    Sustainability, space and supply chains: the role of bamboo in Anji County, China

    Rural modernization in China has been profound as the countryside has moved from agricultural production to industrial and tertiary industry development. Within rural areas these changes can have enormous significance for how we think about their sustainability. One rural county that vividly illustrates both the challenges and opportunities of rural development is Anji in Zhejiang Province in Eastern China. Anji is held up as a model of rural sustainable development. In this paper we analyse the basis for the sustainability claims made of Anji and to do so, we examine how the production and processing of bamboo materials transformed Anji into a place-specific bamboo-making locality that is lauded for its sustainability. We analyse how thinking on a place and a material (bamboo) come together to reinforce thinking on sustainability in rural China. We then go on to critically question the politico-economic arrangements that construct Anji and bamboo as models of sustainability. We argue that whilst both Anji and bamboo do have notable features that characterise them as sustainable and together can make an even more persuasive case for rural sustainability, a more detailed analysis allows us to uncover the deep-rooted tensions that persist in Chinese rural development between environmental protection and economic growth. The paper draws on a mixture of published and unpublished material to provide a detailed examination of the ways in which bamboo supply chains operate within and through Anji. The paper concludes that local constructions of sustainability are driven by economic rather than environmental values

    Incremental Learning on Food Instance Segmentation

    Food instance segmentation is essential to estimate the serving size of dishes in a food image. The recent cutting-edge techniques for instance segmentation are deep learning networks with impressive segmentation quality and fast computation. Nonetheless, they are hungry for data and expensive for annotation. This paper proposes an incremental learning framework to optimize the model performance given a limited data labelling budget. The power of the framework is a novel difficulty assessment model, which forecasts how challenging an unlabelled sample is to the latest trained instance segmentation model. The data collection procedure is divided into several stages, each in which a new sample package is collected. The framework allocates the labelling budget to the most difficult samples. The unlabelled samples that meet a certain qualification from the assessment model are used to generate pseudo-labels. Eventually, the manual labels and pseudo-labels are sent to the training data to improve the instance segmentation model. On four large-scale food datasets, our proposed framework outperforms current incremental learning benchmarks and achieves competitive performance with the model trained on fully annotated samples

    蠻野蒙昧的時代,原始人對於大自然所發生的各種現象,例如風吹雨打、 打雷閃電等,都是一無所知,因此,感到十分驚奇與敬畏,認為是有一種超人的力量在支配着大自然的一舉一動,原始人叫它做「神」,他們這種富於想像力的解釋就產生出神話。 可是隨着時代的邁進,文明程度的提高,神話也隨之變形。以西王母神話為例,據《山海經》描寫,西王母本來是一個豹尾、虎齒、蓬髮、戴勝,而司天 之厲及五殘的獰厲之神,有三青鳥為之取食。到《穆天子傳》,西王母與周穆王在瑤池中詩歌唱答,王母變成了一個氣象雍容的帝女了。《淮南子》裡,西王母變成了擁有長生不死的吉神;《漢武帝內傳》進一步成為了年三十許,容顏絕世的美人,以前的三青鳥也成了一群美麗的侍女,與漢武帝載歌載舞。 這種變化,正好用蕭兵在《神話學導論》的解釋:神話是「滿足著初民潛在的審美欲求。隨着文化的進步,它就情不自禁地由神聖的祭壇滑向快樂的歌場。不但語言本身能夠自我增殖,以語言為載體的『神話:幻想性口頭文學』更會自我擴張。」另外,也可以運用王孝廉《中國的神話與傳說‧神話與詩》對神話變化四大原因作出解釋。 現在試就西王母形象轉變的三個階段:神格化、人格化、仙話化加以作出分析與討論,從而也帶出神話與文學之間的關係

    A targeted gene panel that covers coding, non-coding and short tandem repeat regions improves the diagnosis of patients with neurodegenerative diseases

    Genetic testing for neurodegenerative diseases (NDs) is highly challenging because of genetic heterogeneity and overlapping manifestations. Targeted-gene panels (TGPs), coupled with next-generation sequencing (NGS), can facilitate the profiling of a large repertoire of ND-related genes. Due to the technical limitations inherent in NGS and TGPs, short tandem repeat (STR) variations are often ignored. However, STR expansions are known to cause such NDs as Huntington\u27s disease and spinocerebellar ataxias type 3 (SCA3). Here, we studied the clinical utility of a custom-made TGP that targets 199 NDs and 311 ND-associated genes on 118 undiagnosed patients. At least one known or likely pathogenic variation was found in 54 patients; 27 patients demonstrated clinical profiles that matched the variants; and 16 patients whose original diagnosis were refined. A high concordance of variant calling were observed when comparing the results from TGP and whole-exome sequencing of four patients. Our in-house STR detection algorithm has reached a specificity of 0.88 and a sensitivity of 0.82 in our SCA3 cohort. This study also uncovered a trove of novel and recurrent variants that may enrich the repertoire of ND-related genetic markers. We propose that a combined comprehensive TGPs-bioinformatics pipeline can improve the clinical diagnosis of NDs

    TRAPPC9 Mediates the Interaction between p150Glued and COPII Vesicles at the Target Membrane

    Background: The transport of endoplasmic reticulum (ER)-derived COPII vesicles toward the ER-Golgi intermediate compartment (ERGIC) requires cytoplasmic dynein and is dependent on microtubules. p150Glued, a subunit of dynactin, has been implicated in the transport of COPII vesicles via its interaction with COPII coat components Sec23 and Sec24. However, whether and how COPII vesicle tether, TRAPP (Transport protein particle), plays a role in the interaction between COPII vesicles and microtubules is currently unknown. Principle Findings: We address the functional relationship between COPII tether TRAPP and dynactin. Overexpressed TRAPP subunits interfered with microtubule architecture by competing p150Glued away from the MTOC. TRAPP subunit TRAPPC9 bound directly to p150Glued via the same carboxyl terminal domain of p150Glued that binds Sec23 and Sec24. TRAPPC9 also inhibited the interaction between p150Glued and Sec23/Sec24 both in vitro and in vivo, suggesting that TRAPPC9 serves to uncouple p150Glued from the COPII coat, and to relay the vesicle-dynactin interaction at the target membrane. Conclusions: These findings provide a new perspective on the function of TRAPP as an adaptor between the ERGIC membrane and dynactin. By preserving the connection between dynactin and the tethered and/or fused vesicles, TRAPP allows nascent ERGIC to continue the movement along the microtubules as they mature into the cis-Golgi

    The impact of positron emission tomography on primary tumour delineation and dosimetric outcome in intensity modulated radiotherapy of early T-stage nasopharyngeal carcinoma

    Background: In intensity modulated radiotherapy (IMRT) of nasopharyngeal carcinoma (NPC), accurate delineation of the gross tumour volume (GTV) is important. Image registration of CT and MRI has been routinely used in treatment planning. With recent development of positron emission tomography (PET), the aims of this study were to evaluate the impact of PET on GTV delineation and dosimetric outcome in IMRT of early stage NPC patients.Methods: Twenty NPC patients with T1 or T2 disease treated by IMRT were recruited. For each patient, 2 sets of NP GTVs were delineated separately, in which one set was performed using CT and MRI registration only (GTVCM), while the other set was carried out using PET, CT and MRI information (GTVCMP). A 9-field IMRT plan was computed based on the target volumes generated from CT and MRI (PTVCM). To assess the geometric difference between the GTVCM and GTVCMP, GTV volumes and DICE similarity coefficient (DSC), which measured the geometrical similarity between the two GTVs, were recorded. To evaluate the dosimetric impact, the Dmax, Dmin, Dmean and D95 of PTVs were obtained from their dose volume histograms generated by the treatment planning system.Results: The overall mean volume of GTVCMP was greater than GTVCM by 4.4 %, in which GTVCMP was slightly greater in the T1 group but lower in the T2 group. The mean DSC of the whole group was 0.79 ± 0.05. Similar mean DSC values were also obtained from the T1 and T2 groups separately. The dosimetric parameters of PTVCM fulfilled the planning requirements. When applying this plan to the PTVCMP, the average Dmin (56.9 Gy) and D95 (68.6 Gy) of PTVCMP failed to meet the dose requirements and demonstrated significant differences from the PTVCM (p = 0.001 and 0.016 respectively), whereas the doses to GTVCMP did not show significant difference with the GTVCM.Conclusion: In IMRT of early stage NPC, PET was an important imaging modality in radiotherapy planning so as to avoid underdosing the PTV, although its effect on GTV delineation was not significant.     Prolonged exposure to bacterial toxins downregulated expression of toll-like receptors in mesenchymal stromal cell-derived osteoprogenitors

    <p>Abstract</p> <p>Background</p> <p>Human mesenchymal stromal cells (MSCs, also known as mesenchymal stem cells) are multipotent cells with potential therapeutic value. Owing to their osteogenic capability, MSCs may be clinically applied for facilitating osseointegration in dental implants or orthopedic repair of bony defect. However, whether wound infection or oral microflora may interfere with the growth and osteogenic differentiation of human MSCs remains unknown. This study investigated whether proliferation and osteogenic differentiation of MSCs would be affected by potent gram-positive and gram-negative derived bacterial toxins commonly found in human settings.</p> <p>Results</p> <p>We selected lipopolysaccharide (LPS) from <it>Escherichia coli </it>and lipoteichoic acid (LTA) from <it>Streptococcus pyogenes </it>as our toxins of choice. Our findings showed both LPS and LTA did not affect MSC proliferation, but prolonged LPS challenge upregulated the osteogenic differentiation of MSCs, as assessed by alkaline phosphatase activity and calcium deposition. Because toll-like receptors (TLRs), in particularly TLR4 and TLR2, are important for the cellular responsiveness to LPS and LTA respectively, we evaluated their expression profiles serially from MSCs to osteoblasts by quantitative PCR. We found that during osteogenic differentiation, MSC-derived osteoprogenitors gradually expressed TLR2 and TLR4 by Day 12. But under prolonged incubation with LPS, MSC-derived osteoprogenitors had reduced TLR2 and TLR4 gene expression. This peculiar response to LPS suggests a possible adaptive mechanism when MSCs are subjected to continuous exposure with bacteria.</p> <p>Conclusion</p> <p>In conclusion, our findings support the potential of using human MSCs as a biological graft, even under a bacterial toxin-rich environment.</p
