    Assessing the quality of ionogram interpretation using the HF Doppler technique

    International audienceThe first joint common volume measurements by the Digisonde Portable Sounder (DPS-4) and a new Doppler type system has been run at the Pruhonice ionospheric observatory (49.99° N, 14.54° E) since January 2004. The measurement of the Doppler shift is carried out continuously on a frequency of 3.6 MHz, thus the radio wave is reflected predominantly from the ionospheric F layer. To compare digisonde measurements with the Doppler data, a phase path was calculated from both Doppler and digisonde records. Under stormy conditions and in the case where a sporadic E layer was present, a significant disagreement between both measurements has been found. The discrepancies could be related to the uncertainties of the observational inputs and to the interpretation of the digisonde data. The comparison of the phase paths shows that during geomagnetically quiet days, in the absence of the sporadic E layer, and when high quality ionograms are available and correctly scaled, the electron density N(h) profiles, calculated by the Automatic Real Time Ionogram Scaler with True height algorithm (ARTIST), can be considered reliable

    Rapidly developing sequencing technologies have provided the possibility for identification of a DNA nucleotide sequence of a whole individual human genome just in a couple of weeks. Working capacity of some sequencers is already measured in thousands of milliards of base pairs per an operating cycle. Reviewed have been the basic principles and analytical potential of the modern methods for DNA sequencing which are nominally subdivided into three major types: classical - capillary electrophoresis sequencing and pyrosequencing, novel (Next Generation Sequencing - NGS) - simultaneous sequencing of millions of DNA fragments, each one of which is a cluster containing many thousands or hundreds of thousands of their clones - high-performance pyrosequencing, cyclic ligase and semiconducting sequencing, molecular-cluster-based sequencing using fluorescent-labeled precursors; and cutting-edge methods - (Next-Next Generation Sequencing - NNGS) - the ones that read millions of single DNA fragments without pre-cloning

    Phenotypic and Genetic Analysis of Altered Variants of <i>Vibrio cholerae</i> Biovar El Tor

    Determined is cholera toxin production by altered Vibrio cholerae O1 eltor strains, isolated on the territory of Russia in 1993-2010, which contain classical type ctxB gene. Studied are their genetic characteristics related to the production of this virulence factor. Determined is that altered eltor variants yield 10 to 20 times more cholera toxin than typical eltor strains. Changes in promotor region of ctxAB operon are not detected but elucidated is that 90 % of studied altered strains contain two copies of CTXϕ prophage in their genome whereas typical strains carry one copy of CTXϕ prophage. Despite the higher copy number of CTXϕ prophage, the identified differences in production of cholera toxin can be associated mostly with a change of transcriptional activity of regulatory genes. That causes the active production of cholera toxin by the altered variants of Vibrio cholerae eltor in non typical environment

Determined is a significant genetic diversity of the gene-variants, probably, due to their polyclonal origin and continuous alterations within genome under the influence of varying environmental factors

    Comparative Analysis of the Sequenced Zaire ebolavirus Samples from the Republic of Guinea

    An unprecedented on its scale Ebola fever epidemic, caused by Zaire ebolavirus, has been registered in the territory of West Africa in 2014-2015. The virus quickly spread from the Republic of Guinea into the neighboring countries. Presented are the results of comparative analysis between three Zaire ebolavirus genomes, the complementary DNA of which was obtained from the samples from Ebola fever patients in the Republic of Guinea in 2014. The nucleotide sequences of the genomes received have allowed for identification of phylogenetic affinity to certain references, displayed in the database of NCBI GenBank. General phylogenetic analysis by means of representative sampling has revealed two trends of genome evolutionary development from one and the same ancestral form. The first branch of evolution is represented by a relatively small number of Zaire ebolavirus isolates, disseminated in Guinea. The second one, the most numerous trend, is generated by four variants of Zaire ebolavirus genome, which have been identified starting from May - early June, 2014 in Sierra-Leone, but within a short period of time they have won the whole territory of the country and the neighboring Guinea and Liberia
