    HMGA1 variant IVS5-13insC is associated with insulin resistance and type 2 diabetes: an updated meta-analysis

    Background: High-mobility group A1 (HMGA1) polymorphism has been suspected as a gene variant associated with type 2 diabetes (T2D). However, conflicting outcomes have been reported. Objective: This meta-analysis aimed to predict the association between the HMGA1 variant IVS5-13insC and T2D. Methods: Statistical analyses were performed using Stata/SE 12.0 software. Results: A total of 11 case-control studies in 6 articles were included. Results suggested that the HMGA1 variant IVS5-13insC was associated with an increased risk of insulin resistance (OR = 0.61, 95% CI 0.56 to 0.66, P < 0.0001), T2D (OR = 0.67, 95% CI 0.61 to 0.73, P < 0.0001), particularly for Caucasians with increased risks of T2D (OR = 0.56, 95% CI 0.49 to 0.65, P < 0.0001) compared with wild-type subjects. Conclusion: This meta-analysis indicated that the HMGA1 variant IVS5-13insC can be a risk factor of T2D development, particularly among Caucasians. Significant risks were also found (Asian: OR = 0.74, 95% CI: 0.63 to 0.86, P < 0.0001, Hispanic-American: OR = 0.81, 95% CI: 0.65 to 1.01, P < 0.0001) in non-Caucasian population. However, ethnical studies should be conducted to reveal whether the HMGA1 variant IVS5-13insC is associated with an increased risk of T2D.Keywords: HMGA1, type 2 diabetes, insulin resistance, variant, meta-analysis

    The functions and clinical applications of tumor-derived exosomes

    Exosomes are extracellular vesicles with diameters ranging from 30 to 150 nm. They can be secreted by all cell types and transfer information in the form of their contents, which include proteins, lipids and nucleic acids, to other cells throughout the body. They have roles in normal physiological processes as well as in disease development. Here, we review recent findings regarding tumor-derived exosomes, including methods for their extraction and preservation. We also describe the actions of exosomes in tumorigenesis. The exosomal antigen-presenting effect during antitumor immune responses and its suppressive function in immune tolerance are discussed. Finally, we describe the potential application of exosomes to cancer therapy and liquid biopsy

    Laser-assisted synthesis of two-dimensional transition metal dichalcogenides: a mini review

    The atomically thin two-dimensional (2D) transition metal dichalcogenides (TMDCs) have attracted the researcher’s interest in the field of flexible electronics due to their high mobility, tunable bandgaps, and mechanical flexibility. As an emerging technique, laser-assisted direct writing has been used for the synthesis of TMDCs due to its extremely high preparation accuracy, rich light–matter interaction mechanism, dynamic properties, fast preparation speed, and minimal thermal effects. Currently, this technology has been focused on the synthesis of 2D graphene, while there are few literatures that summarize the progress in direct laser writing technology in the synthesis of 2D TMDCs. Therefore, in this mini-review, the synthetic strategies of applying laser to the fabrication of 2D TMDCs have been briefly summarized and discussed, which are divided into top-down and bottom-up methods. The detailed fabrication steps, main characteristics, and mechanism of both methods are discussed. Finally, prospects and further opportunities in the booming field of laser-assisted synthesis of 2D TMDCs are addressed

    Introduction: Oxytocin might be used therapeutically as an ally to rescue osteopathy resulting from diabetes. However, the in vivo effects of oxytocin on marrow adipogenesis in diabetes remain unknown. In this longitudinal study, we aimed to investigate the protective ef­fects of oxytocin on diabetes-induced marrow adiposity in rabbits using proton MR spectroscopy. Material and methods: Forty-five female New Zealand rabbits were randomly divided into controls, diabetes, and diabetes treated with oxytocin (ip, 0.78 mg/kg) for six months. Marrow fat fraction (FF) was determined by proton MR spectroscopy at baseline, and at three and six months. Bone mineral density was measured by dual-energy X-ray absorptiometry. Serum biomarkers, glycolipid metabolism, and histological analysis of marrow adipocytes were determined. Results: Oxytocin treatment had positive metabolic effects in diabetic rabbits, which was based on the changes in glucose metabolism, insulin sensitivity, and lipid profiles. The diabetic rabbits demonstrated dramatic marrow adiposity in a time-dependent manner; at three and six months the FF percentage changes from baseline were 10.1% and 25.8%, respectively (all P < 0.001). Moreover, oxytocin treatment significantly reversed FF values and quantitative parameters of marrow adipocyte in diabetic rabbits to levels of naive control rabbits. Oxytocin improved bone formation marker in diabetic rabbits compared to the saline group. Also, treatment of diabetic rabbits with oxytocin significantly mitigated bone deterioration when compared with the saline-treated diabetic group (all P < 0.05). Conclusions: Oxytocin appears to alleviate harmful effects of hyperglycaemia on marrow adiposity. Proton MR spectroscopy may be a valuable tool, providing complementary information on efficacy assessments.Wstęp: Oksytocyna może być stosowana terapeutycznie w osteopatii wynikającej z cukrzycy, jednakże jej wpływ in vivo na stłuszczenie szpiku kostnego w przebiegu cukrzycy pozostaje niezbadany. Niniejsze badanie przekrojowe ma na celu zbadać ochronne działanie oksy­tocyny na wywołane cukrzycą stłuszczenie szpiku kostnego u królików przy użyciu protonowej spektroskopii rezonansu magnetycznego. Materiał i metody: Czterdzieści pięć samic królików nowozelandzkich podzielono losowo na grupę kontrolną, grupę z cukrzycą oraz grupę z cukrzycą leczoną oksytocyną (0.78 mg/kg, i.p.) przez sześć miesięcy. Frakcja tłuszczu (ang. fat fraction; FF) szpiku kostnego została określona za pomocą protonowej spektroskopii rezonansu magnetycznego na początku badania oraz po trzech i sześciu miesiącach. Gęstość mineralną kości zmierzono za pomocą absorpcjometrii promieniowania rentgenowskiego o podwójnej energii. Określono również biomarkery surowicy krwi, metabolizm glikolipidów oraz sporządzono analizę histologiczną adipocytów szpiku kostnego. Wyniki: Leczenie oksytocyną przyniosło pozytywne efekty metaboliczne u królików z cukrzycą, co stwierdzono na podstawie zmian w me­tabolizmie glukozy, wrażliwości na insulinę oraz profili lipidowych. Zauważono drastyczny wzrost stłuszczenia szpiku kostnego u królików z cukrzycą w sposób zależny od czasu; po trzech i sześciu miesiącach, procentowe zmiany frakcji tłuszczu w stosunku do wartości wyjściowej wynosiły odpowiednio 10,1% i 25,8% (wszystkie P &lt; 0.001). Co więcej, leczenie oksytocyną znacząco odwracało wartości frakcji tłuszczu oraz ilościowe parametry adipocytów szpiku kostnego u królików z cukrzycą do poziomu królików z grupy kontrolnej. Oksytocyna poprawiała marker tworzenia kości u królików z cukrzycą w porównaniu do grupy, której podawano sól fizjologiczną. Ponadto, leczenie oksytocyną królików z cukrzycą znacząco łagodziło niszczenie kości w porównaniu do grupy z cukrzycą, której podawano sól fizjologiczną (wszystkie P &lt; 0.05). Wnioski: Oksytocyna wydaje się zmniejszać szkodliwy wpływ hiperglikemii na stłuszczenie szpiku kostnego. Protonowa spektroskopia rezonansu magnetycznego może być cennym narzędziem, dostarczającym uzupełniających informacji na temat oceny skuteczności leczenia

    The effect of a strong magnetic field on the dominant neutrino emissivity in strange stars is investigated. In ultra strong magnetic field, there exists an enhanced neutrino emission because the charged particles are confined to the lowest Landau level. The concept of virtual and real integration operation mode of fully-mechanized mining face based on virtual reality-digital twin-cyber physical system-industrial metaverse is proposed. It has six connotation characteristics, such as display and off-line simulation, monitoring and auxiliary operation, online simulation and preview. It is an evolution process from low-level display simulation to high-level deep integration function. Finally, it have four abilities : the ability of reproduction mapping from real to virtual precision, the ability of reasoning and forecasting decision-making in virtual iteration, the ability of reproduction control from virtual to real, the ability of seamless cooperation between virtual and real human-computer, and the ability of lean management.
