    Meta-Analysis: Contrast-Enhanced Ultrasound Versus Conventional Ultrasound for Differentiation of Benign and Malignant Breast Lesions

    This meta-analysis aimed to compare the diagnostic performance of contrast-enhanced ultrasound (CEUS), conventional ultrasound (US) combined with CEUS (US + CEUS) and US for distinguishing breast lesions. From thorough literature research, studies that compared the diagnostic performance of CEUS versus US or US + CEUS versus US, using pathology results as the gold standard, were included. A total of 10 studies were included, of which 9 compared the diagnostic performance of CEUS and US, and 5 studies compared US + CEUS and US. In those comparing CEUS versus US, the pooled sensitivity was 0.93 (95% CI: 0.91–0.95) versus 0.87 (95% CI: 0.85–0.90) and pooled specificity was 0.86 (95% CI: 0.84–0.88) versus 0.72 (95% CI: 0.69–0.75). In studies comparing US + CEUS versus US, the pooled sensitivity was 0.94 (95% CI: 0.92–0.96) versus 0.87 (95% CI: 0.84–0.90) and pooled specificity was 0.86 (95% CI: 0.82–0.89) versus 0.80 (95% CI: 0.76–0.84). In terms of diagnosing breast malignancy, areas under the curve of the summary receiver operating characteristic (of both CEUS (p = 0.003) and US + CEUS (p = 0.000) were statistically higher than that of US. Both CEUS alone and US + CEUS had better diagnostic performance than US in differentiation of breast lesions, and US + CEUS also had low negative likelihood ratio

    Spatiotemporal features of the soil moisture across Northwest China using remote sensing data, reanalysis data, and global hydrological model

    Soil moisture is an important factor affecting the change of land surface hydrological processes and the distribution of material and energy exchanges between the land and atmosphere and vegetation’s temporal and spatial distributions, especially in arid and semi-arid regions. This paper focuses on soil moisture features across Northwest China, the core region of the Silk Road Economic Belt. Six soil moisture datasets from the period 1981–2020 were employed, which included ERA5 (the European Centre for Medium-Range Weather Forecasts Atmospheric Reanalysis 5), ESA-CCI (European Space Agency Climate Change Initiative), GLDAS (Global Land Data Assimilation System), MERRA-2 (The Modern-Era Retrospective Analysis for Research and Applications, Version 2), RSSSM (A Remote Sensing-based global 10-day resolution Surface Soil Moisture dataset), and SSM-Feng (Regional multimodal fusion of surface soil moisture data in China). The temporal and spatial variation of the linear trend and abrupt change characteristics at seasonal and annual scale were explored. The results are as follows: 1) ESA-CCI, GLDAS, and MERRA-2 showed a slow increase in annual soil moisture tendency at a rate of less than 0.001 m3/m3/year, while ERA5 and SSM-Feng showed a significant decreasing linear trend at a rate of 1.31 × 10−4 m3/m3/year and 1.01 × 10−4 m3/m3/year (p < 0.05), respectively. 2) In autumn and winter, only GLDAS and MERRA-2 showed significant increasing trends. In the growing season (i.e., from April to October), the soil moisture of ESA-CCI, GLDAS, and MERRA-2 significantly increased at the rates of 3.29 × 10−4 m3/m3/year, 3.30 × 10−4 m3/m3/year, and 6.64 × 10−4 m3/m3/year (p < 0.05), respectively. 3) ERA5 and ESA-CCI have frequent abrupt changes in 1984, 1987, and 2006 for ERA5, 2010–2012 and 2019–2020 for ESA-CCI. 4) In terms of spatial variations, most datasets show that soil moisture has increased across most regions. The ERA5, ESA-CCI, GLDAS, MERRA-2, and SSM-Feng datasets show decreased soil moisture in the Tarim Basin. The conclusions of this study deepen the understanding of temporal and spatial variation in soil moisture in arid areas of Northwest China. Through these conclusions, a certain theoretical basis can be provided for the complex water cycle process in the arid region

    Characteristics and physical mechanisms of a rainstorm in Hotan, Xinjiang, China

    Owing to global warming, extreme precipitation events in the arid regions of Central Asia have increased, resulting in significant consequences for water resources and ecosystems. Hence, to address the features and corresponding physical mechanisms of these rainstorms, we examined the rainstorm that occurred in Hotan, Xinjiang in June 2021 as a case study. We employed multiple datasets, including meteorological stations, sounding observations, satellite precipitation data, and reanalysis datasets. The results indicate that the Global Precipitation Measurement satellite precipitation product accurately captured the temporal and spatial variations in this rainstorm, as verified against hourly in situ observation data. Some meteorological stations recorded values greater than twice their historical records, such as Luopu, Pishan, Moyu, and Hotan. Moreover, the duration of the precipitation was longer than 2 days. For the physical mechanisms of this rainstorm, the water vapor in this rainstorm is sourced from the 45°–65°N region of the North Atlantic Ocean crosses the Ural Mountains and the West Siberian Plain to southern Xinjiang. The low-pressure levels (e.g., 700 hPa and 850 hPa) have the more water vapor flux and specific humidity than the high-pressure levels. Our findings can aid the understanding of extreme precipitation events in Central Asia and provide a reference for dealing with meteorological disasters, including extreme precipitation, in the context of global climate change

    <p>Abstract</p> <p>Background</p> <p>The antiproliferative effect of the Hsp90 inhibitor 17-AAG (17-allylamino-17-demethoxygeldanamycin) on human retinal pigment epithelial cells is investigated.</p> <p>Methods</p> <p>MTT and flow cytometry were used to study the antiproliferative effects of the 17-AAG treatment of ARPE-19 cells. 2D gel electrophoresis (2-DE) and mass spectrometry were applied to detect the altered expression of proteins, which was verified by real-time PCR. Gene Ontology analysis and Ingenuity Pathway Analysis (IPA) were utilized to analyze the signaling pathways, cellular location, function, and network connections of the identified proteins. And SOD assay was employed to confirm the analysis.</p> <p>Results</p> <p>17-AAG suppressed the proliferation of ARPE-19 cells by inducing cell cycle arrest and apoptosis. Proteomic analysis revealed that the expression of 94 proteins was altered by a factor of more than 1.5 following exposure to 17-AAG. Of these 94, 87 proteins were identified. Real-time PCR results indicated that Hsp90 and Hsp70, which were not identified by proteomic analysis, were both upregulated upon 17-AAG treatment. IPA revealed that most of the proteins have functions that are related to oxidative stress, as verified by SOD assay, while canonical pathway analysis revealed glycolysis/gluconeogenesis.</p> <p>Conclusions</p> <p>17-AAG suppressed the proliferation of ARPE-19 cells by inducing cell cycle arrest and apoptosis, and possibly by oxidative stress.</p

    Timing Is Critical for an Effective Anti-Metastatic Immunotherapy: The Decisive Role of IFNγ/STAT1-Mediated Activation of Autophagy

    BACKGROUND: Immunotherapy is often recommended as an adjuvant treatment to reduce the chance of cancer recurrence or metastasis. Interestingly, timing is very important for a successful immunotherapy against metastasis, although the precise mechanism is still unknown. METHODS AND FINDINGS: Using a mouse model of melanoma metastasis induced by intravenous injection of B16-F10 cells, we investigated the mechanism responsible for the diverse efficacy of the prophylactic or therapeutic TLR4 and TLR9 agonist complex against metastasis. We found that the activation of TLR4 and TLR9 prevented, but did not reverse, metastasis because the potency of this combination was neither sufficient to overcome the tumor cell-educated immune tolerance nor to induce efficacious autophagy in tumor cells. The prophylactic application of the complex promoted antimetastatic immunity, leading to the autophagy-associated death of melanoma cells via IFNγ/STAT1 activation and attenuated tumor metastasis. IFNγ neutralization reversed the prophylactic benefit induced by the complex by suppressing STAT1 activation and attenuating autophagy in mice. However, the therapeutic application of the complex did not suppress metastasis because the complex could not reverse tumor cell-induced STAT3 activation and neither activate IFNγ/STAT1 signaling and autophagy. Suppressing STAT3 activation with the JAK/STAT antagonist AG490 restored the antimetastatic effect of the TLR4/9 agonist complex. Activation of autophagy after tumor inoculation by using rapamycin, with or without the TLR4/9 agonist complex, could suppress metastasis. CONCLUSION AND SIGNIFICANCE: Our studies suggest that activation of IFNγ/STAT1 signaling and induction of autophagy are critical for an efficacious anti-metastatic immunotherapy and that autophagy activators may overcome the timing barrier for immunotherapy against metastasis

    The second King’s College London Symposium on Ageing and Long-term Care in China was convened from 4 to 5th July 2019 at King’s College London in London. The aim of the Symposium was to have a better understanding of health and social challenges for aging and long-term care in China. This symposium draws research insights from a wide range of disciplines, including economics, public policy, demography, gerontology, public health and sociology. A total of 20 participants from eight countries, seek to identify the key issues and research priorities in the area of aging and long-term care in China. The results published here are a synthesis of the top four research areas that represent the perspectives from some of the leading researchers in the field. © The Author(s) 2020
