Artesunate Enhances the Cytotoxicity of 5-Aminolevulinic Acid-Based Sonodynamic Therapy against Mouse Mammary Tumor Cells In Vitro

Sonodynamic therapy (SDT) kills tumor cells through the synergistic effects of ultrasound (US) and a sonosensitizer agent. 5-Aminolevulinic acid (5-ALA) has been used as a sonodynamic sensitizer for cancer treatment. However, studies have shown that 5-ALA-based SDT has limited efficacy against malignant tumors. In this study, we examined whether artesunate (ART) could enhance the cytotoxicity of 5-ALA-based SDT against mouse mammary tumor (EMT-6) cells in vitro. In the ART, ART + US, ART + 5-ALA, and ART + 5-ALA + US groups, the cell survival rate correlated with ART concentration, and decreased with increasing concentrations of ART. Morphologically, many apoptotic and necrotic cells were observed in the ART + 5-ALA + US group. The percentage of reactive oxygen species-positive cells in the ART + 5-ALA + US group was also significantly higher than that in the 5-ALA group (p = 0.0228), and the cell death induced by ART + 5-ALA + US could be inhibited by the antioxidant N-acetylcysteine. These results show that ART offers great potential in enhancing the efficacy of 5-ALA-based SDT for the treatment of cancer. However, these results are only based on in vitro studies, and further in vivo studies are required

Acute effect of dynamic stretching or running on endurance running performance in well-trained male runners

BACKGROUND: The purpose of this study was to compare the acute effects of dynamic stretching and running on relative high-intensity endurance running performance in well-trained male runners. METHODS: The endurance running performances of 16 well-trained long-distance male runners were assessed on a treadmill at about 5 minutes after 2 types of intervention. The interventions were running intervention and dynamic stretching intervention. In the running intervention, each participant ran on the treadmill at a velocity equivalent to his 70% maximal oxygen uptake (V̇O2max) in each participant for 15 minutes. In the dynamic stretching intervention, dynamic stretching was performed for 10 repetitions as quickly as possible for the 5 muscle groups of the lower extremities for 3 minutes and 40±9 seconds. Endurance running performance was evaluated by time to exhaustion during running at a velocity equivalent to 90% V̇O2max in each participant. RESULTS: The time to exhaustion (870.7±237.6 seconds) after dynamic stretching intervention was significantly (d=0.75, P=0.01) prolonged compared with that (745.4±225.1 seconds) after running intervention. CONCLUSIONS: The results demonstrated that the dynamic stretching intervention improved the endurance performance of running at a velocity equivalent to 90% V̇O2max in well-trained male runners. Our findings suggested that performing dynamic stretching was effective for improving performance compared with performing only running during the warm-up

Purpose: The purpose of this study was to compare the acute effects of general warm-up (GWU) and GWU plus dynamic stretching (GWU + DS) on endurance running performance in well-trained male runners. Method: The endurance running performances of eight well-trained long-distance male runners were assessed on a treadmill after 2 types of intervention for 5 min after running on the treadmill at a velocity equivalent to 70% maximal oxygen uptake (VO_2max) in each athlete for 15 min. The interventions were GWU and GWU + DS. In the GWU + DS intervention, dynamic stretching was performed for ten repetitions as quickly as possible for the five muscle groups of the lower extremities. The total duration of the dynamic stretching was 3 min and 45 s. Endurance running performance was assessed at 1 min 15 s after the dynamic stretching. The endurance running performance was evaluated by the time to exhaustion (TTE) during running at a velocity equivalent to 90% V˙O_2max in each athlete. Results: The TTE (640.6 ± 220.4 s) after GWU + DS intervention was significantly (d = 1.02, p = .03) shorter than that (760.6 ± 249.1 s) after GWU intervention. Conclusions: The results demonstrated that GWU + DS intervention impaired immediate endurance performance of running at a velocity equivalent to 90% V˙O_2max in well-trained male runners compared with GWU intervention. Thus, we are not able to recommend that well-trained runners and their coaches use the protocol for GWU + DS described in this study during actual warm-ups

A Case of Pneumothorax after Treatment with Lenvatinib for Anaplastic Thyroid Cancer with Lung Metastasis

A 63-year-old man was diagnosed with multiple lung metastases from anaplastic thyroid cancer and received lenvatinib. Follow-up computed tomography on day 34 of lenvatinib treatment showed pneumothorax. The pneumothorax was temporarily improved with chest drainage. However, pleurodesis was performed to treat a relapse of the pneumothorax. Pneumothorax during chemotherapy for a malignant tumor is considered a relatively rare complication. This case is the first documentation that pneumothorax may develop during lenvatinib treatment. The possible development of pneumothorax should be considered when lenvatinib is used in patients with lung metastasis

Nephrectomy for Metastatic Kidney Tumor in Patients with Differentiated Thyroid Cancer: A Report of Two Cases

    No full text
The occurrence of renal tumors originating from thyroid cancer is extremely rare with a few effective treatments for renal metastases. Here, we report the cases of two patients with differentiated thyroid cancer who underwent nephrectomy for a metastatic kidney tumor. Case 1 was a 74-year-old man who was diagnosed with right kidney tumor 10 years after initial surgery for papillary thyroid cancer (PTC). Right nephrectomy was performed, and the pathology was metastatic PTC. Case 2 was a 68-year-old woman who was diagnosed with left kidney tumor 24 years after surgery for follicular thyroid carcinoma (FTC). Left nephrectomy was performed, and the pathology was metastatic FTC. Nephrectomy for single renal metastasis could be considered a treatment option if the patients’ general condition is positive
