    Leakage-Aware Interconnect for On-Chip Network

    On-chip networks have been proposed as the interconnect fabric for future systems-on-chip and multi-processors on chip. Power is one of the main constraints of these systems and interconnect consumes a significant portion of the power budget. In this paper, we propose four leakage-aware interconnect schemes.     A Comprehensive and Additive System for Child-Focused Assessment and Evaluation in EI/ECSE

    One of the more serious problems facing the field of early intervention/early childhood special education is the disconnect between the community programs that offer screening, eligibility determination, curricular assessment (i.e., programmatic assessment), and progress monitoring services. First, we provide definitions of these service components to ensure clarity. Second, the substantial disconnect found between the screening, eligibility determination, curricular assessment, and progress monitoring components in most communities is discussed. Third, an alternative conceptual framework and its rationale are offered. This framework is designed to ensure that information collected is comprehensive, and that information/data gathered during each component serves as the platform for the next component.     Flux density measurements for 32 pulsars in the 20 cm band

    Flux density measurements provide fundamental observational parameters that describe a pulsar. In the current pulsar catalogue, 27% of radio pulsars have no flux density measurement in the 20 cm observing band. Here, we present the first measurements of the flux densities in this band for 32 pulsars observed using the Parkes radio telescope and provide updated pulse profiles for these pulsars. We have used both archival and new observations to make these measurements. Various schemes exist for measuring flux densities.     A Targeted Library Screen Reveals a New Inhibitor Scaffold for Protein Kinase D

    Protein kinase D (PKD) has emerged as a potential therapeutic target in multiple pathological conditions, including cancer and heart diseases. Potent and selective small molecule inhibitors of PKD are valuable for dissecting PKD-mediated cellular signaling pathways and for therapeutic application. In this study, we evaluated a targeted library of 235 small organic kinase inhibitors for PKD1 inhibitory activity at a single concentration. Twenty-eight PKD inhibitory chemotypes were identified and six exhibited excellent PKD1 selectivity. Five of the six lead structures share a common scaffold, with compound 139 being the most potent and selective for PKD vs PKC and CAMK. Compound 139 was an ATP-competitive PKD1 inhibitor with a low double-digit nanomolar potency and was also cell-active. Kinase profiling analysis identified this class of small molecules as pan-PKD inhibitors, confirmed their selectivity again PKC and CAMK, and demonstrated an overall favorable selectivity profile that could be further enhanced through structural modification. Furthermore, using a PKD homology model based on similar protein kinase structures, docking modes for compound 139 were explored and compared to literature examples of PKD inhibition. Modeling of these compounds at the ATP-binding site of PKD was used to rationalize its high potency and provide the foundation for future further optimization. Accordingly, using biochemical screening of a small number of privileged scaffolds and computational modeling, we have identified a new core structure for highly potent PKD inhibition with promising selectivity against closely related kinases. These lead structures represent an excellent starting point for the further optimization and the design of selective and therapeutically effective small molecule inhibitors of PKD. © 2012 Tandon et al

    Feasibility of Using a Commercial Fitness Tracker as an Adjunct to Family-Based Weight Management Treatment: Pilot Randomized Trial.

    BACKGROUND: Fitness trackers can engage users through automated self-monitoring of physical activity. Studies evaluating the utility of fitness trackers are limited among adolescents, who are often difficult to engage in weight management treatment and are heavy technology users. OBJECTIVE: We conducted a pilot randomized trial to describe the impact of providing adolescents and caregivers with fitness trackers as an adjunct to treatment in a tertiary care weight management clinic on adolescent fitness tracker satisfaction, fitness tracker utilization patterns, and physical activity levels. METHODS: Adolescents were randomized to 1 of 2 groups (adolescent or dyad) at their initial weight management clinic visit. Adolescents received a fitness tracker and counseling around activity data in addition to standard treatment. A caregiver of adolescents in the dyad group also received a fitness tracker. Satisfaction with the fitness tracker, fitness tracker utilization patterns, and physical activity patterns were evaluated over 3 months. RESULTS: A total of 88 adolescents were enrolled, with 69% (61/88) being female, 36% (32/88) black, 23% (20/88) Hispanic, and 63% (55/88) with severe obesity. Most adolescents reported that the fitness tracker was helping them meet their healthy lifestyle goals (69%) and be more motivated to achieve a healthy weight (66%). Despite this, 68% discontinued use of the fitness tracker by the end of the study. There were no significant differences between the adolescent and the dyad group in outcomes, but adolescents in the dyad group were 12.2 times more likely to discontinue using their fitness tracker if their caregiver also discontinued use of their fitness tracker (95% CI 2.4-61.6). Compared with adolescents who discontinued use of the fitness tracker during the study, adolescents who continued to use the fitness tracker recorded a higher number of daily steps in months 2 and 3 of the study (mean 5760 vs 4148 in month 2, P=.005, and mean 5942 vs 3487 in month 3, P=.002). CONCLUSIONS: Despite high levels of satisfaction with the fitness trackers, fitness tracker discontinuation rates were high, especially among adolescents whose caregivers also discontinued use of their fitness tracker. More studies are needed to determine how to sustain the use of fitness trackers among adolescents with obesity and engage caregivers in adolescent weight management interventions

    Vicinal Feature Statistics Augmentation for Federated 3D Medical Volume Segmentation

    Federated learning (FL) enables multiple client medical institutes collaboratively train a deep learning (DL) model with privacy protection. However, the performance of FL can be constrained by the limited availability of labeled data in small institutes and the heterogeneous (i.e., non-i.i.d.) data distribution across institutes. Though data augmentation has been a proven technique to boost the generalization capabilities of conventional centralized DL as a "free lunch", its application in FL is largely underexplored. Notably, constrained by costly labeling, 3D medical segmentation generally relies on data augmentation. In this work, we aim to develop a vicinal feature-level data augmentation (VFDA) scheme to efficiently alleviate the local feature shift and facilitate collaborative training for privacy-aware FL segmentation. We take both the inner- and inter-institute divergence into consideration, without the need for cross-institute transfer of raw data or their mixup. Specifically, we exploit the batch-wise feature statistics (e.g., mean and standard deviation) in each institute to abstractly represent the discrepancy of data, and model each feature statistic probabilistically via a Gaussian prototype, with the mean corresponding to the original statistic and the variance quantifying the augmentation scope. From the vicinal risk minimization perspective, novel feature statistics can be drawn from the Gaussian distribution to fulfill augmentation. The variance is explicitly derived by the data bias in each individual institute and the underlying feature statistics characterized by all participating institutes.     Opposite changes in the expression of clathrin and caveolin-1 in normal and cancerous human prostate tissue: putative clathrin-mediated recycling of EGFR

    Endocytosis, an important macromolecule uptake process in cells, is known to be dysregulated in cancer. Clathrin and caveolin-1 proteins play a major role in receptor-mediated endocytosis. We have used a quantitative, unbiased and semi-automated method to measure in situ protein expression of clathrin and caveolin-1 in cancerous and paired normal (cancer adjacent, non-cancerous) human prostate tissue. There was a significant (p < 0.0001) increase in the expression of clathrin in prostate cancer samples (N = 29, n = 91) compared to normal tissue (N = 29, n = 67) (N = number of patients, n = number of cores in tissue arrays). Conversely, there was a significant (p < 0.0001) decrease in expression of caveolin-1 in prostate cancer tissue compared to normal prostate tissue. The opposite change in expression of the two proteins was highly correlated to increasing cancer aggressiveness. There was also a concurrent increase in the expression of epidermal growth factor receptor (EGFR), a key receptor in carcinogenesis, with clathrin in prostate cancer tissue, indicating recycling of EGFR through clathrin-mediated endocytosis (CME). These results indicate that in prostate cancer, caveolin-1-mediated endocytosis (CavME) may be acting as a brake and increase in CME may facilitate tumorigenicity and aggressiveness of prostate cancer through recycling of EGFR. Changes in the expression of these proteins can also potentially be used as a biomarker for prostate cancer to aid in diagnosis and prognosis and clinical decision-making
