LLNL 10(a)(1)(A) Annual Report (TE-053672-2)--2006
This report summarizes research related to Lawrence Livermore National Laboratory's (LLNL) Experimental Test Site, Site 300 (S300), located within Alameda and San Joaquin Counties (Figure 1) and conducted under the 10(a)(1)(A) (Recovery) permit TE-053672-2. The U.S. Department of Energy/National Nuclear Security Administration (NNSA) holds this property in ownership. The 2006 recovery research at S300 involved fieldwork associated with two species: the California whipsnake (Masticophis lateralis) (MALA) and the California red-legged frog (Rana aurora draytonii) (CRLF). A research project (Biological Opinion 1-1-02-F-0064) investigating the direct effects (fatality/harm) and indirect impacts (habitat alteration) of prescribed burning of coastal sage scrub on MALA was permitted by the U.S. Fish and Wildlife Service (Service), Sacramento Office, in the Spring of 2002. Several different public resource management agencies were approved by the Service to implement recovery-oriented (fuels treatment) studies associated with this research plan. LLNL's proposed seven-year study was initiated in 2002 and is anticipated to conclude in 2008. Results reflected in this report apply to information gathered during the first five years of the project; because of the low MALA sample size collected onsite and the unforeseen wildland fire that burned both of the study sites in 2005, long term conservation-related measures for MALA are not yet evident from study analyses. The CRLF research (Biological Opinion 1-1-02-F-0062) involved translocating individuals from two wetland locations that had received artificial (potable) water discharges for the last 15-20 years. CRLF's that occupied the wetland sites were moved to an enhanced wetland area further downstream (engineered pools in a pre-existing perennial drainage) in an area called ''Mid-Elk Ravine''. The aboveground, potable water flows were terminated once all CRLF translocations had been completed (Summer of 2006)

The 2005 Recovery research at S300 involved fieldwork associated with only two species: Alameda whipsnake (Masticophis lateralis euryxanthus) and the California red-legged frog (Rana aurora draytonii) (RLF). Note: the whipsnake subspecies existing at S300 shows taxonomic variation (generally 50% chaparral whipsnake [Masticophis lateralis] traits) when compared to the Alameda whipsnake (Riemer 1954) and therefore it will be referred to as ''California whipsnake (Masticophis lateralis)'' (CWS) for classification purposes in this report (Swaim 2004)

Lawrence Livermore National Laboratory Pre-project Rare Plant and Wildlife Surveys For the Pit 7 Drainage Diversion and Groundwater Extraction and Treatment Facility
In January 2007, the Department of Energy (DOE) released the final Environmental Assessment for the Proposed Environmental Remediation at the Lawrence Livermore National Laboratory (LLNL) Site 300 Pit 7 Complex. At the same time, the Department of Toxic Substances Control (DTSC) released the final Negative Declaration and Initial Study covering the Pit 7 remediation. No substantial adverse effect on wildlife species of concern was anticipated from the project. However, it was proposed that wildlife surveys should be conducted prior to construction because species locations and breeding areas could potentially change by the time construction activities began. Although no known populations of rare or endangered/threatened plant species were known to occur within the project impact area at the time these documents were released, rare plants listed by the California Native Plant Society had been observed in the vicinity. As such, both DOE and DTSC proposed that plant surveys would be undertaken at the appropriate time of year to determine if rare plants would be impacted by project construction. This document provides the results of wildlife and rare plant surveys taken prior to the start of construction at the Pit 7 Complex

A high throughput live transparent animal bioassay to identify non-toxic small molecules or genes that regulate vertebrate fat metabolism for obesity drug development
<p>Abstract</p> <p>Background</p> <p>The alarming rise in the obesity epidemic and growing concern for the pathologic consequences of the metabolic syndrome warrant great need for development of obesity-related pharmacotherapeutics. The search for such therapeutics is severely limited by the slow throughput of animal models of obesity. Amenable to placement into a 96 well plate, zebrafish larvae have emerged as one of the highest throughput vertebrate model organisms for performing small molecule screens. A method for visually identifying non-toxic molecular effectors of fat metabolism using a live transparent vertebrate was developed. Given that increased levels of nicotinamide adenine dinucleotide (NAD) via deletion of CD38 have been shown to prevent high fat diet induced obesity in mice in a SIRT-1 dependent fashion we explored the possibility of directly applying NAD to zebrafish.</p> <p>Methods</p> <p>Zebrafish larvae were incubated with daily refreshing of nile red containing media starting from a developmental stage of equivalent fat content among siblings (3 days post-fertilization, dpf) and continuing with daily refreshing until 7 dpf.</p> <p>Results</p> <p>PPAR activators, beta-adrenergic agonists, SIRT-1 activators, and nicotinic acid treatment all caused predicted changes in fat, cholesterol, and gene expression consistent with a high degree of evolutionary conservation of fat metabolism signal transduction extending from man to zebrafish larvae. All changes in fat content were visually quantifiable in a relative fashion using live zebrafish larvae nile red fluorescence microscopy. Resveratrol treatment caused the greatest and most consistent loss of fat content. The resveratrol tetramer Vaticanol B caused loss of fat equivalent in potency to resveratrol alone. Significantly, the direct administration of NAD decreased fat content in zebrafish. Results from knockdown of a zebrafish G-PCR ortholog previously determined to decrease fat content in <it>C. elegans </it>support that future GPR142 antagonists may be effective non-toxic anti-obesity therapeutics.</p> <p>Conclusion</p> <p>Owing to the apparently high level of evolutionary conservation of signal transduction pathways regulating lipid metabolism, the zebrafish can be useful for identifying non-toxic small molecules or pharmacological target gene products for developing molecular therapeutics for treating clinical obesity. Our results support the promising potential in applying NAD or resveratrol where the underlying target protein likely involves Sirtuin family member proteins. Furthermore data supports future studies focused on determining whether there is a high concentration window for resveratrol that is effective and non-toxic in high fat obesity murine models.</p
‘They’re more like ordinary stroppy British women’: Attitudes and expectations of maternity care professionals to UK-born ethnic minority women
Objective To explore the attitudes and expectations of maternity care professionals to UK-born ethnic minority mothers. Methods Qualitative in-depth interviews with 30 professionals from eight NHS maternity units in England that provide services for large proportions of women of black Caribbean, black African, Indian, Pakistani and Irish descent. Results All the professionals reported providing care to both UK-born and migrant mothers from ethnic minorities. Most of them felt that they could differentiate between UK-born and migrant mothers based mainly on language fluency and accent. ‘Westernized dress’ and ‘freedom’ were also cited as indicators. Overall, professionals found it easier to provide services to UK-born mothers and felt that their needs were more like those of white English mothers than those of migrant mothers. UK-born mothers were generally thought to be assertive and expressive, and in control of care-related decision-making whereas some South Asian Muslim women were thought to be constrained by family influences. Preconceived ideas about ethnic minority mothers' tolerance of pain in labour, use of pharmacological pain relief measures and mode of delivery were recurring themes. Women's education and social class were felt to be major influences on the uptake of maternity care, regardless of ethnicity.
Conclusions Professionals appeared to equate the needs of UK-born ethnic minority women with those of white English women. Overall, this has positive implications for care provision. Despite this, specific behavioural expectations and unconscious stereotypical views were evident and have the potential to affect clinical practice
Puffins, Pigs, Cod, and Barley: Palaeoeconomy at Undir Junkarinsfløtti, Sandoy, Faroe Islands
This paper reports on the zooarchaeological and archaeobotanical remains from the initial season of excavations at the Norse period site at Undir Junkarinsfløtti in the Faroe islands. These remains represent the first zooarchaeological analysis undertaken for the Faroes and only the third archaeobotanical assemblage published from the islands. The excavated deposits are described and the key findings from the palaeoenvironmental remains highlighted within the context of the wider North Atlantic environmental archaeology of the Norse period
