    Young Engels Is Still Engels: A Response to Terrell Carver

    Terrel Carver assumes that existing biographies of Engels unfairly seek to explain his ideas in relation to Marx and seek to reconstruct the life journey, especially the ideological journey, of young Engels. He proposes two arguments: first, Engels\u27 ideological growth had no direct purpose or direction and was full of uncertainties; but also, second, that the young Engels\u27 theoretical achievements have been unduly underestimated as they were much more profound than Marx\u27s, such that Marx was the main beneficiary of their initial meeting. One problem with Carver’s argument is that his new insights are often not that new at all. His anti-teleological Engels is a liberal, but this is a more-or-less static form of liberalism that seems innocent of any immanent mechanism through which Engels’s trajectory toward revolutionary socialism might be understood. “Engels vs. Marx” is a trite trope. The proposition of “Engels vs. Marx” functions to deny the theory and practice of Marxist revolutionaries and politicians after Engels. This is something that the left academia should be vigilant about

    ShenQiWan ameliorates renal injury in type 2 diabetic mice by modulating mitochondrial fusion and endoplasmic reticulum stress

    Background: ShenQiWan is commonly used in traditional Chinese medicine for the treatment of diabetic nephropathy, which is closely related to mitochondrial fusion and endoplasmic reticulum stress. This study aimed to investigate the intervention effect and molecular mechanisms of ShenQiWan on renal injury in KKAy mice.Methods: C57BL/6J mice (11 weeks old) were fed a regular diet upon arrival, while KKAy mice (11 weeks old) were fed a high-fat diet upon arrival. At 12 weeks of age, KKAy mice with random blood glucose ≥13.9 mmol/L were identified as diabetic mice and randomly divided into the model group (n = 30) and the treatment group (n = 30), while C57BL/6J mice of 12 weeks old (n = 30) served as the control group. The treatment group received daily aqueous decoction of ShenQiWan (13.5 g/kg), while the control group and model group received daily equal amounts of saline from 12 weeks old to 24 weeks old. The general status of mice was observed regularly, and fasting blood glucose and 24-hour urine microalbumin were measured. Ten mice were euthanized in each group at the age of 16, 20, and 24 weeks, serum samples were used for biochemical indexes and kidney tissues were used for morphological studies. GRP78, OPA1, MFN1, MFN2 mRNA and protein expression were detected by Real-time PCR, immunohistochemistry and Western blot.Results: The mice in the model group exhibited symptoms of lethargy, slow movement, obesity, polyuria and proteinuria. Morphological observation revealed pathological changes, including thickening of the glomerular basement membrane and interstitial fibrosis. After treatment with ShenQiWan, the fasting blood glucose level of KKAy mice was significantly reduced, urinary albuminuria was decreased, serum biochemical indexes were improved, renal tissue pathological changes were significantly alleviated. The results also showed a significant reduction in the expression of endoplasmic reticulum stress-related factor GRP78 and an increase in the expression of mitochondrial fusion-related factors OPA1, MFN1 and MFN2 after treatment with ShenQiWan.Conclusion: ShenQiWan can protect diabetic mice from renal damage by modulating mitochondrial fusion and alleviating endoplasmic reticulum stress, exerting its protective effects

    New insight into the microbiome, resistome, and mobilome on the dental waste water in the context of heavy metal environment

    ObjectHospital sewage have been associated with incorporation of antibiotic resistance genes (ARGs) and mobile genetic elements (MGEs) into microbes, which is considered as a key indicator for the spread of antimicrobial resistance (AMR). The compositions of dental waste water (DWW) contain heavy metals, the evolution of AMR and its effects on the water environment in the context of heavy metal environment have not been seriously investigated. Thus, our major aims were to elucidate the evolution of AMR in DWW.MethodsDWW samples were collected from a major dental department. The presence of microbial communities, ARGs, and MGEs in untreated and treated (by filter membrane and ozone) samples were analyzed using metagenomics and bioinformatic methods.ResultsDWW-associated resistomes included 1,208 types of ARGs, belonging to 29 antibiotic types/subtypes. The most abundant types/subtypes were ARGs of multidrug resistance and of antibiotics that were frequently used in the clinical practice. Pseudomonas putida, Pseudomonas aeruginosa, Chryseobacterium indologenes, Sphingomonas laterariae were the main bacteria which hosted these ARGs. Mobilomes in DWW consisted of 93 MGE subtypes which belonged to 8 MGE types. Transposases were the most frequently detected MGEs which formed networks of communications. For example, ISCrsp1 and tnpA.5/4/11 were the main transposases located in the central hubs of a network. These significant associations between ARGs and MGEs revealed the strong potential of ARGs transmission towards development of antimicrobial-resistant (AMR) bacteria. On the other hand, treatment of DWW using membranes and ozone was only effective in removing minor species of bacteria and types of ARGs and MGEs.ConclusionDWW contained abundant ARGs, and MGEs, which contributed to the occurrence and spread of AMR bacteria. Consequently, DWW would seriously increase environmental health concerns which may be different but have been well-documented from hospital waste waters

    Retrospective evaluation of whole exome and genome mutation calls in 746 cancer samples

    Funder: NCI U24CA211006Abstract: The Cancer Genome Atlas (TCGA) and International Cancer Genome Consortium (ICGC) curated consensus somatic mutation calls using whole exome sequencing (WES) and whole genome sequencing (WGS), respectively. Here, as part of the ICGC/TCGA Pan-Cancer Analysis of Whole Genomes (PCAWG) Consortium, which aggregated whole genome sequencing data from 2,658 cancers across 38 tumour types, we compare WES and WGS side-by-side from 746 TCGA samples, finding that ~80% of mutations overlap in covered exonic regions. We estimate that low variant allele fraction (VAF < 15%) and clonal heterogeneity contribute up to 68% of private WGS mutations and 71% of private WES mutations. We observe that ~30% of private WGS mutations trace to mutations identified by a single variant caller in WES consensus efforts. WGS captures both ~50% more variation in exonic regions and un-observed mutations in loci with variable GC-content. Together, our analysis highlights technological divergences between two reproducible somatic variant detection efforts

    The Impact of Negative Informal Information Before a Change on Performance: A Within-Person Approach

    We live in a rapidly changing business environment where change has become the norm for organizations to maintain competitiveness. Although both scholars and practitioners agree that organizational change communication is important to help employees adjust to change, little is known about how negative informal information before the change affects employees&rsquo; reaction to the change and occurrence of possible within-person dynamics of resistance intention over time. Based on the construal-level theory, we used SPSS 22, AMOS 20, and HLM 6.0 as tools to explore how negative informal information affects individual performance. We used a multilevel approach to probe within-person processes among 215 MBA students in China. The results show that (1) negative informal information provided before the organizational change is positively related to the resistance intention, (2) resistance intention decreases significantly over time, and (3) negative informal information is negatively related to individual performance during the organizational change. The results from this study extend the literature on informal communication before the change and provide a dynamic perspective on the occurrence of possible within-person dynamics of resistance intention over time

    Spatiotemporal Variations of Extreme Precipitation in Wuling Mountain Area (China) and Their Connection to Potential Driving Factors

    Changes in extreme precipitation have become a significant issue of regional disaster risk assessment and water resources management. Extreme precipitation variability is affected by multiple factors and shows disparities across different regions. Especially in mountain areas, geographic feature and local characteristics put more complexity and uncertainty on the changes of precipitation extremes. In this study, ten extreme precipitation indices of Wuling Mountain Area (WMA) during 1960&ndash;2019 have been used to analyzed the spatiotemporal variations of precipitation extremes. The relationships between extreme precipitation and potential driving factors, including geographic factors, global warming, local temperature, and climate indices, were investigated via correlation analysis. The results indicated that extreme precipitation tends to have a shorter duration and stronger intensity in WMA. Decreasing trends in R10mm, R20mm, R25mm, and the consecutive wet days (CWD) series account for 92%, 68%, 52%, and 96% of stations, while most stations in WMA have rising trends in Rx1day (68%), SDII (64%), R95p (72%), and R99p (72%). Significant abrupt changes in extreme precipitation indices mainly occurred in the 1980s&ndash;1990s. Geographic factors, local temperature, and climate indices exert different impacts on extreme precipitation. Longitude and elevation instead of latitude significantly affect extreme precipitation indices except for the maximum duration of wet spells. Global warming is likely to increase the intensity and decrease the duration of extreme precipitation, while the influence of local temperature is not exactly the same as that of global warming. The study reveals that summer monsoon indices are the dominant climate factor for variations of precipitation extremes in WMA. The correlation coefficient between extreme precipitation indices (such as Rx1day, R95p, R99p) and the East Asian summer monsoon index is around 0.5 and passed the significant test at the 0.01 level. The weakening of the summer monsoon indices tends to bring extreme precipitation with stronger intensity. The findings provide more understanding of the drivers and reasons of extreme precipitation changes in the mountain area

    Effects of soy protein and β-conglycinin on microbiota and in vitro antioxidant and immunomodulatory capacities of human faecal cultures

    To clarify the effects of soy protein (SP) and β-conglycinin (BC) on human gut microbiota, human stool samples, collected from four healthy volunteers, were incubated without (control) or with 3% (w/v) SP and BC at 37 °C for 24 h under anaerobic conditions.During incubation, the amount of dissolved protein decreased and that of ammonia increased, although indole content was lower in both SP and BC cultures than in control cultures.The degraded protein band pattern, obtained by sodium dodecyl sulphate-polyacrylamide gel electrophoresis, varied across individuals.Total organic acids tended to increase in amount in SP and BC cultures, with n-butyric acid levels increasing significantly (p < 0.05) in the BC culture.The gut microbiota were analysed using 16S rRNA (V4) gene amplicon sequencing. Genera regarded as butyrate producers, such as Roseburia, Butyricicoccus, and Faecalibacterium, were high in both SP and BC cultures, and the genera and amplicon sequence variants of butyrate producers varied across individuals.The incubation with SP and BC increased the superoxide anion radical scavenging capacity and ferric reducing power of the faecal cultures. As an in vitro immunomodulatory activity, increased promotion of nitric oxide secretion in murine macrophage-like RAW264.7 cells was observed in the BC cultures.Overall, the effects of SP and BC were considered positive with respect to the host.公開日: 2023-02-1
