    Status of the spawning biomass of the Pacific sardine, 1984-85

    State law provides for the initiation of a 1000-ton fishery for Pacific sardines, Sardinops sagax, when the spawning biomass of sardines reaches 20 000 short tons. Data from ichthyoplankton surveys, an aerial spotter index, sea survey cruises, the mackerel fishery, and the live bait fishery are discussed as indicators of the relative abundance of sardines compared to previous years. It appears the spawning biomass of the northern subpopulation of Pacific sardine remains below 20 000 tons. A proposed survey plan is described for determining the relative magnitude of the sardine spawning biomass on an annual basis. Legislation is discussed which was passed in 1984 and pertains to the sardine resource. (23pp.

It appears that the spawning biomass of the northern stock of Pacific sardines remains below 20,000 tons.     Southern California marine sport fishing from privately owned boats: catch and effort for October-December 1982

    The catch landed and effort expended by private-boat sport fishermen were studied in southern California marine waters between October and December 1982, to determine the impact of one segment of the sport fishery on local marine resources. Fishermen returning from fishing trips were interviewed at launch ramps, hoists, and boat-rental facilities. This report contains quantitative data and statistical estimates of total effort, total catch, catch of preferred species, and length frequencies for those species whose catches are regulated by minimum size limits. An estimated 157,000 organisms were landed by 57,700 anglers and 4800 divers. The major components of the angler catch were Pacific mackerel, Scomber japonicus (28,700 estimated catch): white croaker, Genyonemus lineatus (24,000 estimated catch); and Pacific bonito, Sarda chiliensis (16,700 estimated catch). These three species represented almost half the total estimated angler catch. Rockfishes, Sebastes spp: were a major catch component; the 35 rockfish species landed made up 24% of the estimated catch. Divers landed an estimated 14,700 fishes and invertebrates. Chief among these were abalone, Haliotis spp. (4200 estimated catch); rock scallop, Hinnites rugosus (3600 estimated catch); and California spiny lobster, Panulirus interruptus (2500 estimated catch). Angler and diver compliance with size-limit regulations was generally favorable, with diver compliance being particularly scrupulous, especially with invertebrate species. The compliance rate for California halibut, Paralichthys californicus, showed a drop from the previous quarter (July - September 1982) from 70 to 59% legal. During the same time period, size-limit compliance rose from 4 to 20% for Pacific bonito, a species with a tolerance allowing the take of some under-sized fish. (28p.

    Versatile toolbox for high throughput biochemical and functional studies with fluorescent fusion proteins

    Fluorescent fusion proteins are widely used to study protein localization and interaction dynamics in living cells. However, to fully characterize proteins and to understand their function it is crucial to determine biochemical characteristics such as enzymatic activity and binding specificity. Here we demonstrate an easy, reliable and versatile medium/high-throughput method to study biochemical and functional characteristics of fluorescent fusion proteins. Using a new system based on 96-well micro plates comprising an immobilized GFP-binding protein (GFP-mulitTrap), we performed fast and efficient one-step purification of different GFP- and YFP-fusion proteins from crude cell lysate. After immobilization we determined highly reproducible binding ratios of cellular expressed GFP-fusion proteins to histone-tail peptides, DNA or selected RFP-fusion proteins. In particular, we found Cbx1 preferentially binding to di-and trimethylated H3K9 that is abolished by phosphorylation of the adjacent serine. DNA binding assays showed, that the MBD domain of MeCP2 discriminates between fully methylated over unmethylated DNA and protein-protein interactions studies demonstrate, that the PBD domain of Dnmt1 is essential for binding to PCNA. Moreover, using an ELISA-based approach, we detected endogenous PCNA and histone H3 bound at GFP-fusions. In addition, we quantified the level of H3K4me2 on nucleosomes containing different histone variants. In summary, we present an innovative medium/high-throughput approach to analyse binding specificities of fluroescently labeled fusion proteins and to detect endogenous interacting factors in a fast and reliable manner in vitro

    Managing regional innovation strategy projects

    This paper presents a qualitative interview study with 28 RIS project managers that aimed at understanding whether or not this is true in the context of regional innovation and what the specifics of managing regional innovation projects are. In taking up a recent claim for policy intervention studies which allow to “derive precise suggestions for their design and management”.  The study investigated the interrelation between the agility of the management approach and the achievements of RIS projects. Findings raise serious issues concerning methodological approaches for the management of regional innovation projects and question whether the recent public funding mechanisms can lead to sustainable project results

    Bright, excellent, ignored: the contribution of Luhmann's system theory and its problem of non-connectivity to academic management research

    "Niklas Luhmann's theory has been largely ignored in organization studies, compared to other sociological approaches like Weick's sensemaking, Giddens' structuration theory, or Latour's Actor-Network Theory. While having being applied on a number of fields, such as philosophy, sociology, theology, law and political sciences, application of Luhmann's theory is still limited in organization studies (e.g. Munro 2010). In this paper, the authors attempt to explain the reasons of this ignorance and limited use, focusing on Luhmann's writing style, but also on the theoretical and empirical limitations his theory poses. Believing that Luhmann's theory holds great potential to be applied to organizational studies and explain organizational phenomena, they then discuss how it could contribute to radically changing the conventional ways of studying and analyzing organizational phenomena, fostering at the same time, the debate about the value of his theory. The authors analyze organizations as communicative processes that continuously produce and reproduce themselves to create knowledge, make decisions, structure expectations, and redefine organizational boundaries. Finally, they address the empirical challenges of applying Luhmann's theory on organization studies." (author's abstract
