    Antioxidant Activity of Sulfated Seaweeds Polysaccharides by Novel Assisted Extraction

    Seaweeds have an extremely numerous of species in the world and been able to be divided into several developmental systems. Broadly, three types of seaweeds can be defined according to their color: brown seaweeds, green seaweeds, and red seaweeds. Thousands of years ago, mankind used seaweeds as food and medicine. Seaweed extracts are gaining increasing attention due to their unique composition and the potential for widespread use in industry. A variety of novel (green) extraction techniques have been devised for converting seaweed biomass into seaweed extracts, such as enzyme-assisted extraction (EAE), microwave-assisted extraction (MAE), pressurized liquid extraction (PLE), supercritical fluid extraction (SFE), and ultrasound-assisted extraction (UAE), which are capable of extracting seaweeds’ biologically active compounds without causing degradation. Seaweed extracts contain compounds, such as carbohydrates, proteins, minerals, oils, fats, and polyunsaturated fatty acids and abundant bioactive compounds, such as antioxidants, pigments, and sulfated seaweed polysaccharides (SWP), as well as antibacterial, antifungal, anti-inflammatory, antioxidation, antitumor, antiviral, and hypolipidemic effect.     The study on SFLAB GanedenBC30 viability on baking products during storage

    AbstractFor understanding Bacillus coagulans, GanedenBC30 was used in different ways to added in raw dough and examine their viability after baking. Eight different baking products: (1) chrysanthemum cookies, (2) egg pastry cakes, (3) mooncakes, (4) muffins, (5) polo breads, (6) soda cookies, (7) sponge cakes, and (8) toasts were made from 0.5% GanedenBC30 added to their dough in two ways: (a) flour powder or (b) egg yolk. Then the (a) pH value, (b) titratable acidity, (c) GanedenBC30 counts, and (d) viability GanedenBC30 of eight different baking products were determined after storing at 4oC for 0, 3, 6, 9, 12, 15 days, or 25oC for 0, 3, 6 days. The eight types of raw dough had relatively lower pH values and rise after baking. The titratable acidity of the eight types of dough was relatively higher, and declined after baking. However, the pH value and titratable acidity of the eight baking products remained the same after 9 days at 4oC. On the other hand, the GanedenBC30 counts in the eight baking products were less than their raw dough GanedenBC30 levels. For storage at both 4 and 25oC, the results show the GanedenBC30 viability of baking products decreased with storage days. The dough made by flour powder and baking showed higher GanedenBC30 viability than by egg yolk. GanedenBC30 are good candidates for baking product use, both in lactic acid production and probiotic preparations

    A structural analysis of in vitro catalytic activities of hammerhead ribozymes

    <p>Abstract</p> <p>Background</p> <p>Ribozymes are small catalytic RNAs that possess the dual functions of sequence-specific RNA recognition and site-specific cleavage. <it>Trans</it>-cleaving ribozymes can inhibit translation of genes at the messenger RNA (mRNA) level in both eukaryotic and prokaryotic systems and are thus useful tools for studies of gene function. However, identification of target sites for efficient cleavage poses a challenge. Here, we have considered a number of structural and thermodynamic parameters that can affect the efficiency of target cleavage, in an attempt to identify rules for the selection of functional ribozymes.</p> <p>Results</p> <p>We employed the Sfold program for RNA secondary structure prediction, to account for the likely population of target structures that co-exist in dynamic equilibrium for a specific mRNA molecule. We designed and prepared 15 hammerhead ribozymes to target GUC cleavage sites in the mRNA of the breast cancer resistance protein (BCRP). These ribozymes were tested, and their catalytic activities were measured <it>in vitro</it>. We found that target disruption energy owing to the alteration of the local target structure necessary for ribozyme binding, and the total energy change of the ribozyme-target hybridization, are two significant parameters for prediction of ribozyme activity. Importantly, target disruption energy is the major contributor to the predictability of ribozyme activity by the total energy change. Furthermore, for a target-site specific ribozyme, incorrect folding of the catalytic core, or interactions involving the two binding arms and the end sequences of the catalytic core, can have detrimental effects on ribozyme activity.</p> <p>Conclusion</p> <p>The findings from this study suggest rules for structure-based rational design of <it>trans</it>-cleaving hammerhead ribozymes in gene knockdown studies.     The Role of lncRNAs in the Distant Metastasis of Breast Cancer

    Breast cancer (BC) remains the most frequently diagnosed cancer worldwide. Among breast cancer patients, distant metastasis and invasion is the leading cause of BC related death. Recently, long non-coding RNAs (lncRNAs), which used to be considered a genetic byproduct (owing to their unknown biological function), have been reported to be highly implicated in the development and progression of BC. In this review, we produce a summary of the functions and mechanisms of lncRNAs implicated in the different distant metastases of BC. The functions of lncRNAs have been divided into two types: oncogenic type and tumor suppressor. Furthermore, the majority of them exert their roles through the regulation of invasion, migration, epithelial—mesenchymal transition (EMT), and the metastasis process. In the final part, we briefly addressed future research prospects of lncRNAs, especially the testing methods through which to detect lncRNAs in the clinical work, and introduced several different tools with which to detect lncRNAs more conveniently. Although lncRNA research is still in the initial stages, it is a promising prognosticator and a novel therapeutic target for BC metastasis, which requires more research in the future

    Near-Surface Attenuation and Velocity Structures in Taiwan from Wellhead and Borehole Recordings Comparisons

    By analyzing the data from 28 seismic borehole stations deployed by the Central Weather Bureau Seismic Network throughout Taiwan from 2007 to 2014, we estimated the near-surface velocity (Vp and Vs) and attenuation (Qp and Qs) structures from surface to depths of approximately 300 m. To ensure that the deeper recordings were on the ray path between the seismic source and upper receiver, only events with an incidence angle of less than 35° were selected. Local magnitudes of analyzed events were between 1.1 and 6.6. The subsurface Qp and Qs were well modeled in the 5 - 40 Hz frequency band using the spectral ratio of direct P- and S-waves, respectively, at each station, under frequency-independent Q and ω2 source model assumptions. The estimated Vp in the Coastal Plain, the Western Foothills, the Longitudinal Valley, and the Yilan Plain were approximately 1000 - 2000 m s-1, which was lower than the Vp of 2500 - 4000 m s-1 in the Central Mountain Range. In addition, the Vs in the plain areas were lower than that in the Central Mountain Range. The low Vp and Vs and high Vp/Vs ratio in the Coastal Plain and the Western Foothills can be attributed to the unconsolidated soil and high pore-fluid content of subsurface sediments in the plain areas. In contrast to the velocity distribution, low Qp and Qs were observed in the Central Mountain Range. The low Qp and Qs with low Vp/Vs and low Qs/Qp ratios in the Central Mountain Range was consistent with the high thermal temperature observed in the field investigation. The obtained velocity and attenuation structures near surface could also provide important constraints in validation of the crustal structure of Taiwan
