    Bioethanol from Germinated Grains.

    The most well-known way to produce bioethanol is by the enzymatic hydrolysis and fermentation of starch. In a new project “BioConcens” (2007) sponsored by DARCOF (DAnish Research Center for Organic Food and farming) one aim is to develop a combined ethanol and biogas production for use in organic farming using starch containing biomass. Natural enzymes from cereals will be used for hydrolysis of starch to glucose in accordance with technology in brewing technology. Commercial enzymes are often produced from gene-modified organisms and will therefore not be used in the suggested organic context or process. A preliminary study was performed in which grains of wheat, rye, and barley were germinated using traditional methods applied in malting for beer production. During malting the amylase enzymes present in the grain are activated (autoamylolytic effect). Three steps were applied in the malting process; steeping, germination, and drying of the grains. After malting the grains were milled and mixed with water to 13% DM, cooked at 57.5C for 2 hours (to activate the enzymes), and cooled to 30C before adding Bakers Yeast. The results of this study indicate that efficient hydrolysis of starch can be achieved by activation of autoamylolytic enzymes in cereal grains after a malting process. The ethanol yields obtained in the autoamylolytic hydrolysis was comparable (or slightly higher) to that of reference experiments using commercial enzymes (amylases). The highest ethanol yield was obtained with wheat (0.34 g/g DM grain), followed by barley (0.31 g/g DM grain), and rye (0.29 g/g DM grain)

    Inhibition of bacterial adhesion to HT-29 cells by lipoteichoic acid extracted from Clostridium butyricum

    The aim of this experiment was to study the effect of the lipoteichoic acid (LTA) extracted from Clostridium butyricum on the adhesion of C. butyricum and Escherichia coli to HT-29 human intestinal cells. The method of extraction of lipoteichoic acid form C. butyricum by TX114 was evaluated. The purification of the LTA by DEAE-cellulose 52 anion exchange chromatography was also investigated. In addition, the LTA was assayed for its inhibition of the adhesion of C. butyricum and E. coli to HT-29 cells and antimicrobial activity. Our results showed that LTA could be extracted by TX114 and purified by DEAE-cellulose 52 anion exchange chromatography and could inhibit the adhesion of C. butyricum and E. coli to HT-29 cells. This result also revealed that the LTA from C. butyricum could inhibit the adhesion of C. butyricum and E. coli to intestinal cells.Key words: Lipoteichoic acid, Clostridium butyricum, HT-29 cells, adhesion, Escherichia col

    Diversity of methanogens in the hindgut of grower and finisher pigs

    This study examined the diversity of the methanogens in the hindgut of two different weight groups of pigs and correlated it with the amount of digested organic carbon (OC) and various components of dietary fiber. Five grower (58.9 ± 1.15 kg) and five finisher (89.4 ± 0.85 kg) Duroc × Landrace × Large Yorkshire female pigs were allocated into two groups and individually housed in cages. During the experiment, feed intake and fecal output were recorded for determination of apparent digestibility of OC, crude fiber (CF), neutral detergent fiber (NDF) and acid detergent fiber (ADF). At the end of the digestibility trial, pigs were sacrificed, and the contents of five segments of hindgut were sterilely collected to determine diversity of methanogens. Total microbial DNA of the hindgut contents was used as template for amplification of the methanogen16S rRNA gene, and the PCR products were further subjected to denaturing gradient gel electrophoresis (DGGE) analysis. Results show that the number of DGGE bands and Shannon diversity index for the 90 kg pigs were higher (P<0.05) than those for the 60 kg pigs. Methanogen communities did not alter along the different segments of the hindgut for the two weight groups. In addition, the amount of OC, CF, NDF and ADF digested (g/d) for the 90 kg pigs (1018.77, 23.11, 268.86 and 99.16, respectively) was higher (P<0.05) than the respective values for the 60 kg pigs (669.27, 13.77, 222.31 and 69.07), indicating that the higher diversity of  methanogens in the former group was related to the higher quantity of fiber materials fermented in the hindgut. The positive correlation (p<0.05) between number of DGGE bands and Shannon diversity index with quantity of digested OC and ADF further reaffirmed the above suggestion.Key words: Methanogen, pig, Shannon diversity index, polymerase chain reaction-denaturing gradient gel electrophoresis (PCR-DGGE)

    β-Catenin/Tcf-4 signaling pathway plays important roles in colorectal tumorigenesis. RT-PCR, western blotting and immunoprecipitation were used to study the effects of curcumin on β-catenin/Tcf-4 signaling pathway in HT-29 cells. Treatment of curcumin could induce cleavage of β-catenin and the cleavage could be inhibited by caspase inhibitors. The association of β-catenin with Tcf-4 in nucleus could be inhibited by curcumin. The expression of c-myc and cyclinD1 was downregulated by curcumin, which could not be blocked by Z-DEVD-FMK. The results showed curcumin could induce thecleavage of β-catenin by activition of caspases and downregulate the activity of β-catenin/Tcf signaling pathway independent of the caspases in HT-29 cells

    Orbital superfluidity in the PP-band of a bipartite optical square lattice

    The successful emulation of the Hubbard model in optical lattices has stimulated world wide efforts to extend their scope to also capture more complex, incompletely understood scenarios of many-body physics. Unfortunately, for bosons, Feynmans fundamental "no-node" theorem under very general circumstances predicts a positive definite ground state wave function with limited relevance for many-body systems of interest. A promising way around Feynmans statement is to consider higher bands in optical lattices with more than one dimension, where the orbital degree of freedom with its intrinsic anisotropy due to multiple orbital orientations gives rise to a structural diversity, highly relevant, for example, in the area of strongly correlated electronic matter. In homogeneous two-dimensional optical lattices, lifetimes of excited bands on the order of a hundred milliseconds are possible but the tunneling dynamics appears not to support cross-dimensional coherence. Here we report the first observation of a superfluid in the PP-band of a bipartite optical square lattice with SS-orbits and PP-orbits arranged in a chequerboard pattern. This permits us to establish full cross-dimensional coherence with a life-time of several ten milliseconds. Depending on a small adjustable anisotropy of the lattice, we can realize real-valued striped superfluid order parameters with different orientations Px±PyP_x \pm P_y or a complex-valued Px±iPyP_x \pm i P_y order parameter, which breaks time reversal symmetry and resembles the π\pi-flux model proposed in the context of high temperature superconductors. Our experiment opens up the realms of orbital superfluids to investigations with optical lattice models.Comment: 5 pages, 5 figure

    Immunomodulatory Potential of Patchouli Alcohol Isolated from Pogostemon cablin (Blanco) Benth (Lamiaceae) in Mice

    Purpose: To isolate and purify patchouli alcohol (PA), a tricyclic sesquiterpene constituent of Pogostemon cablin, and investigate its immunomodulatory potential in Kunming mice.Methods: PA was prepared from an ethanol aqueous extract of P. cablin by silica gel column chromatography, and further purified by crystallization using n-hexane. Purity was assessed by analytical gas chromatography (GC) and confirmation of chemical structure performed by Fourier transform infrared (FTIR), nuclear magnetic resonance (NMR) spectroscopy and mass spectrometry (MS). The effect of PA from Pogostemon cablin on immunological function was studied by macrophage phagocytosis, immune organ index, serum immunoglobulin level and delayed type  hypersensitivity (DTH) in mice that were administered orally doses of 20, 40 and 80 mg/kg.Results: The purity of PA was 99.3%. The oral administration of PA (40, or 80 mg/kg body weight) significantly increased the phagocytic index (p < 0.05), compared with prednisone acetate (PR) group. Administration of PA (80 mg/kg) boosted the production of circulating serum IgM (0.081 ± 0.010) and IgG (1.296 ± 0.120), while IgM and IgG in PR group was 0.069 ± 0.011 (p < 0.01) and 1.180 ± 0.070 (p < 0.01) respectively. However, PA (20 mg/kg) treatment elicited significant decrease in DTH induced by 2, 4-dinitro-chlorobenzene (DNCB) in mice (1.03 ± 0.40, p < 0.05), in comparison to DNCB-induced group (1.67 ± 0.84 mg).Conclusion: These results suggest that PA has significant immunomodulatory properties which probably act by activating mononuclear phagocytic system, augmenting humoral immune response while suppressing cellular immune response.Keywords: Patchouli alcohol, Pogostemon cablin, Immunomodulatory, Phagocytic index, Macrophag

    An integrated bidirectional multi-channel opto-electro arbitrary waveform stimulator for treating motor neurone disease

    This paper presents a prototype integrated bidirectional stimulator ASIC capable of mixed opto-electro stimulation and electrophysiological signal recording. The development is part of the research into a fully implantable device for treating motor neurone disease using optogenetics and stem cell technology. The ASIC consists of 4 stimulator units, each featuring 16-channel optical and electrical stimulation using arbitrary current waveforms with an amplitude up to 16 mA and a frequency from 1.5 Hz to 50 kHz, and a recording front-end with a programmable bandwidth of 1 Hz to 4 kHz, and a programmable amplifier gain up to 74 dB.
