    Inherent SM Voltage Balance for Multilevel Circulant Modulation in Modular Multilevel DC--DC Converters

    The modularity of a modular multilevel dc converter (MMDC) makes it attractive for medium-voltage distribution systems. Inherent balance of submodule (SM) capacitor voltages is considered as an ideal property, which avoids a complex sorting process based on many measurements thereby reducing costs and enhancing reliability. This article extends the inherent balance concept previously shown for square-wave modulation to a multilevel version for MMDCs. A switching duty matrix dU is introduced: it is a circulant matrix of preset multilevel switching patterns with multiple stages and multiple durations. Inherent voltage balance is ensured with a full-rank dU . Circulant matrix theory shows that this is equivalent to a simplified common factor criterion. A nonfull rank dU causes clusters of SM voltage rather than a single common value, with the clusters indicated by the kernel of the matrix. A generalized coprime criterion is developed into several deductions that serve as practical guidance for design of multilevel circulant modulation. The theoretical development is verified through full-scale simulations and downscaled experiments. The effectiveness of the proposed circulant modulation in achieving SM voltage balance in an MMDC is demonstrated

    ABSCISIC ACID-INSENSITIVE 5-KIP-RELATED PROTEIN 1-SHOOT MERISTEMLESS modulates reproductive development of Arabidopsis

    Soil (or plant) water deficit accelerates plant reproduction. However, the underpinning molecular mechanisms remain unknown. By modulating cell division/number, ABSCISIC ACID-INSENSITIVE 5 (ABI5), a key bZIP (basic (region) leucine zippers) transcription factor, regulates both seed development and abiotic stress responses. The KIP-RELATED PROTEIN (KRP) cyclin-dependent kinases (CDKs) play an essential role in controlling cell division, and SHOOT MERISTEMLESS (STM) plays a key role in the specification of flower meristem identity. Here, our findings show that abscisic acid (ABA) signaling and/or metabolism in adjust reproductive outputs (such as rosette leaf number and open flower number) under water-deficient conditions in Arabidopsis (Arabidopsis thaliana) plants. Reproductive outputs increased under water-sufficient conditions but decreased under water-deficient conditions in the ABA signaling/metabolism mutants abscisic acid2-1 (aba2-1), aba2-11, abscisic acid insensitive3-1 (abi3-1), abi4-1, abi5-7, and abi5-8. Further, under water-deficient conditions, ABA induced-ABI5 directly bound to the promoter of KRP1, which encodes a CDK that plays an essential role in controlling cell division, and this binding subsequently activated KRP1 expression. In turn, KRP1 physically interacted with STM, which functions in the specification of flower meristem identity, promoting STM degradation. We further demonstrate that reproductive outputs are adjusted by the ABI5–KRP1–STM molecular module under water-deficient conditions. Together, our findings reveal the molecular mechanism by which ABA signaling and/or metabolism regulate reproductive development under water-deficient conditions. These findings provide insights that may help guide crop yield improvement under water deficiency

    Clinical values of multiple Epstein-Barr virus (EBV) serological biomarkers detected by xMAP technology

    <p>Abstract</p> <p>Background</p> <p>Serological examination of Epstein-Barr virus (EBV) antibodies has been performed for screening nasopharyngeal carcinoma (NPC) and other EBV-associated diseases.</p> <p>Methods</p> <p>By using xMAP technology, we examined immunoglobulin (Ig) A antibodies against Epstein-Barr virus (EBV) VCA-gp125, p18 and IgA/IgG against EA-D, EBNA1 and gp78 in populations with distinct diseases, or with different genetic or geographic background. Sera from Cantonese NPC patients (n = 547) and healthy controls (n = 542), 90 members of high-risk NPC families and 52 non-endemic healthy individuals were tested. Thirty-five of NPC patients were recruited to observe the kinetics of EBV antibody levels during and after treatment. Patients with other EBV-associated diseases were collected, including 16 with infectious mononucleosis, 28 with nasal NK/T cell lymphoma and 14 with Hodgkin's disease.</p> <p>Results</p> <p>Both the sensitivity and specificity of each marker for NPC diagnosis ranged 61–84%, but if combined, they could reach to 84.5% and 92.4%, respectively. Almost half of NPC patients displayed decreased EBV immunoactivities shortly after therapy and tumor recurrence was accompanied with high EBV antibody reactivates. Neither the unaffected members from high-risk NPC families nor non-endemic healthy population showed statistically different EBV antibody levels compared with endemic controls. Moreover, elevated levels of specific antibodies were observed in other EBV-associated diseases, but all were lower than those in NPC.</p> <p>Conclusion</p> <p>Combined EBV serological biomarkers could improve the diagnostic values for NPC. Diverse EBV serological spectrums presented in populations with different EBV-associated diseases, but NPC patients have the highest EBV activity.</p

    The quasar SDSS J142507.32+323137.4 : dual AGNs?

    We analyze the optical spectrum of type 1 QSO SDSS J1425+3231. This ob- ject is interesting since its narrow emission lines such as [O III]{\lambda}{\lambda}4959, 5007 are double- peaked, and the line structure can be modeled well by three Gaussian components: two components for the two peaks (we refer the peaks at low/high redshift as "the blue/red component") and another one for the line wing which has the same line center as that of the blue component, but ~ 3 times broader. The separation between the blue and red components is ~ 500 km/s with blue component ~ 2 times broader than the red one. The H{\beta} emission can be separated into four components: two for the double-peaked narrow line and two for the broad line which comes from the broad line region (BLRs). The black hole mass estimated from the broad H{\beta} emission line using the typical reverberation map- ping relation is 0.85 \times 108M\odot, which is consistent with that derived from parameters of [O III]{\lambda} 5007 of the blue component. We suggest this QSO might be a dual AGN system, the broad H{\beta} emission line is mainly contributed by the primary black hole (traced by the blue component) while the broad H{\beta} component of the secondary black hole (traced by the red component) is hard to be separated out considering a resolution of ~2000 of SDSS spectra or it is totally obscured by the dusty torus.Comment: 9 pages, 2 figures, accepted for publication in RA

    A pilot study on acute inflammation and cancer: a new balance between IFN-γ and TGF-β in melanoma

    Recent data have redefined the concept of inflammation as a critical component of tumor progression. However, there has been little development on cases where inflammation on or near a wound and a tumor exist simultaneously. Therefore, this pilot study aims to observe the impact of a wound on a tumor, to build a new mouse tumor model with a manufactured surgical wound representing acute inflammation, and to evaluate the relationship between acute inflammation or wound healing and the process of tumor growth. We focus on the two phases that are present when acute inflammation influences tumor. In the early phase, inhibitory effects are present. The process that produces these effects is the functional reaction of IFN-γ secretions from a wound inflammation. In the latter phase, the inhibited tumor is made resistant to IFN-γ through the release of TGF-β to balance the inflammatory factor effect on the tumor cells. A pair of cytokines IFN-γ/TGF-β established a new balance to protect the tumor from the interference effect of the inflammation. The tumor was made resistant to IFN-γ through the release of TGF-β to balance the inflammatory effect on the tumor cells. This balance mechanism that occurred in the tumor cells increased proliferation and invasion. In vitro and in vivo experiments have confirmed a new view of clinical surgery that will provide more detailed information on the evaluation of tumors after surgery. This study also provides a better understanding of the relationship between tumor and inflammation, as well as tumor cell attacks on inflammatory factors

    Glycosylation on Spermatozoa, a Promise for the Journey to the Oocyte

    Spermatozoa experience a long and tough transit in male and female genital tracts before successful fertilization. Glycosylation helps spermatogenesis, epididymal maturation, passing through cervical mucus, avoiding killing of the female immunologic system, and shaking hands between sperm and egg. Changes in glycosylations along the transit ensure that the right things happen at the right time and place on spermatozoa. Aberrant glycosylations on spermatozoa will negatively affect their fertility. Thus, we developed a lectin array method to examine the glycocalyx of spermatozoa, which will help observe glycosylations occurring on spermatozoa in a normal or abnormal conditions, such as spermatozoa with DEF126 mutation and poor freezability. Intriguingly, binding levels of ABA (Agaricus bisporus agglutinin), a lectin marking the inner layer of the glycocalyx, were changed in these subfertile spermatozoa, which indicates that the integrity of glycocalyx is critical for sperm fertility.     Leveraging Large Language Models for Pre-trained Recommender Systems

    Recent advancements in recommendation systems have shifted towards more comprehensive and personalized recommendations by utilizing large language models (LLM). However, effectively integrating LLM's commonsense knowledge and reasoning abilities into recommendation systems remains a challenging problem. In this paper, we propose RecSysLLM, a novel pre-trained recommendation model based on LLMs. RecSysLLM retains LLM reasoning and knowledge while integrating recommendation domain knowledge through unique designs of data, training, and inference. This allows RecSysLLM to leverage LLMs' capabilities for recommendation tasks in an efficient, unified framework. We demonstrate the effectiveness of RecSysLLM on benchmarks and real-world scenarios. RecSysLLM provides a promising approach to developing unified recommendation systems by fully exploiting the power of pre-trained language models.Comment: 13 pages, 4 figure

    Nuciferine downregulates Per-Arnt-Sim kinase expression during its alleviation of lipogenesis and inflammation on oleic acid-induced hepatic steatosis in HepG2 cells

    Nonalcoholic fatty liver disease (NAFLD) is a prevalent liver disease associated with lipotoxicity, lipid peroxidation, oxidative stress and inflammation. Nuciferine, an active ingredient extracted from the natural lotus leaf, has been reported to be effective for the prevention and treatment of NAFLD. Per-Arnt-Sim kinase (PASK) is a nutrient responsive protein kinase that regulates lipid and glucose metabolism, mitochondrial respiration and gene expression. The aim of the present study was to investigate the protective effect of nuciferine against NAFLD and its inhibitory effect on PASK, exploring the possible underlying mechanism of nuciferine-mediated inhibition on NAFLD. Relevant biochemical parameters (lipid accumulation, extent of oxidative stress and release of inflammation cytokines) in oleic acid (OA)-induced HepG2 cells that mimicked steatosis in vitro were measured and compared with the control. It was found that nuciferine and silenced-PASK (siRNA PASK) both inhibited triglyceride (TG) accumulation and was effective in decreasing fatty acid (FFAs). The content of total antioxidant capacity (T-AOC) and superoxide dismutase (SOD) were increased respectively by nuciferine and siRNA PASK without increase in glutathione (GSH). Malondialdehyde (MDA) was decreased respectively by nuciferine and siRNA PASK. In addition, nuciferine decreased TNF-a, IL-6 and IL-8 as well as the siRNA PASK group. IL-10 was increased by nuciferine and siRNA PASK respectively. Further investigation revealed that nuciferine and siRNA PASK could respectively regulate the expression of target genes involved in lipogenesis and inflammation, suggesting that nuciferine may be a potential therapeutic treatment for NAFLD. Furthermore, the modulated effect of nuciferine on (OA)-induced HepG2 cells lipogenesis and inflammation, which was accompanied with PASK inhibition, was also consistent with siRNA PASK, implying that PASK might play a role in nuciferine-mediated regulation on NAFLD

    Enhancing Recommender Systems with Large Language Model Reasoning Graphs

    Recommendation systems aim to provide users with relevant suggestions, but often lack interpretability and fail to capture higher-level semantic relationships between user behaviors and profiles. In this paper, we propose a novel approach that leverages large language models (LLMs) to construct personalized reasoning graphs. These graphs link a user's profile and behavioral sequences through causal and logical inferences, representing the user's interests in an interpretable way. Our approach, LLM reasoning graphs (LLMRG), has four components: chained graph reasoning, divergent extension, self-verification and scoring, and knowledge base self-improvement. The resulting reasoning graph is encoded using graph neural networks, which serves as additional input to improve conventional recommender systems, without requiring extra user or item information. Our approach demonstrates how LLMs can enable more logical and interpretable recommender systems through personalized reasoning graphs. LLMRG allows recommendations to benefit from both engineered recommendation systems and LLM-derived reasoning graphs. We demonstrate the effectiveness of LLMRG on benchmarks and real-world scenarios in enhancing base recommendation models.Comment: 12 pages, 6 figure
