Estudios efectivos de potencial para algunas nuevas moléculas híbridas para su actividad contra el cáncer de próstata
Objective: The present work aimed at developing novel hybrid molecules for targeting the prostate
cancer. It is observed that two human shock proteins Hsp70 and Hsp90 are over-expressed in prostate
cancer making them one of the important drug targets. We have designed and developed twelve new
hybrid molecules 6a-j for targeting these proteins.
Methods: The designed molecules were prepared following a four step reaction protocol and characterized
on the basis of proton NMR and Mass spectrometry. These were subjected to in vitro studies by
means of Oncotest and CCK-8 assays with two cell lines DU145 and 22Rv1. The selected molecules 6b
and 6i were subjected to molecular docking and then for SPR based affinity assay.
Results: Compounds 6b and 6i were found to be highly active anticancer compounds comparable to
standard drug enzalutamide. They have significant IC50 and high dock score for the Hsp70 and Hsp90.
These compounds are selective and have good binding affinity for the Hsp70 due to high Kd.
Conclusion: Compound 6b and 6i can serve as lead molecules for the development of antiprostate cancer
drugs with Hsp70 as target.Objetivo: El presente trabajo tuvo como objetivo desarrollar nuevas moléculas híbridas para atacar el
cáncer de próstata. Se observa que dos proteínas de choque humano, Hsp70 y Hsp90, se sobreexpresan
en el cáncer de próstata, lo que las convierte en uno de los objetivos farmacológicos importantes. Hemos
diseñado y desarrollado doce nuevas moléculas híbridas 6a-j para dirigir estas proteínas.
Métodos: Las moléculas diseñadas se prepararon siguiendo un protocolo de reacción de cuatro etapas
y se caracterizaron sobre la base de RMN de protón y espectrometría de masas. Estos se sometieron a
estudios in vitro por medio de ensayos Oncotest y CCK-8 con dos líneas celulares DU145 y 22Rv1. Las
moléculas seleccionadas 6b y 6i se sometieron a acoplamiento molecular y luego a ensayo de afinidad
basado en SPR.
Resultados: Se descubrió que los Compuestos 6b y 6i son compuestos anticancerígenos muy activos
comparables al fármaco estándar enzalutamida. Tienen un IC50 significativo y una puntuación alta para
el muelle de Hsp70 y Hsp90. Estos compuestos son selectivos y tienen una buena afinidad de unión por
la Hsp70 debido a la alta Kd.
Conclusión: Los compuestos 6b y 6i pueden servir como moléculas principales para el desarrollo de
fármacos antiprostáticos contra el cáncer con Hsp70 como objetivo
REST APIs: A large-scale analysis of compliance with principles and best practices
Quickly and dominantly, REST APIs have spread over the Web and percolated into modern software development practice, especially in the Mobile Internet where they conveniently enable offloading data and computations onto cloud services. We analyze more than 78GB of HTTP traffic collected by Italy’s biggest Mobile Internet provider over one full day and study how big the trend is in practice, how it changed the traffic that is generated by applications, and how REST APIs are implemented in practice. The analysis provides insight into the compliance of state-of-the-art APIs with theoretical Web engineering principles and guidelines, knowledge that affects how applications should be developed to be scalable and robust. The perspective is that of the Mobile Internet
Design and Synthesis of Vitamin Drug Conjugate for its Probable Potential Against SARS-COV-2 Infections
The novel corona virus infection had become a global epidemic due to its rapid spread. So, there is an urgent need to treat COVID-19 patients. The aim of this research was to hypothesize and examine vitamin drug con-jugate as targeted moiety. The present scaffold may have potential role to fight against COVID-19 infection due to its antimicrobial, antioxidants and immunomodulatory activities. Here, we've highlighted the term Vit-amin Drug Conjugate as possible therapy approach for SARS-COV-2 infection. As a result, we synthesize, characterized, and evaluated a Hydroxychloroquine — Folic Acid conjugate (HCQ-FA) by esterification mechanism to provide effective treatment against SARS-CoV-2 infection by enhancing therapeutic effect through synergistic mechanism, masking undesired side effects, and improving cellular internalization. By us-ing prodrug, the efficacy and bioavailability of existing antiviral drugs could be improved. The structure of the conjugate was determined by spectroscopic data like IR, NMR, and mass spectra, which indicates that HCQ-FA conjugate formed by esteric conjugation. Molecular docking studies revealed that HCQ-FA conju-gate shows good level of docking as well as binding interaction with main protease moiety. Molecular dy-namic stimulation revealed that this conjugate shows good stability at the binding site of SARS main protease moiety and exhibits inhibitory activity against COVID-19 infectio
Mechanical Strain Stabilizes Reconstituted Collagen Fibrils against Enzymatic Degradation by Mammalian Collagenase Matrix Metalloproteinase 8 (MMP-8)
Collagen, a triple-helical, self-organizing protein, is the predominant structural protein in mammals. It is found in bone, ligament, tendon, cartilage, intervertebral disc, skin, blood vessel, and cornea. We have recently postulated that fibrillar collagens (and their complementary enzymes) comprise the basis of a smart structural system which appears to support the retention of molecules in fibrils which are under tensile mechanical strain. The theory suggests that the mechanisms which drive the preferential accumulation of collagen in loaded tissue operate at the molecular level and are not solely cell-driven. The concept reduces control of matrix morphology to an interaction between molecules and the most relevant, physical, and persistent signal: mechanical strain.The investigation was carried out in an environmentally-controlled microbioreactor in which reconstituted type I collagen micronetworks were gently strained between micropipettes. The strained micronetworks were exposed to active matrix metalloproteinase 8 (MMP-8) and relative degradation rates for loaded and unloaded fibrils were tracked simultaneously using label-free differential interference contrast (DIC) imaging. It was found that applied tensile mechanical strain significantly increased degradation time of loaded fibrils compared to unloaded, paired controls. In many cases, strained fibrils were detectable long after unstrained fibrils were degraded.In this investigation we demonstrate for the first time that applied mechanical strain preferentially preserves collagen fibrils in the presence of a physiologically-important mammalian enzyme: MMP-8. These results have the potential to contribute to our understanding of many collagen matrix phenomena including development, adaptation, remodeling and disease. Serum Uric Acid and Adiposity: Deciphering Causality Using a Bidirectional Mendelian Randomization Approach
Background: Although the relationship between serum uric acid (SUA) and adiposity is well established, the direction of the causality is still unclear in the presence of conflicting evidences. We used a bidirectional Mendelian randomization approach to explore the nature and direction of causality between SUA and adiposity in a population-based study of Caucasians aged 35 to 75 years. Methods and Findings: We used, as instrumental variables, rs6855911 within the SUA gene SLC2A9 in one direction, and combinations of SNPs within the adiposity genes FTO, MC4R and TMEM18 in the other direction. Adiposity markers included weight, body mass index, waist circumference and fat mass. We applied a two-stage least squares regression: a regression of SUA/adiposity markers on our instruments in the first stage and a regression of the response of interest on the fitted values from the first stage regression in the second stage. SUA explained by the SLC2A9 instrument was not associated to fat mass (regression coefficient [95 % confidence interval]: 0.05 [20.10, 0.19] for fat mass) contrasting with the ordinary least square estimate (0.37 [0.34, 0.40]). By contrast, fat mass explained by genetic variants of the FTO, MC4R and TMEM18 genes was positively and significantly associated to SUA (0.31 [0.01, 0.62]), similar to the ordinary least square estimate (0.27 [0.25, 0.29]). Numerous risk factors for the development of gout have been established, including hyperuricaemia, genetic factors, dietary factors, alcohol consumption, metabolic syndrome, hypertension, obesity, diuretic use and chronic renal disease.
