Evaluation of the Kinetic Properties of the Sporulation Protein SpoIIE of Bacillus subtilis by Inclusion in a Model Membrane
Starvation induces Bacillus subtilis to initiate a developmental process (sporulation) that includes asymmetric cell division to form the prespore and the mother cell. The integral membrane protein SpoIIE is essential for the prespore-specific activation of the transcription factor σ(F), and it also has a morphogenic activity required for asymmetric division. An increase in the local concentration of SpoIIE at the polar septum of B. subtilis precedes dephosphorylation of the anti-anti-sigma factor SpoIIAA in the prespore. After closure and invagination of the asymmetric septum, phosphatase activity of SpoIIE increases severalfold, but the reason for this dramatic change in activity has not been determined. The central domain of SpoIIE has been seen to self-associate (I. Lucet et al., EMBO J. 19:1467-1475, 2000), suggesting that activation of the C-terminal PP2C-like phosphatase domain might be due to conformational changes brought about by the increased local concentration of SpoIIE in the sporulating septum. Here we report the inclusion of purified SpoIIE protein into a model membrane as a method for studying the effect of local concentration in a lipid bilayer on activity. In vitro assays indicate that the membrane-bound enzyme maintains dephosphorylation rates similar to the highly active micellar state at all molar ratios of protein to lipid. An antimicrobial impregnated urinary catheter that reduces mineral encrustation and prevents colonisation by multi-drug resistant organisms for up to 12 weeks
Two major complications of indwelling urinary catheterisation include infection and mineral encrustation of the catheter. Our antimicrobial urinary catheter (AUC) impregnated with rifampicin, triclosan, and sparfloxacin has demonstrated long-term protective activity against major uropathogens. This study aimed to firstly assess the ability of the AUC to resist mineral encrustation in the presence and absence of bacteria. Secondly, it aimed to investigate the AUC’s anti-biofilm activity against multi-drug resistant organisms. There was no difference in surface roughness between AUC and control segments. In a static and a perfusion model, phosphate deposition was significantly reduced on AUCs challenged with P. mirabilis. Furthermore, none of the AUCs blocked during the 28 day test period, unlike controls. The AUC prevented colonisation by methicillin-resistant Staphylococcus aureus, methicillin-resistant Staphylococcus epidermidis, extended-spectrum beta-lactamase producing E. coli, and carbapenemase-producing E. coli for 12 consecutive weekly challenges. All three drugs impregnated into the catheter continued to exert protective activity throughout 12 weeks of constant perfusion. The drugs appear to migrate into the crystalline biofilm to continually protect against bacteria not it direct contact with the catheter surface. In conclusion, the AUC reduces mineral encrustation and may increase time to blockage in the presence of P. mirabilis, and does not predispose to mineral deposition under other conditions. It also offers 12 weeks of protection against multi-drug resistant bacteria
Spatial and Temporal Variation of Soil Salinity During Dry and Wet Seasons in the Southern Coastal Area of Laizhou Bay, China
260-270The southern coastal area of Laizhou Bay is subjected to severe soil salinization due to saline groundwater. The degree of spatial variability is strongly affected by seasonal changes during an annual cycle. In this paper, the spatio-temporal variability of soil salinity in Laizhou Bay, China, was examined to ascertain the current situation of soil salinization in the study area and to reveal the characteristics of seasonal variation of soil salinity. The classical statistical methods and geostatistical methods were applied to soil salinity data collected from four soil layers, i.e., 0-30, 30-60, 60-90, and 0-100 cm, during summer and autumn in 2014. The results indicated that the variation of soil salinity of all the soil layers in summer and autumn was moderate. The soil salinity in the 0-30 cm layer showed a moderate spatial autocorrelation, whereas the spatial autocorrelations of soil salinity in other layers were strong. The overall spatial distribution of soil salinity showed a clear banding distribution and the degree of salinization in the eastern area was lower than that in the western and northern regions.A high ratio of evaporation/precipitation is one of the important reasons for the soil salinity in July is significantly higher than that in November. The rank of soil salinity under different land-use types was: salt pan > orchard > weeds > soybean > woods > cotton > maize > ginger > sweet potato. The research findings can provide theoretical guidance for accurate assessment and soil partition management of regional soil salinization
Scutellarin Attenuates Hypertension-Induced Expression of Brain Toll-Like Receptor 4/Nuclear Factor Kappa B
Hypertension is associated with low-grade inflammation, and Toll-like receptor 4 (TLR4) has been shown to be linked to the development and maintenance of hypertension. This study aimed to investigate the effects of scutellarin (administered by oral gavage daily for 2 weeks) on brain TLR4/nuclear factor kappa B-(NF-κB-) mediated inflammation and blood pressure in renovascular hypertensive (using the 2-kidney, 2-clip method) rats. Immunofluorescence and western immunoblot analyses revealed that hypertension contributed to the activation of TLR4 and NF-κB, accompanied by significantly enhanced expression of proinflammatory mediators, such as tumor necrosis factor-α (TNF-α), interleukin-1β (IL-1β), and interleukin-18 (IL-18). Furthermore, expression of the antiapoptotic protein, myeloid cell leukemia-1 (Mcl1), was decreased, and the pro-apoptotic proteins, Bax and cleavedcaspase-3 p17 were increased in combined cerebral cortical/striatal soluble lysates. Scutellarin significantly lowered blood pressure and attenuated the number of activated microglia and macrophages in brains of hypertensive rats. Furthermore, scutellarin significantly reduced the expression of TLR4, NF-κB p65, TNF-α, IL-1β, IL-18, Bax and cleaved-caspase-3 p17, and increased the expression of Mcl1. Overall, these results revealed that scutellarin exhibits anti-inflammatory and anti-apoptotic properties and decreases blood pressure in hypertensive rats. Therefore, scutellarin may be a potential therapeutic agent in hypertension-associated diseases
Hydrochemical Characteristics and Quality Assessment of Groundwater under the Impact of Seawater Intrusion and Anthropogenic Activity in the Coastal Areas of Zhejiang and Fujian Provinces, China
AbstractCoastal groundwater is an important resource in the developed region associated with human health and sustainable economic development. To identify the origins of salinity and evaluate the impact of water-rock interactions, seawater intrusion (SWI), and evaporation on groundwater in the coastal areas of Zhejiang and Fujian provinces, a comprehensive investigation was performed. Meanwhile, nitrate and fluoride indicators resulting from the anthropogenic activity and SWI were also considered. At last, the water quality index (WQI) of coastal groundwater was evaluated with geochemical and multivariate statistical methods. The results indicated that (1) the groundwater in coastal areas of Zhejiang and Fujian provinces has been affected by SWI to varying degrees. The analysis of selected ion ratios (Na+/Cl− and Br−/Cl−) and isotopic compositions showed that SWI is the predominant cause of increasing salinity in the groundwater of Zhejiang Province, while the cause is water-rock interactions (ion exchange and mineral weathering) in Fujian Province. The hydrochemical evolution path of groundwater in Zhejiang Province is Ca/Mg-HCO3 to Na-Cl, while a different pattern of Ca/Mg-HCO3 to Na (Mg/Ca)-Cl occurs in Fujian Province. However, the trend of SWI development in both provinces was freshening. (2) Nitrification, sewage infiltration, and SWI increased the NO3− content in groundwater. Some of the NO3− concentration in Fujian Province exceeds the standard, and the nitrogen pollution was more serious than in Zhejiang Province. The F− content in coastal groundwater was affected by SWI and mineral dissolution; the F− content in Zhejiang Province was higher than in Fujian Province, which was close to the groundwater standard limit. The average WQI value of Zhejiang was 103.61, and the WQI of Fujian was 61.69, indicating that the coastal groundwater quality in Fujian Province was better than in Zhejiang Province. The results of the study revealed the impact of SWI and anthropogenic activity on groundwater in the southern coastal zone of China and will be valuable for sustainable groundwater resource management
Distribution of 222Rn in Seawater Intrusion Area and Its Implications on Tracing Submarine Groundwater Discharge on the Upper Gulf of Thailand
AbstractRadon (222Rn) has been widely employed as a tracer for estimating submarine groundwater discharge (SGD). However, the uncertainty of the SGD estimation remains significant, due to the spatial variability of radon in groundwater. In this study, we analyzed the hydrochemical proprieties of seawater and coastal groundwater in the Upper Gulf of Thailand and discussed the distribution characteristics of 222Rn in aquifers in terms of aquifer lithology, groundwater system recharge conditions, and water retention time. The results suggested that the residence time of groundwater and the process of groundwater salinization have the greatest impact on the distribution of 222Rn activity. A 222Rn mass balance model, synthesizing the distribution characteristics of 222Rn in groundwater and tidal influences on SGD, was built to estimate the submarine groundwater discharge in the Upper Gulf of Thailand. The result showed that the SGD flux of the Upper Gulf of Thailand was 0.0203 m/d. Moreover, there is a positive correlation between tidal height and the activity of 222Rn in groundwater. The SGD observed during the low tide was about 1.25 times higher than that observed during the high tide. This may influence the marine geochemical cycles of elements and their impact on marine ecosystems
A novel lytic phage potentially effective for phage therapy against Burkholderia pseudomallei in the tropics.
BACKGROUND: Burkholderia pseudomallei is a tropical pathogen that causes melioidosis. Its intrinsic drug-resistance is a leading cause of treatment failure, and the few available antibiotics require prolonged use to be effective. This study aimed to assess the clinical potential of B. pseudomallei phages isolated from Hainan, China. METHODS: Burkholderia pseudomallei strain (HNBP001) was used as the isolation host, and phages were recovered from domestic environmental sources, which were submitted to the host range determination, lytic property assays, and stability tests. The best candidate was examined via the transmission electron microscope for classification. With its genome sequenced and analyzed, its protective efficacy against B. pseudomallei infection in A549 cells and Caenorhabditis elegans was evaluated, in which cell viability and survival rates were compared using the one-way ANOVA method and the log-rank test. RESULTS: A phage able to lyse 24/25 clinical isolates was recovered. It was classified in the Podoviridae family and was found to be amenable to propagation. Under the optimal multiplicity of infection (MOI) of 0.1, an eclipse period of around 20 min and a high titer (1012 PFU/ml) produced within 1 h were demonstrated. This phage was found stabile at a wide range of temperatures (24, 37, 40, 50, and 60 °C) and pH values (3-12). After being designated as vB_BpP_HN01, it was fully sequenced, and the 71,398 bp linear genome, containing 93 open reading frames and a tRNA-Asn, displayed a low sequence similarity with known viruses. Additionally, protective effects of applications of vB_BpP_HN01 (MOI = 0.1 and MOI = 1) alone or in combination with antibiotics were found to improve viability of infected cells (70.6 ± 6.8%, 85.8 ± 5.7%, 91.9 ± 1.8%, and 96.8 ± 1.8%, respectively). A significantly reduced mortality (10%) and a decreased pathogen load were demonstrated in infected C. elegans following the addition of this phage. CONCLUSIONS: As the first B. pseudomallei phage was isolated in Hainan, China, phage vB_BpP_HN01 was characterized by promising lytic property, stability, and efficiency of bacterial elimination during the in vitro/vivo experiments. Therefore, we can conclude that it is a potential alternative agent for combating melioidosis
Whole-genome sequencing of the snub-nosed monkey provides insights into folivory and evolutionary history
Colobines are a unique group of Old World monkeys that principally eat leaves and seeds rather than fruits and insects. We report the sequencing at 146× coverage, de novo assembly and analyses of the genome of a male golden snub-nosed monkey (Rhinopithecus roxellana) and resequencing at 30× coverage of three related species (Rhinopithecus bieti, Rhinopithecus brelichi and Rhinopithecus strykeri). Comparative analyses showed that Asian colobines have an enhanced ability to derive energy from fatty acids and to degrade xenobiotics. We found evidence for functional evolution in the colobine RNASE1 gene, encoding a key secretory RNase that digests the high concentrations of bacterial RNA derived from symbiotic microflora. Demographic reconstructions indicated that the profile of ancient effective population sizes for R. roxellana more closely resembles that of giant panda rather than its congeners. These findings offer new insights into the dietary adaptations and evolutionary history of colobine primates
Green Tea Powder Decreased Egg Weight Through Increased Liver Lipoprotein Lipase and Decreased Plasma Total Cholesterol in an Indigenous Chicken Breed
Whether or not green tea promotes egg production is unclear. Huainan partridge chickens at 20 weeks of age were divided into two groups, with one group fed a basal diet (control) and one fed a basal diet plus 10 g/kg green tea powder (GTP) for 12 weeks. Egg production (EP) and feed intake (FI) were recorded daily. Plasma lipid parameters, and apolipoprotein-B (Apo-B), 3-hydroxy-3-methylglutaryl coenzyme A reductase (HMGR), and lipoprotein lipase (LPL) expression were determined every four weeks. Egg production and FI showed no significant difference between the two groups (p > 0.05). Egg weight was 47.58 g in the control group, which was higher than that of the GTP group, and the feed-to-egg ratio (FCR) was 4.62 in the control group, which was lower than that of the GTP group after 12 weeks feeding. Compared with the control group, plasma orexin A (p < 0.05), high-density lipoprotein (HDL), apolipoprotein A (Apo A), and very high-density lipoprotein (VHDL) (p < 0.01, respectively) were increased. Plasma glucose (Glu), free fatty acid (FFA), apolipoprotein B (Apo B), triglyceride (TG), total cholesterol (TC) (p < 0.01, respectively), and low density lipoprotein (LDL) (p < 0.05) were decreased in the GTP group after 8 weeks feeding. The LPL expression in the liver was increased in the GTP group after 8 to 12 weeks feeding when compared to the control group (p < 0.05). Chickens fed GTP did not affect EP, but decreased egg weight, which might be because of lower plasma lipid concentration, increased plasma Orexin A, and liver LPL expression
