    An ultra-sensitive and easy-to-use assay for sensing human UGT1A1 activities in biological systems

    The human UDP-glucuronosyltransferase 1A1 (UGT1A1), one of the most essential conjugative enzymes, is responsible for the metabolism and detoxification of bilirubin and other endogenous substances, as well as many different xenobiotic compounds. Deciphering UGT1A1 relevance to human diseases and characterizing the effects of small molecules on the activities of UGT1A1 requires reliable tools for probing the function of this key enzyme in complex biological matrices. Herein, an easy-to-use assay for highly-selective and sensitive monitoring of UGT1A1 activities in various biological matrices, using liquid chromatography with fluorescence detection (LC-FD), has been developed and validated. The newly developed LC-FD based assay has been confirmed in terms of sensitivity, specificity, precision, quantitative linear range and stability. One of its main advantages is lowering the limits of detection and quantification by about 100-fold in comparison to the previous assay that used the same probe substrate, enabling reliable quantification of lower amounts of active enzyme than any other method. The precision test demonstrated that both intra- and inter-day variations for this assay were less than 5.5%. Furthermore, the newly developed assay has also been successfully used to screen and characterize the regulatory effects of small molecules on the expression level of UGT1A1 in living cells. Overall, an easy-to-use LC-FD based assay has been developed for ultra-sensitive UGT1A1 activities measurements in various biological systems, providing an inexpensive and practical approach for exploring the role of UGT1A1 in human diseases, interactions with xenobiotics, and characterization modulatory effects of small molecules on this conjugative enzyme.     Th2 cytokine bias induced by silver nanoparticles in peripheral blood mononuclear cells of common bottlenose dolphins (Tursiops truncatus)

    Background Silver nanoparticles (AgNPs) have been widely used in many commercial products due to their excellent antibacterial ability. The AgNPs are released into the environment, gradually accumulate in the ocean, and may affect animals at high trophic levels, such as cetaceans and humans, via the food chain. Hence, the negative health impacts caused by AgNPs in cetaceans are of concern. Cytokines play a major role in the modulation of immune system and can be classified into two types: Th1 and Th2. Th1/Th2 balance can be evaluated by the ratios of their polarizing cytokines (i.e., interferon [IFN]-Îł/Interleukin [IL]-4), and animals with imbalanced Th1/Th2 response may become more susceptible to certain kinds of infection. Therefore, the present study evaluated the in vitro cytokine responses of cetacean peripheral blood mononuclear cells (cPBMCs) to 20 nm citrate-AgNPs (C-AgNP20) by quantitative reverse transcriptase polymerase chain reaction (qRT-PCR). Methods Blood samples were collected from six captive common bottlenose dolphins (Tursiops truncatus). The cPBMCs were isolated and utilized for evaluating the in vitro cytokine responses. The cytokines evaluated included IL-2, IL-4, IL-10, IL-12, interferon (IFN)-Îł, and tumor necrosis factor (TNF)-α. The geometric means of two housekeeping genes (HKGs), glyceraldehyde 3-phosphate dehydrogenase (GAPDH) and ÎČ2-microglobulin (B2M), of each sample were determined and used to normalize the mRNA expression levels of target genes. Results The ratio of late apoptotic/necrotic cells of cPBMCs significantly increased with or without concanavalin A (ConA) stimulation after 24 h of 10 ”g/ml C-AgNP20 treatment. At 4 h of culture, the mRNA expression level of IL-10 was significantly decreased with 1 ”g/ml C-AgNP20 treatment. At 24 h of culture with 1 ”g/ml C-AgNP20, the mRNA expression levels of all cytokines were significantly decreased, with the exceptions of IL-4 and IL-10. The IFN-Îł/IL-4 ratio was significantly decreased at 24 h of culture with 1 ”g/ml C-AgNP20 treatment, and the IL-12/IL-4 ratio was significantly decreased at 4 or 24 h of culture with 0.1 or 1 ”g/ml C-AgNP20 treatment, respectively. Furthermore, the mRNA expression level of TNF-α was significantly decreased by 1 ”g/ml C-AgNP20 after 24 h of culture. Discussion The present study demonstrated that the sublethal dose of C-AgNP20 (≀1 ”g/ml) had an inhibitory effect on the cytokine mRNA expression levels of cPBMCs with the evidence of Th2 cytokine bias and significantly decreased the mRNA expression level of TNF-α. Th2 cytokine bias is associated with enhanced immunity against parasites but decreased immunity to intracellular microorganisms. TNF-α is a contributing factor for the inflammatory response against the infection of intracellular pathogens. In summary, our data indicate that C-AgNP20 suppresses the cellular immune response and thereby increases the susceptibility of cetaceans to infection by intracellular microorganisms

    Plasma Interleukin-37 is elevated in patients with rheumatoid arthritis: Its correlation with disease activity and Th1/Th2/Th17-related cytokines

    Interleukin- (IL-) 37 is a novel anti-inflammatory cytokine that suppresses immune response and inflammation. This study was performed to determine whether IL-37 was elevated in patients with rheumatoid arthritis (RA) and investigate the correlation between IL-37 level and disease activity and the concentration of Th1/Th2/Th17-related cytokines. Clinical parameters of disease activity, including the 28-joint disease activity score (DAS28) and C-reactive protein (CRP), were collected in 34 RA patients and 34 age- and sex-matched healthy controls. Plasma IL-37 was measured by ELISA. Plasma levels of TNF-α, IL-1ÎČ, IL-2, IL-4, IL-5, IL-6, IL-7, IL-8, IL-10, IL-12, IL-13, IL-17, G-CSF, GM-CSF, IFN-Îł, MCP-1, and MIP-1ÎČ were analyzed using the Bio-Plex suspension array system. It was found that IL-37 levels were elevated markedly in RA patients and almost undetectable in healthy controls. In addition, IL-37 levels in patients with active RA were significantly enhanced as compared with those in patients of remission. More importantly, IL-37 showed a significant correlation with disease activity (DAS28) and IL-4, IL-7, IL-10, IL-12, and IL-13 concentrations in RA patients. These findings suggest that IL-37 plays an important role in the pathogenesis of RA and may prove to be a potential biomarker of active RA

    A simulation study on the measurement of D0-D0bar mixing parameter y at BES-III

    We established a method on measuring the \dzdzb mixing parameter yy for BESIII experiment at the BEPCII e+e−e^+e^- collider. In this method, the doubly tagged ψ(3770)→D0D0‟\psi(3770) \to D^0 \overline{D^0} events, with one DD decays to CP-eigenstates and the other DD decays semileptonically, are used to reconstruct the signals. Since this analysis requires good e/πe/\pi separation, a likelihood approach, which combines the dE/dxdE/dx, time of flight and the electromagnetic shower detectors information, is used for particle identification. We estimate the sensitivity of the measurement of yy to be 0.007 based on a 20fb−120fb^{-1} fully simulated MC sample.Comment: 6 pages, 7 figure

    Proactive Personality and Career Adaptability of Chinese Female Pre-Service Teachers in Primary Schools: The Role of Calling

    Career adaptability has attracted extensive attention from international scholars as one of the important factors predicting the future sustainability of individuals. Proactive personality and calling have a positive influence on individual career development and life satisfaction, with reference to promoting the sustainable development of individuals. This study focuses on the positive effects of career adaptability and calling on shaping female primary school pre-service teachers and provides suggestions for training excellent elementary school teachers and building quality primary education. This study investigated the relationship between proactive personality, calling, and career adaptability among 707 female pre-service elementary school teachers in Chinese universities. The results showed that proactive personality positively predicts calling and that proactive personality and calling positively predict career adaptability. Calling partially mediated the relationship between proactive personality and career adaptability. Therefore, it is possible to enhance calling by cultivating the proactive personality of female pre-service teachers in elementary schools, thus further promoting the career adaptability of this group. On this basis, effective strategies should be adopted in the future to improve the career adaptability of female pre-service teachers in elementary schools for the sustainable development of this group

    Charge-state effects on charge transfer of helium ions in gold nanosheet

    We investigate the charge-state effects on charge transfer of helium ions in gold nanosheet using the time-dependent density functional theory non-adiabatically coupled to the molecular dynamics. In order to characterize and extract the charge-state information of incident particles inside the nanosheet, we develop two novel computational methods. It is found that the charge transfer behavior of He ion in gold nanosheet is sensitive to its charge state at the time of incident. Analysis of these results allows us to gain new insights in the interaction between He ions and gold nanosheet. This work validates the ability of current methodology in dealing with ion collisions in materials

    Landfill leachate treatment by coagulation/flocculation combined with microelectrolysis-Fenton processes

    <p>Landfill leachate was pretreated by chemical flocculation with polyaluminum chloride (PAC) as a flocculant, and subsequently purified by the microelectrolysis-Fenton (MEF) process. Response surface methodology was employed to optimize the MEF process, and the optimal conditions were initial pH 3.20, H<sub>2</sub>O<sub>2</sub> concentration 3.57 g/L, and Fe-C dosage 104.52 g/L. The PAC coagulation combined with MEF processes obtained a superior decontamination performance, and the predicted chemical oxygen demand (COD) and humic acids (HA) removal were respectively 90.27% and 93.79%. The strong fluorescence peak at 425 nm and the trapping experiment showed that <math><mi></mi><mo>⋅</mo>OH</math> was generated during MEF, which had a strong oxidation ability to degrade organic recalcitrant pollutants. The ultraviolet–visible spectra and three-dimensional excitation–emission matrices spectra (3D-EEMs) indicated that PAC coagulation could preferentially remove protein-like substances, while the MEF process was effective in destructing organic recalcitrant pollutants, especially humic-like and fulvic-like substances.</p
