    Protective effect of vanillin in streptozotocin-induced diabetes in neonatal rats via attenuation of oxidative stress and inflammation

    Purpose: To evaluate the antidiabetic activity of vanillin in streptozotocin (STZ)-induced diabetic rats.Methods: Diabetes was induced in 2-day old male pups by intraperitoneal (i.p.) administration of STZ (90 mg/kg). The pups were then randomly assigned to four groups: control group which received citrate buffer only in place of STZ; negative control group, i.e., diabetic group; and vanillin-treated groups which received vanillin (100 or 200 mg/kg, p.o.) continuously from the 6th week of age to the 10th week. The antidiabetic effect of vanillin was determined by measuring the serum levels of insulin, triglycerides and glucose in the diabetic rats. Oral glucose tolerance, kidney and liver function tests were also performed at the end of the protocol. Moreover, the oxidative stress and inflammatory cytokines in liver tissues, and histopathological changes in pancreatic tissues were assessed.Results: Vanillin treatment significantly decreased serum glucose and triglyceride levels and increased the level of insulin, when compared to the negative control group. There was higher insulin sensitivity in the vanillin-treated group than in the negative control group. In addition, vanillin improved liver and renal functions in STZ-induced diabetic neonatal rats. Hepatic oxidative stress and inflammatory mediators, as well as histopathological changes in pancreas were attenuated by vanillin treatment.Conclusion: These results reveal that vanillin attenuates hyperglycemia in STZ-induced neonatal diabetic rat model by decreasing oxidative stress and inflammatory cytokines.     Development and validation of risk prediction and neural network models for dilated cardiomyopathy based on WGCNA

    BackgroundDilated cardiomyopathy (DCM) is a progressive heart condition characterized by ventricular dilatation and impaired myocardial contractility with a high mortality rate. The molecular characterization of DCM has not been determined yet. Therefore, it is crucial to discover potential biomarkers and therapeutic options for DCM.MethodsThe hub genes for the DCM were screened using Weighted Gene Co-expression Network Analysis (WGCNA) and three different algorithms in Cytoscape. These genes were then validated in a mouse model of doxorubicin (DOX)-induced DCM. Based on the validated hub genes, a prediction model and a neural network model were constructed and validated in a separate dataset. Finally, we assessed the diagnostic efficiency of hub genes and their relationship with immune cells.ResultsA total of eight hub genes were identified. Using RT-qPCR, we validated that the expression levels of five key genes (ASPN, MFAP4, PODN, HTRA1, and FAP) were considerably higher in DCM mice compared to normal mice, and this was consistent with the microarray results. Additionally, the risk prediction and neural network models constructed from these genes showed good accuracy and sensitivity in both the combined and validation datasets. These genes also demonstrated better diagnostic power, with AUC greater than 0.7 in both the combined and validation datasets. Immune cell infiltration analysis revealed differences in the abundance of most immune cells between DCM and normal samples.ConclusionThe current findings indicate an underlying association between DCM and these key genes, which could serve as potential biomarkers for diagnosing and treating DCM

    The Impact of Spatial Structure on Economic Efficiency of Beijing-Tianjin-Hebei Megalopolis in China

    This study constructs a theoretical model and empirical framework concerning how spatial structure affects economic efficiency using data on the Beijing-Tianjin-Hebei (BTH) megaregion between 2008 and 2017. The study finds the following: ① the development of the internal spatial structure of the BTH urban agglomeration is unequal. The populations of most cities in the urban agglomeration are still in a dispersed state. Although urban populations have tended to agglomerate around multiple subcenter units in the cities, the trend towards population agglomeration around city centers is not found to be significant. ② The total factor productivity (TFP) of the BTH urban agglomeration was not high in most years between 2008 and 2017, showing a fluctuating downward trend overall. The TFP of the urban agglomeration showed differential regional patterns. The decline of TFP growth in the BTH urban agglomeration is mainly due to declining technological progress, technological efficiency, and scale efficiency. Resource input remains the major driving force behind the development of the BTH megaregion. ③ Concerning how the spatial structure of the urban agglomeration affected economic efficiency, the study finds that primacy, urban Gini index, urban population size, human capital, informatization level, industrial structure, and science and technology levels have positive effects on economic efficiency, whereas dispersion, governmental role, economic openness, and land input have negative effects. This study has several policy implications. Achieving coordinated and integrated development of the BTH urban agglomeration will require constructing a scientific and regional spatial system, improving the development levels of regional central cities, divesting Beijing of noncapital functions, and reshaping the industrial layout of the BTH megaregion in an orderly manner, while continuously improving the internal hierarchical structure of urban agglomeration and strengthening intercity economic connections

    FOG De-Noising Algorithm Based on Augmented Nonlinear Differentiator and Singular Spectrum Analysis

    A novel algorithm based on singular spectrum analysis (SSA) and augmented nonlinear differentiator (AND) for extracting the useful signal from a noisy measurement of fiber optic gyroscope (FOG) is proposed in this paper. As a novel type of tracking differentiator, augmented nonlinear differentiator (AND) has the advantages of dynamical performance and noise-attenuation ability. However, there is a contradiction in AND, i.e., selecting a larger acceleration factor may cause faster convergence but bad random noise reduction, whereas selecting a smaller acceleration factor may lead to signal delay but effective random noise reduction. To overcome the contradiction of AND, multi-scale transformation is introduced. Firstly, the noisy signal is decomposed into components by SSA, and the correlation coefficients between each component and original signal are calculated, then the component with biggest correlation coefficient is reserved and other components are filtered by AND with designed selection criterion of acceleration factor, finally the de-noising result is obtained after reconstruction process. There are mainly two prominent advantages of the proposed SSA-AND algorithm: (i) Compared to traditional tracking differentiators, better de-noising ability can be achieved without signal delay; and (ii) compared to other widely used hybrid de-noising methods based on multi-scale transformation, a parameter determination method is given based on the correlation coefficient of each decomposed component, which improves the reliability of the proposed algorithm

    Comparative neonatal outcomes in singleton births from blastocyst transfers or cleavage-stage embryo transfers: a systematic review and meta-analysis

    Abstract Background Comparative neonatal outcomes with respect to singleton births from blastocyst transfers or cleavage-state embryo transfers are controversial with respect to which method is superior. Many studies have yielded contradictory results. We performed a systematic review and meta-analysis for the purpose of comparing neonatal outcomes in single births following IVF/ICSI. Methods We searched the Medline, Embase and Cochrane Central Register of Clinical Trials (CCTR) databases until October 2016. Studies and trials that contained neonatal outcomes for singleton births were included. Data were extracted in 2 × 2 tables. The analysis was performed using Rev Man 5.1 software. Risk ratios (RRs) and risk differences, with 95% confidence intervals, were calculated to assess the results of each outcome. Subgroups were applied in all outcomes. Newcastle-Ottawa scale (NOS) checklists were used to assess the quality of the referenced studies. Results Twelve studies met the criteria in this meta-analysis. There was a high risk of preterm birth after blastocyst embryo transfer versus the risk after cleavage-stage transfer (RR: 1.11, 95% CI: 1.01–1.22). For the “only fresh” subgroup, the outcome was coincident (RR: 1.16, 95% CI: 1.06–1.27). For the “fresh and frozen” and “only frozen” subgroups, there were no differences. Patients who received fresh blastocyst embryo transfers had a high risk of very preterm births (RR: 1.16, 95% CI: 1.02–1.31). Finally, cleavage-stage embryo transfers were associated with a high risk of infants who were small for gestational age (0.83, 95% CI: 0.76–0.92) and a low risk of those who were large for gestation age (1.14, 95% CI: 1.04–1.25). Conclusions The risks of preterm and very preterm births increased after fresh blastocyst transfers versus the risks after fresh cleavage-stage embryo transfers. However, in frozen embryo transfers, there were no differences. Blastocyst embryo transfers resulted in high risks of infants who were large for gestational age, and cleavage-stage embryo transfers resulted in high risks of infants who were small for gestational age

    The Impact of Preconception TSH on the Reproductive Outcomes of Infertile Women Undergoing the First Fresh D3 Embryo Transfer Cycle

    Purpose. To investigate the association between high-normal preconception TSH levels and reproductive outcomes in infertile women undergoing the first fresh D3 embryo transfer. Methods. This was a retrospective study. Euthyroid patients undergoing the first fresh D3 embryo transfer from January 2018 to May 2019 were initially included. The patients were divided into a low-TSH (0.27–2.5 mIU/L) group and a high-normal TSH (2.5–4.2 Miu) group. The reproductive outcomes were compared between the groups. Results. A total of 1786 women were ultimately included, in which 1008 of whom had serum TSH levels between 0.27 and 2.5 mIU/L and 778 of whom had serum TSH levels between 2.5 and 4.2 mIU/L. The patients were highly homogeneous in terms of general characteristics. High-normal TSH levels had no adverse impact on the clinical pregnancy rate, miscarriage rate, or live birth rate (respectively, aOR = 0.92, 1.30, and 0.88 and P = 0.416, 0.163, and 0.219). No significant differences were observed in terms of gestational age, single live birth rates, and birth weight, or birth length. Conclusion. High-normal TSH levels did not significantly influence reproductive outcomes in infertile women undergoing the first fresh D3 embryo transfer. Further studies are needed to test whether the results might be applicable to a wider population
