Antihyperalgesia by α2-GABAA Receptors Occurs Via a Genuine Spinal Action and Does Not Involve Supraspinal Sites
Drugs that enhance GABAergic inhibition alleviate inflammatory and neuropathic pain after spinal application. This antihyperalgesia occurs mainly through GABAA receptors (GABAARs) containing α2 subunits (α2-GABAARs). Previous work indicates that potentiation of these receptors in the spinal cord evokes profound antihyperalgesia also after systemic administration, but possible synergistic or antagonistic actions of supraspinal α2-GABAARs on spinal antihyperalgesia have not yet been addressed. Here we generated two lines of GABAAR-mutated mice, which either lack α2-GABAARs specifically from the spinal cord, or, which express only benzodiazepine-insensitive α2-GABAARs at this site. We analyzed the consequences of these mutations for antihyperalgesia evoked by systemic treatment with the novel non-sedative benzodiazepine site agonist HZ166 in neuropathic and inflammatory pain. Wild-type mice and both types of mutated mice had similar baseline nociceptive sensitivities and developed similar hyperalgesia. However, antihyperalgesia by systemic HZ166 was reduced in both mutated mouse lines by about 60% and was virtually indistinguishable from that of global point-mutated mice, in which all α2-GABAARs were benzodiazepine insensitive. The major (α2-dependent) component of GABAAR-mediated antihyperalgesia was therefore exclusively of spinal origin, whereas supraspinal α2-GABAARs had neither synergistic nor antagonistic effects on antihyperalgesia. Our results thus indicate that drugs that specifically target α2-GABAARs exert their antihyperalgesic effect through enhanced spinal nociceptive control. Such drugs may therefore be well-suited for the systemic treatment of different chronic pain conditions
THE PRESENT STATUS OF THE GERM-CELL PROBLEM IN VERTEBRATES
(i) Morphological studies relating to the origin and differentiation of the definitive germ cells in vertebrates have, as indicated, resulted in conflicting views. In many instances two or more competent investigators who have studied the same form have reached different conclusions. (2) Some contend that the germ cells are set aside from the soma during the early stages of embryonic development, and that these alone serve as the progenitors of the functional sex cells. (3) Others recognize an early differentiation of sex cells but hold that these are supplemented by others produced from the somatic epithelium of the gonad in late embryonic or post-embryonic stages. (4) Another group recognizes the early differentiated cells as germ cells but contend that these all degenerate and that the definitive ones are formed from the germinal epithelium. These degenerating germ cells are believed by certain authors to be a phylogenetic recapitulation of the condition in lower forms. (5) Finally, yet another group contends that the so-called primordial germ cells are not germ cells at all but are enlarged cells in some stage of mitosis or in some specific metabolic phase. This group believes that all germ cells are derived from the somatic cells of the germinal epithelium. (6) Experimental work supports the view that the primordial germ cells, which are recognized early, are the progenitors of the definitive sex cells. When these primordial germ cells are prevented from reaching the site of the developing gonad the individual fails to develop sex cells, although a sterile gonad and its associated structures may develop. (7) I suggest that the observed proliferation of germ cells from the germinal epithelium, reported by numerous investigators, can be interpreted in another way by a thorough study of the enlarged germ cells in relation to the epithelium. Effects of dietary carotenoids on mouse lung genomic profiles and their modulatory effects on short-term cigarette smoke exposures
Male C57BL/6 mice were fed diets supplemented with either β-carotene (BC) or lycopene (LY) that were formulated for human consumption. Four weeks of dietary supplementations results in plasma and lung carotenoid (CAR) concentrations that approximated the levels detected in humans. Bioactivity of the CARs was determined by assaying their effects on the activity of the lung transcriptome (~8,500 mRNAs). Both CARs activated the cytochrome P450 1A1 gene but only BC induced the retinol dehydrogenase gene. The contrasting effects of the two CARs on the lung transcriptome were further uncovered in mice exposed to cigarette smoke (CS) for 3 days; only LY activated ~50 genes detected in the lungs of CS-exposed mice. These genes encoded inflammatory-immune proteins. Our data suggest that mice offer a viable in vivo model for studying bioactivities of dietary CARs and their modulatory effects on lung genomic expression in both health and after exposure to CS toxicants
Bill color, not badge size, indicates testosterone-related information in house sparrows
The honesty of ornamental signals of quality is often argued to be enforced via costs associated with testosterone. It is still poorly understood, however, how seasonal variation of testosterone within individuals is related to the timing and extent of ornament development. Here, we studied inter- and intra-individual variability of plasma testosterone levels in a population of 150 captive male house sparrows (Passer domesticus) through the course of a full year. We further analyzed the relationship between plasma testosterone levels and two sexually dimorphic ornaments: badge size and bill coloration. Also, because of a known negative relation between molt and circulating testosterone levels, we analyzed the relationship between ornamentation and molt status during the fall. We found that testosterone levels increased towards the breeding season and decreased before the onset of annual molt. However, within individuals, relative testosterone titers demonstrated low repeatability between seasons. Plasma testosterone levels were not correlated with badge size in any season but were correlated strongly with bill coloration during all periods, except the breeding season when variation in bill color was low. Finally, we found that bill coloration strongly correlated with molt status during fall. Our results indicate that bill coloration, not badge size, is the best ornamental indicator of a “running average” of male testosterone in house sparrows and therefore the best potential indicator of qualities and/or behavioral strategies associated with testosterone
Vitamin A and Retinoid Derivatives for Lung Cancer: A Systematic Review and Meta Analysis
Despite reported antiproliferative activity of vitamin A and its common use for cancer, there is no comprehensive synthesis of its safety and efficacy in lung cancers. To address this issue we conducted a systematic review of the safety and efficacy of vitamin A for the treatment and prevention of lung cancers.Two independent reviewers searched six electronic databases from inception to July 2009 for clinical, observational, and preclinical evidence pertaining to the safety and efficacy of vitamin A and related retinoids for lung cancers. 248 studies were included for full review and analysis. Five RCTs assessed treatment of lung cancers, three assessed primary prevention, and three looked at secondary prevention of lung cancers. Five surrogate studies, 26 phase I/II, 32 observational, and 67 preclinical studies were also included. 107 studies were included for interactions between vitamin A and chemo- or radiation-therapy. Although some studies demonstrated benefits, there was insufficient evidence overall to support the use of vitamin A or related retinoids for the treatment or prevention of lung cancers. Retinyl palmitate combined with beta carotene increased risk of lung cancer in smokers in the large CARET trial. Pooling of three studies pertaining to treatment and three studies on secondary prevention revealed no significant effects on response rate, second primary tumor, recurrence, 5-year survival, and mortality. There was a small improvement in event free survival associated with vitamin A compared to controls, RR 1.24 (95% CI 1.13-1.35). The synthetic rexinoid bexarotene increased survival significantly among a subset of patients in two RCTs (p<0.014, <0.087).There is a lack of evidence to support the use of naturally occurring retinoids for the treatment and prevention of lung cancers. The rexinoid bexarotene may hold promise for use among a subset of patients, and deserves further study

The SIB Swiss Institute of Bioinformatics (www.isb-sib.ch) provides world-class bioinformatics databases, software tools, services and training to the international life science community in academia and industry. These solutions allow life scientists to turn the exponentially growing amount of data into knowledge. Here, we provide an overview of SIB's resources and competence areas, with a strong focus on curated databases and SIB's most popular and widely used resources. In particular, SIB's Bioinformatics resource portal ExPASy features over 150 resources, including UniProtKB/Swiss-Prot, ENZYME, PROSITE, neXtProt, STRING, UniCarbKB, SugarBindDB, SwissRegulon, EPD, arrayMap, Bgee, SWISS-MODEL Repository, OMA, OrthoDB and other databases, which are briefly described in this article
Auswirkungen der Handhabung von Pailletten während der Lagerung auf die Qualität von Stiersamen
Das Ziel der vorliegenden Studie bestand darin, die Auswirkungen der Handhabung von Samendosen durch die Besamungstechniker auf die Qualität des Tiefgefriersamens zu untersuchen. Dazu wurden insgesamt 10 Ejakulate von 7 Stieren kryokonserviert und die Pailletten während 7 Monaten in den Arbeitsgefässen von je 10 Besamungstechnikern in drei verschiedenen Regionen der Schweiz (A, B und C) sowie im Hauptlager aufbewahrt. Nach der Lagerung erfolgten die Bestimmung der Motilität im aufgetauten Samen mittels Computer-assistierter Spermienanalyse (CASA) und die Beurteilung der Akrosomintaktheit (vitale Spermien mit intaktem Akrosom) mit Hilfe der Durchflusszytometrie. Zusätzlich wurden für jeden Besamungstechniker die Non-Return-Rate 56 (NRR) berechnet und der Zusammenhang zwischen NRR der einzelnen Besamungstechniker und der Samenqualität überprüft. Die Ergebnisse zeigen, dass die regionale Handhabung und das Ejakulat einen signifikanten Einfluss sowohl auf die Motilität wie auch auf die Akrosomintaktheit der aufgetauten Samendosen hatten. Die Motilität war nach Lagerung bei den Besamungstechnikern in der Region B signifikant (P < 0.05) tiefer als in der Region C. In der Region B waren der Anteil akrosomintakter Spermien und die NRR signifikant (P < 0.05) tiefer als in der Region A. Bei den einzelnen Besamungstechnikern konnte ein signifikanter Zusammenhang zwischen Motilität bzw. Akrosomintaktheit der bei ihnen gelagerten Samendosen und der NRR festgestellt werden. Aufgrund unserer Ergebnisse kann gefolgert werden, dass die Qualität des Tiefgefriersamens deutliche Unterschiede zwischen den Besamungstechnikern der 3 Regionen aufwies und einen Einfluss auf die NRR hatte
The aim of the present study was to investigate the influence of semen handling by different inseminators on quality of frozen-thawed bovine semen. For this study a total of 10 ejaculates were collected from 7 bulls and semen stored frozen for 7 months in vessels of 10 inseminators each, in three different regions of Switzerland (A, B and C). After storage motility and acrosome integrity (live spermatozoa with intact acrosomes) were measured in frozen-thawed semen by computer-assisted sperm analysis (CASA) and flowcytometry, respectively. In addition, the 56 days non-return rate (NRR) was determined for each inseminator and the relation between NRR of single inseminators and semen quality examined. Results demonstrate that motility as well as acrosome integrity of stored frozen semen were significantly influenced by the specific delivery system used in the different regions and by the ejaculate. After storage of straws in region B semen motility was significantly (P < 0.05) lower than in region C. In region B the percentage live and acrosome intact spermatozoa as well as the NRR of the inseminators were significantly (P < 0.05) lower than in region A. Motility and acrosome integrity of semen stored by single inseminators were significantly correlated with the inseminator's NRR. We conclude that the quality of frozen semen clearly varied between inseminators of the three regions with an impact on NRR
