey. They had an average stigma score of 83.70 ± 13.88, consisting of self-stigma (48.64 ± 8.04) and perceived stigma (35.06 ± 6.80) scores. Latent profile analysis identified three distinct and comparable subtypes: the low-self-low-perceived group (14.69%), the high-self-medium-perceived group (49.38%), and the high-self-high-perceived group (35.91%). These subtypes exhibited statistically significant differences in all dimensions and the overall stigma score (p < 0.05) and were found to be correlated with the patient’s level of education, marital status, drinking habits, number of chronic illnesses, presence of diabetes, and frequency of urinary leakage (p < 0.05). Conclusion: This study demonstrates that elderly females with SUI face elevated levels of stigma, and it reveals distinct classification characteristics among them. Additionally, it emphasizes the importance of providing specific support and attention to individuals with higher levels of education, increased fluid intake, marital status, severe urinary leakage, and diabetes

    Recent advances in the study of the CRISPR/Cas9 system have provided a precise and versatile approach for genome editing in various species. However, the applicability and efficiency of this method in large animal models, such as the goat, have not been extensively studied. Here, by co-injection of one-cell stage embryos with Cas9â €‰mRNA and sgRNAs targeting two functional genes (MSTN and FGF5), we successfully produced gene-modified goats with either one or both genes disrupted. The targeting efficiency of MSTN and FGF5 in cultured primary fibroblasts was as high as 60%, while the efficiency of disrupting MSTN and FGF5 in 98 tested animals was 15% and 21% respectively, and 10% for double gene modifications. The on-and off-target mutations of the target genes in fibroblasts, as well as in somatic tissues and testis of founder and dead animals, were carefully analyzed. The results showed that simultaneous editing of several sites was achieved in large animals, demonstrating that the CRISPR/Cas9 system has the potential to become a robust and efficient gene engineering tool in farm animals, and therefore will be critically important and applicable for breeding
