    Study On The Character Of Indoor Air In House With Displacement Ventilation System

    AbstractApplying the method of computational fluid dynamics, velocity and temperature fields of three-dimensional displacement ventilation system with double heat sources are numerically simulated. Comparisons of computational results with experimental data are made. The velocity and temperature distribution of indoor air in houses with displacement ventilation system with single and double heat sources were studied. Two different cases of heat source are analyzed and compared. The results show that there are three layers in vertical temperature fields of displacement ventilation system with single or double heat sources, and the vertical temperature distribution of single heat sources is different from that of double heat sources. Furthermore, under the condition of two heat sources, the displacement ventilation parameters can’t be simply computed according to single heat sources inlet parameters, therefore the interaction between heat sources should be considered

    Inactivation of the fliY gene encoding a flagellar motor switch protein attenuates mobility and virulence of Leptospira interrogans strain Lai

    <p>Abstract</p> <p>Background</p> <p>Pathogenic <it>Leptospira </it>species cause leptospirosis, a zoonotic disease of global importance. The spirochete displays active rotative mobility which may contribute to invasion and diffusion of the pathogen in hosts. FliY is a flagellar motor switch protein that controls flagellar motor direction in other microbes, but its role in <it>Leptospira</it>, and paricularly in pathogenicity remains unknown.</p> <p>Results</p> <p>A suicide plasmid for the <it>fliY </it>gene of <it>Leptospira interrogans </it>serogroup Icterohaemorrhagiae serovar Lai strain Lai that was disrupted by inserting the ampicillin resistance gene (<it>bla</it>) was constructed, and the inactivation of <it>fliY </it>gene in a mutant (<it>fliY</it><sup>-</sup>) was confirmed by PCR and Western Blot analysis. The inactivation resulted in the mRNA absence of <it>fliP </it>and <it>fliQ </it>genes which are located downstream of the <it>fliY </it>gene in the same operon. The mutant displayed visibly weakened rotative motion in liquid medium and its migration on semisolid medium was also markedly attenuated compared to the wild-type strain. Compared to the wild-type strain, the mutant showed much lower levels of adhesion to murine macrophages and apoptosis-inducing ability, and its lethality to guinea pigs was also significantly decreased.</p> <p>Conclusion</p> <p>Inactivation of <it>fliY</it>, by the method used in this paper, clearly had polar effects on downstream genes. The phentotypes observed, including lower pathogenicity, could be a consequence of <it>fliY </it>inactivation, but also a consequence of the polar effects.</p

    Expression and aberrant promoter methylation of Wnt inhibitory factor-1 in human astrocytomas

    <p>Abstract</p> <p>Background</p> <p>Wnt inhibitory factor-1(WIF-1) acts as a Wnt-antagonists and tumor suppressor, but hypermethylation of WIF-1 gene promoter and low expression activate Wnt signaling aberrantly and induce the development of various human tumors. With this work we intended to investigate the expression and promoter methylation status of WIF-1 gene in human astrocytomas.</p> <p>Methods</p> <p>The tissue samples consisted of 53 astrocytomas and 6 normal brain tissues. The expression levels of WIF-1 were determined by immunohistochemistry and semiquantitative RT-PCR. The results were analyzed in correlation with clinicopathological data. Methylation status of WIF-1 gene promoter was investigated using methylation specific PCR. The relationship between methylation and expression of the genes was analyzed.</p> <p>Results</p> <p>The average expression levels of WIF-1 protein and mRNA in astrocytomas were decreased significantly compared with normal control tissues. The protein and mRNA expression of WIF-1 gene in astrocytomas was decreased with the increase of pathological grade. Furthermore, WIF-1 promoter methylation was observed by MS-PCR in astrocytomas which showed significant reduction of WIF-1 expression. The WIF-1 promoter hypermethylation was associated with reduced expression of WIF-1 expression.</p> <p>Conclusion</p> <p>Our results demonstrate that the WIF-1 gene is frequently down-regulated or silenced in astrocytomas by aberrant promoter methylation. This may be an important mechanism in astrocytoma carcinogenesis.</p

    Misuse of Statistical Methods in 10 Leading Chinese Medical Journals in 1998 and 2008

    Statistical methods are vital to biomedical research. Our aim was to find out whether progress has been made in the last decade in the use of statistical methods in Chinese medical research. We reviewed 10 leading Chinese medical journals published in 1998 and in 2008. Regarding statistical methods, using a multiple t-test for multiple group comparison was the most common error in the t-test in both years, which significantly decreased in 2008. In contingency tables, no significant level adjustment for multiple comparison significantly decreased in 2008. In ANOVA, over a quarter of articles misused the method of multiple pair-wise comparison in both years, and no significant difference was seen between the two years. In the rank transformation nonparametric test, the error of using multiple pair-wise comparison for multiple group comparison became less common. Many mistakes were found in the randomised controlled trial (56.3% in 1998; 67.9% in 2008), non- randomised clinical trial (57.3%; 58.6%), basic science study (72.9%; 65.5%), case study or case series study (48.4%; 47.2%), and cross-sectional study (57.1%; 44.2%). Progress has been made in the use of statistical methods in Chinese medical journals, but much is yet to be done

    Lifespan profiles of Alzheimer's disease–associated genes and their products in monkeys and mice.

    Alzheimer's disease (AD) is characterized by plaques of amyloid–beta (Aβ) peptide, cleaved from amyloid–β precursor protein (AβPP). Our hypothesis is that lifespan profiles of AD-associated mRNA and protein levels in monkeys would differ from mice, and that differential lifespan expression profiles would be useful to understand human AD pathogenesis. We compared profiles of AβPP mRNA, AβPP protein, and Aβ levels in rodents and primates. We also tracked a transcriptional regulator of the AβPP gene, specificity protein 1 (SP1), and the β amyloid precursor cleaving enzyme (BACE1). In mice, AβPP and Sp1 mRNA and their protein products were elevated late in life; Aβ levels declined in old age. In monkeys, Sp1, AβPP, and BACE1 mRNA declined in old age, while protein products and Aβ levels rose. Proteolytic processing in both species did not match production of Aβ. In primates, AβPP and Sp1 mRNA levels coordinate, but an inverse relationship exists with corresponding protein products, as well as Aβ levels. Comparison of human DNA and mRNA sequences to monkey and mouse counterparts revealed structural features that may explain differences in transcriptional and translational processing. These findings are important for selecting appropriate models for AD and other age–related diseases

    Nanoscale Alloying in Electrocatalysts

    In electrochemical energy conversion and storage, existing catalysts often contain a high percentage of noble metals such as Pt and Pd. In order to develop low-cost electrocatalysts, one of the effective strategies involves alloying noble metals with other transition metals. This strategy promises not only significant reduction of noble metals but also the tunability for enhanced catalytic activity and stability in comparison with conventional catalysts. In this report, some of the recent approaches to developing alloy catalysts for electrocatalytic oxygen reduction reaction in fuel cells will be highlighted. Selected examples will be also discussed to highlight insights into the structural and electrocatalytic properties of nanoalloy catalysts, which have implications for the design of low-cost, active, and durable catalysts for electrochemical energy production and conversion reactions
