    ENHANCED PRODUCTION OF BIOSOLIDS BY IMPROVED ACTIVATED SLUDGE CLARIFICATION AND STRUCTURED WATER ANALYSIS

    Biosolids contain high contents of soil-required nutrients, so that have been widely applied in land application. The production of the biosolids depends on the clarification performance and dewaterability of the sludge, which are influenced by bioflocculation and structured water content, respectively. Therefore, research on sludge bioflocculation improvement and structured water content determination were proposed in this study. The result indicated that different activated sludge exhibited various bioflocculation limitations. Influence of the sludge characteristics such as the extracellular polymeric substances (EPS) composition, viscosity and floc size on the structured water content were also investigated. The results indicated that no significant correlation was observed between the EPS composition and the structured water content, however, the sludge floc size was positively correlated with it. The bioflocculation limitations were pinpointed, and how floc size influenced the structured water content needed further studies to improve sludge dewaterability, therefore, enhance the biosolids production quantitatively and qualitatively

    MiR-27b-3p Inhibition Enhances Browning of Epididymal Fat in High-Fat Diet Induced Obese Mice

    Objective: Long-term dysregulation of energy balance is the key component of the obesity epidemic. Given the harm of central obesity and the discovery that beige cells appear within white adipose tissue (WAT), enhancing the energy-expending or “browning” ability of visceral adipose tissue (VAT) has become of therapeutic interest. In this study, we focused on the regulating role of microRNA (miRNA)-27b-3p in mice epididymal white adipose tissue (eWAT) browning.Methods: High-fat diet (HFD) induced obese mice model was constructed. Expression of miR-27b-3p and Ucp1 in eWAT was measured during the course of HFD. Through tail vein injection of antimiR-27b-3p, miR-27b-3p expression was inhibited to analyze the potential role of miR-27b-3p in fat browning and metabolism.Results: miR-27b-3p was predominantly expressed in eWAT and browning ability of eWAT in HFD induced obese mice was impaired. Inhibition of miR-27b-3p enhanced browning capacity of eWAT in mice fed an HFD and led to weight loss and insulin sensitivity improvement.Conclusions: High expression of miR-27b-3p in eWAT inhibits browning ability and leads to visceral fat accumulation. It is suggested miR-27b-3p may become a potential therapeutic option for visceral obesity and its associated diseases

    Prevalence of Common Respiratory Viral Infections and Identification of Adenovirus in Hospitalized Adults in Harbin, China 2014 to 2017

    Background: Respiratory infections pose a great challenge in global health, and the prevalence of viral infection in adult patients has been poorly understood in northeast China. Harbin is one of the major cities in northeast China, and more than half of any given year in Harbin is occupied by winter. To reveal the viral etiology and seasonality in adult patients from Harbin, a 4-year consecutive survey was conducted in Harbin, China.Methods: From January 2014 to December 2017, specimens were obtained from adult patients admitted to the Second Affiliated Hospital of Harbin Medical University with lower respiratory tract infections. Sputum samples were examined by direct immunofluorescence assays to detect seven common respiratory viruses, including influenza virus (type A and B), parainfluenza virus (type 1 to 3), respiratory syncytial virus and adenovirus. Adenovirus positive samples were seeded onto A549 cells to isolate viral strains. Phylogenetic analysis was conducted on the highly variable region of adenoviral hexon gene.Results: A total of 1,300 hospitalized adult patients with lower respiratory tract infections were enrolled, in which 189 patients (14.5%) were detected as having at least one viral infection. The co-infection rate in this study was 25.9% (49/189). The dominant viral pathogen from 2014 to 2017 was parainfluenza virus, with a detection rate of 7.2%, followed by influenza virus, respiratory syncytial virus and adenovirus. Based on the climate seasons determined by daily average temperature, the highest overall viral detection rate was detected in spring (22.0%, 52/236), followed by winter (13.4%, 109/813), autumn (11.4%, 13/114) and summer (10.9%, 15/137). Adenovirus type 3 strains with slight variations were isolated from positive cases, which were closely related to the GB strain from the United States, as well as the Harbin04B strain isolated locally.Conclusion: This study demonstrated that common respiratory viruses were partially responsible for hospitalized lower respiratory tract infections in adult patients from Harbin, China, with parainfluenza virus as the dominant viral pathogen. Climate seasons could be rational indicators for the seasonality analysis of airborne viral infections. Future surveillance on viral mutations would be necessary to reveal the evolutionary history of respiratory viruses

    BVT.2733, a Selective 11β-Hydroxysteroid Dehydrogenase Type 1 Inhibitor, Attenuates Obesity and Inflammation in Diet-Induced Obese Mice

    BACKGROUND: Inhibition of 11β-hydroxysteroid dehydrogenase type 1 (11β-HSD1) is being pursued as a new therapeutic approach for the treatment of obesity and metabolic syndrome. Therefore, there is an urgent need to determine the effect of 11β-HSD1 inhibitor, which suppresses glucocorticoid action, on adipose tissue inflammation. The purpose of the present study was to examine the effect of BVT.2733, a selective 11β-HSD1 inhibitor, on expression of pro-inflammatory mediators and macrophage infiltration in adipose tissue in C57BL/6J mice. METHODOLOGY/PRINCIPAL FINDINGS: C57BL/6J mice were fed with a normal chow diet (NC) or high fat diet (HFD). HFD treated mice were then administrated with BVT.2733 (HFD+BVT) or vehicle (HFD) for four weeks. Mice receiving BVT.2733 treatment exhibited decreased body weight and enhanced glucose tolerance and insulin sensitivity compared to control mice. BVT.2733 also down-regulated the expression of inflammation-related genes including monocyte chemoattractant protein 1 (MCP-1), tumor necrosis factor alpha (TNF-α) and the number of infiltrated macrophages within the adipose tissue in vivo. Pharmacological inhibition of 11β-HSD1 and RNA interference against 11β-HSD1 reduced the mRNA levels of MCP-1 and interleukin-6 (IL-6) in cultured J774A.1 macrophages and 3T3-L1 preadipocyte in vitro. CONCLUSIONS/SIGNIFICANCE: These results suggest that BVT.2733 treatment could not only decrease body weight and improve metabolic homeostasis, but also suppress the inflammation of adipose tissue in diet-induced obese mice. 11β-HSD1 may be a very promising therapeutic target for obesity and associated disease

    Does Loneliness Necessarily Lead to a Decrease in Prosocial Behavior? The Roles of Gender and Situation

    Although previous studies show overwhelming evidence that loneliness is negatively correlated with prosocial behavior, some theories and research have implied that under certain situations, loneliness plays a positive role in an individual’s social functioning. The two studies reported in this article examined loneliness and its associations with prosocial behavior in Chinese adults using subjective reporting and experimental design. Study 1 examined 305 Chinese adults (175 males) using the Social and Emotional Loneliness Scale for Adults and the Prosocial Tendencies Measure to evaluate their loneliness and prosocial tendencies. The results showed that loneliness was negatively associated with all prosocial tendencies except the public prosocial tendency. Study 2 examined 177 Chinese adults (61 males) using an experimental design and found that only lonely women in public situations expressed a greater willingness to help. The results also suggest that loneliness may play a positive role in the social functioning of individuals under certain conditions. The function of loneliness and the implications of the association between loneliness and prosocial behavior are discussed

    Interplay between Children’s Electronic Media Use and Prosocial Behavior: The Chain Mediating Role of Parent–Child Closeness and Emotion Regulation

    In the contemporary digital milieu, children’s pervasive engagement with electronic media is ubiquitous in their daily lives, presenting complex implications for their socialization. Prosocial behavior, a cornerstone of social interaction and child development, is intricately intertwined with these digital experiences. This relation gains further depth, considering the significant roles of parent–child relationships and emotion regulation in shaping children’s social trajectories. This study surveyed 701 families to examine the association between children’s electronic media use and prosocial behavior, specifically exploring the mediating roles of parent–child closeness and emotion regulation. Structural equation modeling was employed for the analysis. Children’s electronic media use negatively correlated with prosocial behavior, parent–child closeness, and emotion regulation. In contrast, a positive association emerged between parent–child closeness, emotion regulation, and prosocial behavior. Emotion regulation also correlated positively with prosocial behavior. Statistical analyses revealed that parent–child closeness and emotion regulation function as both individual and sequential mediators in the relation between electronic media use and prosocial behavior. The study’s analyses reveal that fostering children’s prosocial behavior in the digital era requires strong family ties, effective emotional management, and balanced digital exposure, which are pivotal for their comprehensive development

    Association between Electronic Media Use and Internalizing Problems: The Mediating Effect of Parent–Child Conflict and Moderating Effect of Children’s Age

    In today’s digital world, children are exposed extensively to electronic media, making it an integral part of their daily lives. However, excessive use of electronic media during childhood has been associated with various internalizing problems. Moreover, parent–child conflict and children’s age may be closely associated with children’s problem behaviors. The current study employed a cross-sectional design and conducted a questionnaire survey of 711 parents to examine the association between children’s electronic media use and their internalizing problems. Furthermore, this study probed the mediating role of parent–child conflict within this association and the moderating effect of children’s age. The results of structural equation modeling showed a positive correlation between children’s use of electronic media and their internalizing difficulties. Parent–child conflict served as a mediating factor in this association. Results also showed that the association between parent–child conflict and internalizing problems becomes more pronounced as children grow older. These findings imply that parents should encourage their children to develop healthy habits in using electronic media while fostering positive relationships. Parents should also be mindful of the psychological changes as children age and provide guidance to help them become proficient digital citizens

    Tumor-targeting peptides from combinatorial libraries

    Cancer is one of the major and leading causes of death worldwide. Two of the greatest challenges in fighting cancer are early detection and effective treatments with no or minimum side effects. Widespread use of targeted therapies and molecular imaging in clinics requires high affinity, tumor-specific agents as effective targeting vehicles to deliver therapeutics and imaging probes to the primary or metastatic tumor sites. Combinatorial libraries such as phage-display and one-bead one-compound (OBOC) peptide libraries are powerful approaches in discovering tumor-targeting peptides. This review gives an overview of different combinatorial library technologies that have been used for the discovery of tumor-targeting peptides. Examples of tumor-targeting peptides identified from each combinatorial library method will be discussed. Published tumor-targeting peptide ligands and their applications will also be summarized by the combinatorial library methods and their corresponding binding receptors
