    Malondialdehyde level and some enzymatic activities in subclinical mastitis milk

    The purpose of this study was to evaluate the changes occurring in milk malondialdehyde (MDA) level and some enzymatic activities as a result of subclinical mastitis (SCM) in dairy cows. A total of 124 milk samples were collected from 124 lactating cows from the same herd in the period between the 2nd week after calving and the 10th week postpartum. They were classified by bacterial culture and the California mastitis test (CMT) as positive were deemed to have glands with SCM, and the periodic incidence rate of SCM was 26.6%. The most common bacterial isolates from SCM cases were Staphylococcus aureus (47%) and coagulase negative Staphylococci (CNS) (27%). The mean level of MDA and activities of lactate dehydrogenase (LDH) and alkaline phosphatase (ALP) were significantly higher in SCM milk than in normal milk, while the mean activity of glutathione peroxidase (GPx) was significantly lower in SCM milk than in normal milk. There were no differences in the activities of superoxide dismutase (SOD) and aspartate aminotransferase (AST) between normal milk and SCM milk. Therefore, the measurement of milk MDA level and GPx, LDH and ALP activities, appears to be a suitable diagnostic method for identifying SCM in dairy cows.Key words: Subclinical mastitis, mastitis diagnostic, etiology, malonaldehyde (MDA), enzym

    Undrained cyclic response of K-0-consolidated stiff cretaceous clay under wheel loading conditions

    Optimal whole life design for railways, highways, runways, and metro lines requires an accurate assessment of how their underlying geomaterials respond to large numbers of wheel-loading cycles. This paper presents an experimental study on a natural UK stiff clay with a cyclic triaxial (CT) and hollow cylinder apparatus (CHCA) that imposed K0 and wheel-loading stress conditions. The focus is on Gault clay, a high overconsolidation ratio (OCR) marine clay deposited in the Cretaceous, whose mechanical behavior is significantly anisotropic and in situ K0 values exceed unity. The clay outcrops under sections of most major highways radiating out of London, as well as the HS1 and new HS2 high-speed railways. The experimental investigation explored how the principal stress rotation implicit in wheel loading increases the magnitudes and changes the sign of vertical strain accumulation, as well as accelerating resilient modulus degradation and accentuating stress–strain hysteresis, all of which affect pavement or rail-track serviceability. The clay’s deformation and pore pressure responses are categorized into stable, metastable, and unstable patterns. Comparisons with related studies on low OCR, low K0 soft clay from Wenzhou in southeastern China, confirm the Gault clay’s generally stiffer prefailure behavior and different cyclic response. The stiff clay’s greater brittleness is also emphasized; particle reorientation occurs readily along distinct shear bands, leading to dramatic shear strength reductions that have a major impact on slope and foundation stability and call for appropriate caution in practical design

    Evidence for genetic association of TBX21 and IFNG with systemic lupus erythematosus in a Chinese Han population

    TBX21 recode T-bet which is an important transcription factor that drives the Th1 immune response primarily by promoting expression of the interferon-gamma (IFNG) gene. Recent studies have shown that genetic variants in TBX21 and IFNG are connected with risk of systemic lupus erythematosus (SLE). The aim of the present study was to replicate these genetic associations with SLE in Anhui Chinese population. Genotyping of 3 variants (rs4794067 in TBX21, rs2069705 and rs2069718 in IFNG) was performed. A total of 3732 subjects were included in the final analysis. The study only identified the association of rs2069705 with SLE susceptibility (T vs. C: odds ratio [OR] = 1.12, 95% confidence interval [CI] = 1.00-1.26, P = 0.046). Combined analysis with Hong Kong GWAS showed that the OR for rs2069705 was 1.10 (95% CI: 1.01-1.21, P = 0.027). Further pooled analysis with Korean populations involving 10498 subjects showed a more significant association between rs2069705 and SLE (T vs. C: OR = 1.11, 95%CI = 1.04-1.19, P = 0.002; TT + TC vs. CC: OR = 1.11, 95%CI = 1.02-1.21, P = 0.012; TT vs. TC + CC: OR = 1.28, 95%CI = 1.07-1.54, P = 0.008; TT vs. CC: OR = 1.33, 95%CI = 1.10-1.60, P = 0.003). In addition, we also identified a significant genetic interaction between rs2069705 and rs4794067 in Anhui Chinese population. Our study suggests that IFNG and IFNG-TBX21 interaction are involved in SLE susceptibility.published_or_final_versio

But, the development of simple and effective methods for creating novel assemblies of self-supported patterns of hierarchical architectures to designed materials using a suitable chemical method is important to technology and remains an attractive, but elusive goal.</p> <p>Results</p> <p>The hierarchical structure of Mn<sub>3</sub>O<sub>4 </sub>with radiated spherulitic nanorods was prepared via a simple solution-based coordinated route in the presence of macrocycle polyamine, hexamethyl-1,4,8,11-tetraazacyclotetradeca-4,11-diene (CT) with the assistance of thiourea as an additive.</p> <p>Conclusion</p> <p>This approach opens a new and facile route for the morphogenesis of Mn<sub>3</sub>O<sub>4 </sub>material and it might be extended as a novel synthetic method for the synthesis of other inorganic semiconducting nanomaterials such as metal chalcogenide semiconductors with novel morphology and complex form, since it has been shown that thiourea can be used as an effective additive and the number of such water-soluble macrocyclic polyamines also makes it possible to provide various kinds of ligands for different metals in homogeneous water system.</p

    Identification of Potential Sites for Tryptophan Oxidation in Recombinant Antibodies Using tert-Butylhydroperoxide and Quantitative LC-MS

    Amino acid oxidation is known to affect the structure, activity, and rate of degradation of proteins. Methionine oxidation is one of the several chemical degradation pathways for recombinant antibodies. In this study, we have identified for the first time a solvent accessible tryptophan residue (Trp-32) in the complementary-determining region (CDR) of a recombinant IgG1 antibody susceptible to oxidation under real-time storage and elevated temperature conditions. The degree of light chain Trp-32 oxidation was found to be higher than the oxidation level of the conserved heavy chain Met-429 and the heavy chain Met-107 of the recombinant IgG1 antibody HER2, which have already been identified as being solvent accessible and sensitive to chemical oxidation. In order to reduce the time for simultaneous identification and functional evaluation of potential methionine and tryptophan oxidation sites, a test system employing tert-butylhydroperoxide (TBHP) and quantitative LC-MS was developed. The optimized oxidizing conditions allowed us to specifically oxidize the solvent accessible methionine and tryptophan residues that displayed significant oxidation in the real-time stability and elevated temperature study. The achieved degree of tryptophan oxidation was adequate to identify the functional consequence of the tryptophan oxidation by binding studies. In summary, the here presented approach of employing TBHP as oxidizing reagent combined with quantitative LC-MS and binding studies greatly facilitates the efficient identification and functional evaluation of methionine and tryptophan oxidation sites in the CDR of recombinant antibodies

    One-step fabrication of biocompatible chitosan-coated ZnS and ZnS:Mn2+ quantum dots via a γ-radiation route

    Biocompatible chitosan-coated ZnS quantum dots [CS-ZnS QDs] and chitosan-coated ZnS:Mn2+ quantum dots [CS-ZnS:Mn2+ QDs] were successfully fabricated via a convenient one-step γ-radiation route. The as-obtained QDs were around 5 nm in diameter with excellent water-solubility. These QDs emitting strong visible blue or orange light under UV excitation were successfully used as labels for PANC-1 cells. The cell experiments revealed that CS-ZnS and CS-ZnS:Mn2+ QDs showed low cytotoxicity and good biocompatibility, which offered possibilities for further biomedical applications. Moreover, this convenient synthesis strategy could be extended to fabricate other nanoparticles coated with chitosan

    Synthesis and Characterization of ZnO Nanorods and Nanodisks from Zinc Chloride Aqueous Solution

    ZnO nanorods and nanodisks were synthesized by solution process using zinc chloride as starting material. The morphology of ZnO crystal changed greatly depending on the concentrations of Zn2+ion and ethylene glycohol (EG) additive in the solution. The effect of thermal treatment on the morphology was investigated. Photocatalytic activities of plate-like Zn5(OH)8Cl2 · H2O and rod-like ZnO were characterized.
