    Decoding Fujian’s cervical HPV landscape: unmasking dominance of non-16/18 HR-HPV and tailoring prevention strategies at a large scale

    BackgroundPersistent HR-HPV causes cervical cancer, exhibiting geographic variance. Europe/Americas have higher HPV16/18 rates, while Asia/Africa predominantly have non-16/18 HR-HPV. This study in Fujian, Asia, explores non-16/18 HR-HPV infections, assessing their epidemiology and cervical lesion association for targeted prevention.MethodsA total of 101,621 women undergoing HPV screening at a hospital in Fujian Province from 2013 to 2019 were included. HPV genotyping was performed. A subset of 11,666 HPV-positive women with available histopathology results were analyzed to characterize HPV genotype distribution across cervical diagnoses.ResultsIn 101,621 samples, 24.5% tested positive for HPV. Among these samples, 17.3% exhibited single infections, while 7.2% showed evidence of multiple infections. The predominant non-16/18 high-risk HPV types identified were HPV 52, 58, 53, 51, and 81. Single HPV infections accounted for 64.1% of all HPV-positive cases, with 71.4% of these being non-16/18 high-risk HPV infections. Age-related variations were observed in 11,666 HPV-positive patients with pathological results. Cancer patients were older. In the cancer group, HPV52 (21.8%) and HPV58 (18.6%) were the predominant types, followed by HPV33, HPV31, and HPV53. Compared to single HPV16/18 infection, non-16/18 HPV predominated in LSIL. Adjusted odds ratios (OR) for LSIL were elevated: multiple HPV16/18 (OR 2.18), multiple non-16/18 HR-HPV (OR 2.53), and multiple LR-HPV (OR 2.38). Notably, solitary HPV16/18 conferred higher odds for HSIL and cancer.ConclusionOur large-scale analysis in Fujian Province highlights HPV 52, 58, 53, 51, and 81 as predominant non-16/18 HR-HPV types. Multiple HPV poses increased LSIL risks, while solitary HPV16/18 elevates HSIL and cancer odds. These findings stress tailored cervical cancer prevention, highlighting specific HPV impacts on lesion severity and guiding region-specific strategies for optimal screening in Asia, emphasizing ongoing surveillance in the vaccination era

    YY1 directly interacts with myocardin to repress the triad myocardin/SRF/CArG box-mediated smooth muscle gene transcription during smooth muscle phenotypic modulation

    Yin Yang 1 (YY1) regulates gene transcription in a variety of biological processes. In this study, we aim to determine the role of YY1 in vascular smooth muscle cell (VSMC) phenotypic modulation both in vivo and in vitro. Here we show that vascular injury in rodent carotid arteries induces YY1 expression along with reduced expression of smooth muscle differentiation markers in the carotids. Consistent with this finding, YY1 expression is induced in differentiated VSMCs in response to serum stimulation. To determine the underlying molecular mechanisms, we found that YY1 suppresses the transcription of CArG box-dependent SMC-specific genes including SM22α, SMα-actin and SMMHC. Interestingly, YY1 suppresses the transcriptional activity of the SM22α promoter by hindering the binding of serum response factor (SRF) to the proximal CArG box. YY1 also suppresses the transcription and the transactivation of myocardin (MYOCD), a master regulator for SMC-specific gene transcription by binding to SRF to form the MYOCD/SRF/CArG box triad (known as the ternary complex). Mechanistically, YY1 directly interacts with MYOCD to competitively displace MYOCD from SRF. This is the first evidence showing that YY1 inhibits SMC differentiation by directly targeting MYOCD. These findings provide new mechanistic insights into the regulatory mechanisms that govern SMC phenotypic modulation in the pathogenesis of vascular diseases

    Deglacial biogenic opal peaks revealing enhanced Southern Ocean upwelling during the last 513 ka

    Strength of Southern Ocean upwelling controls the exchange of carbon dioxide (CO2) between deep ocean reservoirs and atmosphere, as well as the communication of dissolved silicon with the euphotic zone of the Southern Ocean. The silicon supply could limit diatom opal productivity in the high-latitudes of Southern Ocean and the subsequent burial of biogenic opal in underlying sediments. Here we report a record of biogenic opal export off the Prydz Bay south of the polar front of the Southern Ocean, indicating strengthened upwelling during the past five glacial terminations. In all five terminations (Isingle bondV), opal peaks occur in line with Northern Hemisphere summer insolation intensity as well as the existing IRDs, indicating that freshwater injection associated with retreat of the Northern Hemisphere ice sheets could be the cause of enhanced upwelling in the Southern Ocean during terminations. This could in turn promote CO2 outgassing, finally accelerating the completion of the terminations. In addition, the enhanced upwelling could export the Si-rich deep water to low latitudes via Antarctic Intermediate Water (AAIW) and Subantarctic Mode Water (SAMW), potentially leading to deglacial opal peaks in subtropical North Atlantic

    Causal effect of gut microbiota on DNA methylation phenotypic age acceleration: a two-sample Mendelian randomization study

    Abstract The causal relationship between gut microbiota and DNA methylation phenotypic age acceleration remains unclear. This study aims to examine the causal effect of gut microbiota on the acceleration of DNA methylation phenotypic age using Mendelian randomization. A total of 212 gut microbiota were included in this study, and their 16S rRNA sequencing data were obtained from the Genome-wide Association Study (GWAS) database. The GWAS data corresponding to DNA methylation phenotypic age acceleration were selected as the outcome variable. Two-sample Mendelian randomization (TSMR) was conducted using R software. During the analysis process, careful consideration was given to address potential biases arising from linkage disequilibrium and weak instrumental variables. The results from inverse-variance weighting (IVW) analysis revealed significant associations (P < 0.05) between single nucleotide polymorphisms (SNPs) corresponding to 16 gut microbiota species and DNA methylation phenotypic age acceleration. Out of the total, 12 gut microbiota species exhibited consistent and robust causal effects. Among them, 7 displayed a significant positive correlation with the outcome while 5 species showed a significant negative correlation with the outcome.     Leaf Area Index Estimation Using Chinese GF-1 Wide Field View Data in an Agriculture Region

    Leaf area index (LAI) is an important vegetation parameter that characterizes leaf density and canopy structure, and plays an important role in global change study, land surface process simulation and agriculture monitoring. The wide field view (WFV) sensor on board the Chinese GF-1 satellite can acquire multi-spectral data with decametric spatial resolution, high temporal resolution and wide coverage, which are valuable data sources for dynamic monitoring of LAI. Therefore, an automatic LAI estimation algorithm for GF-1 WFV data was developed based on the radiative transfer model and LAI estimation accuracy of the developed algorithm was assessed in an agriculture region with maize as the dominated crop type. The radiative transfer model was firstly used to simulate the physical relationship between canopy reflectance and LAI under different soil and vegetation conditions, and then the training sample dataset was formed. Then, neural networks (NNs) were used to develop the LAI estimation algorithm using the training sample dataset. Green, red and near-infrared band reflectances of GF-1 WFV data were used as the input variables of the NNs, as well as the corresponding LAI was the output variable. The validation results using field LAI measurements in the agriculture region indicated that the LAI estimation algorithm could achieve satisfactory results (such as R2 = 0.818, RMSE = 0.50). In addition, the developed LAI estimation algorithm had potential to operationally generate LAI datasets using GF-1 WFV land surface reflectance data, which could provide high spatial and temporal resolution LAI data for agriculture, ecosystem and environmental management researches

    Preparation and Properties of Decellularized Sheep Kidney Derived Matrix Scaffolds

    Abstract Scaffolds from tissues or organs have nanoscale microstructures. Derived matrix scaffolds prepared by decellularized method can provide more cell attachment sites, which is conducive to cell adhesion, proliferation, differentiation and other physiological activities on scaffolds. In this study, the sheep kidney decellularized matrix scaffold was prepared by the method of decellularization. Due to the poor mechanical properties of the decellularized matrix, the cross linking method was adopted to enhance its mechanical properties. The decellularization efficiency of sheep renal matrix scaffolds was observed by scanning electron microscopy and histological staining, and the biocompatibility of the scaffolds was investigated by inoculating adipose derived stem cells. It was found that the scaffold had good decellularization effect and good pore structure

    Correlation between genetic alterations and clinicopathological features of papillary thyroid carcinomas

    Objective To investigate the correlations between multigene alterations and clinicopathological features in papillary thyroid carcinoma (PTC) samples. Methods In this retrospective study, 111 cytological specimens of thyroid nodules, including 74 PTC samples and 37 benign samples, were analyzed using a 22-gene mutation assay employing next-generation sequencing. Clinicopathological information was retrospectively collected and analyzed. Results Gene alterations were associated with a higher rate of lymph node metastasis (LNM) and thyroid capsular invasion, a lower rate of coexisting Hashimoto's thyroiditis, the classical PTC subtype, and younger age (<45 years). Among the 22 genes tested, the BRAF mutation rates showed a significant difference between the PTC and benign groups. In the subgroup analysis, younger age (odds ratio = 12.512, 95% confidence interval: 3.126–50.087) was an independent risk factor for LNM. In further analyses, BRAF mutation was significantly associated with LNM in the older subgroup (age ≥ 45 years), suggesting that the BRAF mutation test has greater value for determining PTC prognosis in the older age group. Conclusions Our findings will provide a more comprehensive understanding of the relationship between gene mutations and PTC and may contribute to improved PTC management

    High Expression Level of &alpha;2-3-Linked Sialic Acids on Salivary Glycoproteins of Breastfeeding Women May Help to Protect Them from Avian Influenza Virus Infection

    Terminal sialic acids (Sia) on soluble glycoprotein of saliva play an important role in the clearance of influenza virus. The aim of this study is to investigate the alteration of sialylation on the salivary proteins of women during the lactation period and its effect on the saliva binding ability to virus. In total, 210 saliva samples from postpartum women with and without breastfeeding were collected, and the expression level of &alpha;2-3/6-linked Sia on the whole salivary proteins and specific glycoproteins of IgA and MUC5B from different groups were tested and verified using lectin microarray, blotting analysis and ELISA based method. The H1N1 vaccine and three strains of Avian influenza virus (AIV) were used for the saliva binding assay. Results showed that the variation in salivary expression level of &alpha;2-3-linked Sia was much more obvious than the &alpha;2-6-linked Sia, which was up-regulated significantly in the breastfeeding groups compared to the non-breastfeeding groups at the same postpartum stage. Furthermore, the binding abilities of salivary glycoproteins to AIV strains and H1N1 vaccine were increased in breastfeeding groups accordingly. This finding adds new evidence for the maternal benefit of breastfeeding and provides new thinking to protect postpartum women from AIV infection
